Inside the nucleus, chromosomes are subjected to direct physical interaction between different components, active forces, and thermal noise, leading to the formation of an ensemble of three-dimensional structures. However, it is still not well understood to what extent and how the structural ensemble varies from one chromosome region or cell-type to another. We designed a statistical analysis technique and applied it to single-cell chromosome imaging data to reveal the fluctuation of individual chromosome structures. By analyzing the resulting structural landscape, we find that the largest dynamic variation is the overall radius of gyration of the chromatin region, followed by domain reorganization within the region. By comparing different human cell-lines and experimental perturbations data using this statistical analysis technique and a network entropy approach, we identify both cell-type and condition-specific features of the structural landscapes. We identify a relationship between epigenetic state and the properties of chromosome structure fluctuation and validate this relationship through polymer simulations. Overall, our study suggests that the types of variation in a chromosome structure ensemble are cell-type as well as region-specific and can be attributed to constraints placed on the structure by factors such as variation in epigenetic state.

Abstract: The purpose of this research was to standardize experiential learning (EL) facilitators in outdoor activities in West Sumatera. The result of this research is expected that the EL facilitator is competent with the issuance of professional certificate by the National Agency for Profession Certification (BNSP). This research uses quantitative method with analytic descriptive approachand has been done in two stages. First, Implementation of workshop program for experiential learning facilitators and produce recommendations for workshop participants. Second, conductied a competency test of EL facilitators by the National Agency for Profession Certification (BNSP). The number of respondents was 20 facilitators from 4 providers (n=20). Data were collected using Indonesian national competence standard of tourism sector in the field of outbound / experiential learning facilitator and BNSP test. The data were processed using descriptive statistical analysis technique.  The results of the study showed that 19 participants were declared competent and 1 incompetent participant. The results of the study showed that 19 participants were declared competent and 1 participants declared incompetent by the National Agency for Profession Certification (BNSP). These results indicate that the EL workshop program resulted 19 participants have been declared competent in guiding outdoor activities using experiential learning approach.

Abstract. We present results from three years of mesospheric and thermospheric wind measurements obtained using full correlation analysis (FCA) and imaging Doppler interferometry (IDI) for the Buckland Park MF radar. The IDI winds show excellent agreement with the FCA winds, both for short (2-min) and longer term (hourly, fortnightly) comparisons. An extension to a commonly used statistical analysis technique is introduced to show that the IDI winds are approximately 10% larger than the FCA winds, which we attribute to an underestimation of the FCA winds rather than an indication that IDI overestimates the wind velocity. Although the distribution of IDI effective scattering positions are shown to be consistent with volume scatter predictions, the velocity comparisons contradict volume scatter predictions that the IDI velocity will be overestimated. However, reanalysis of a 14-day data set suggests the lack of overestimation is due to the radial velocity threshold used in the analysis, and that removal of this threshold produces the volume scatter predicted overestimation of the IDI velocities. The merits of using hourly IDI estimates versus hourly averaged 2-min IDI estimates are presented, suggesting that hourly estimated turbulent velocities are overestimated.

Confidence is important to be possessed by students through the learning process, students who have low self-confidence will be less optimal in developing ability. This study aims to determine the effectiveness of gestalt counseling with the technique around to improve confidence in the learning process in students of class XI MIA 4 SMA Negeri 1 Singaraja academic year 2016/2017. This type of research is pre-experimental with one group pretest-posttest design. Samples of research were six students selected through purposive sampling technique. Methods of data collection using a confident questionnaire that has been calculated the validity and reliability. The subjects from the study were students of class XI MIA 4 who had low self-esteem. Data analysis technique used is t-test statistical analysis technique. The results from the analysis, found that gestalt counseling with the technique of traveling can increase confidence. It is concluded from the analysis result, t count> table with significance level 5% (4, 38> 2, 015). The results from the analysis show that the hypothesis proposed in this study is acceptable. As the implication of this research is expected to be an alternative for counseling teachers in schools in improving student self-confidence.
