Muscle hardness and its relationship with different muscle lengths/positions are important for understanding its underlying physiological status, and yet remained unclear. This study aimed to detect the local muscle hardness at different muscle lengths and identify the influence of muscle position on muscle hardness in healthy adults. A total of 26 healthy adults participated in this study. Shear wave elastography (SWE) was used to measure the muscle hardness of the Rectus Femoris (RF), Tibialis Anterior (TA) and Gastrocnemius Medialis (GM). Each muscle was tested at both resting (RST) and mid-range lengthened (MRL) positions. A novel ultrasound probe placing method was introduced, applied, and evaluated in this study. Moderate to excellent intra-/inter-rater reliability (Intraclass Correlation Coefficient, ICC ≥ 0.70) was found for muscle hardness measurements. The muscle hardness significantly increased from the RST to MRL position for all three muscles (p < 0.001). This study found that the muscle hardness increased at its mid-range lengthened position from the resting position. The mid-range lengthened muscle position of TA and GM could also be sensitive enough to reflect the age-related changes in local muscle hardness. This study also highlights the importance of placing the assessed extremities in an appropriate and consistent position when assessing muscle qualities by ultrasonics in clinical practice.

ABSTRACT A highly sensitive quantitative PCR detection method has been developed and applied to the distribution analysis of human intestinal bifidobacteria by combining real-time PCR with Bifidobacterium genus- and species-specific primers. Real-time PCR detection of serially diluted DNA extracted from cultured bifidobacteria was linear for cell counts ranging from 106 to 10 cells per PCR assay. It was also found that the method was applicable to the detection of Bifidobacterium in feces when it was present at concentrations of >106 cells per g of feces. Concerning the distribution of Bifidobacterium species in intestinal flora, the Bifidobacterium adolescentis group, the Bifidobacterium catenulatum group, and Bifidobacterium longum were found to be the three predominant species by examination of DNA extracted from the feces of 46 healthy adults. We also examined changes in the population and composition of Bifidobacterium species in human intestinal flora of six healthy adults over an 8-month period. The results showed that the composition of bifidobacterial flora was basically stable throughout the test period.

AbstractObjectiveTo describe a new dynamic echodefecography(EDF) using BK ultrasound 8838 transducer and compare its testing results with X-ray defecography.MethodThe BK 8838 ultrasound probe is used to evaluate the static 3D scan, dynamic 3D scan and dynamic 2D scan of pelvic floor and compare its testing results with X-ray defecography and defined its value to evaluate the pelvic floor disease.ResultsFifty-seven patients were studied (24 male and 33 female). Forty seven patients were diagnosed as anismus by EDF and 46 patients were diagnosed as anismus by X-ray defecography. Sixteen patients were diagnosed as rectocele by X-ray defecography, among which eight were classified as mild(6-15mm), 4 as moderate(16-30mm) and 4 as severe(over 30mm).Fourteen patients with constipation and 2 patients with anal pain were diagnosed as intussusception by EDF, but only 3 patients were diagnosed as intussusception by X-ray defecography. Two patients with constipation were diagnosed as perineal descent by EDF and none by X-ray defecography. Two patients were diagnosed as enterocele by EDF as well as X-ray defecography.ConclusionThe EDF established by BK 8838 ultrasound probe can show clear anatomy and real time movement of pelvic floor muscle. The EDF is more sensitive to the diagnosis of intussusception, perineal descent(PD) and anal spincter defect than X-ray defecograpgy. For anismus,rectocele and enterocele,the diagnosis results are comparable between EDF and X ray defecography. Further study is needed to determine its clinical values to evaluate the pelvic disease.

Abstract Background and purpose- MicroRNAs (miRNAs) in exosomes had been implicated differentially expressed in MMD patients, but the miRNAs expression in circulating leukocytes remains unclear. This study was investigated on the differential expression of miRNAs in peripheral leukocytes between MMD patients and healthy adults, and among patients with subtypes of MMD.Methods- A total of 30 patients with MMD and 10 healthy adults were enrolled in a stroke center from October 2017 to December 2018. The gene microarray was used to detect the differential expression profiles of miRNA in leukocytes between MMD patients and controls, and the differentially expressed miRNAs were verified by the method of real-time PCR. The Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) were used to explore the key signaling pathways and possible pathogenesis of MMD.Results- The microarray results showed 12 differentially expressed miRNAs in leukocytes of MMD patients compared with controls (fold change > 2.0 and P < 0.05), of which 7 miRNAs were up-regulated (miRNA-142-5p, miRNA-29b-3p, miRNA-424-5p, MiRNA-582-5p, miRNA-6807-5p, miRNA-142-3p, miRNA-340-5p), and 5 miRNAs were down-regulated (miRNA-144-3p, miRNA-144-5p, miRNA-451a, miRNA-486-5p, miRNA-363-3p). The real-time PCR confirmed 7 differentially expressed miRNAs (P<0.05), of which 4 miRNAs (miRNA-29b-3p, miRNA-142-3p, miRNA-340-5p, miRNA-582-5p) were up-regulated, and 3 miRNAs (miRNA-363-3p, miRNA-451a and miRNA-486-5p) were down-regulated. Both GO and KEGG analysis suggested that the Wnt signaling pathway may be involved in the pathogenesis of MMD. In addition, miRNAs were also differentially expressed among patients with subtypes of MMD. Conclusion- This study indicated that miRNAs are differentially expressed in peripheral leukocytes between MMD patients and healthy adults, and among patients with subtypes of MMD. The Wnt signaling pathway is probably involved in the pathogenesis of MMD.
