Dysregulation of ion and potential homeostasis in the scala media is the most prevalent cause of hearing loss in mammals. However, it is not well understood how the development and function of the stria vascularis regulates this fluid homeostasis in the scala media. From a mouse genetic screen, we characterize a mouse line, named 299, that displays profound hearing impairment. Histology suggests that 299 mutant mice carry a severe, congenital structural defect of the stria vascularis. The in vivo recording of 299 mice using double-barreled electrodes shows that endocochlear potential is abolished and potassium concentration is reduced to ∼20 mM in the scala media, a stark contrast to the +80 mV endocochlear potential and the 150 mM potassium concentration present in healthy control mice. Genomic analysis revealed a roughly 7-kb-long, interspersed nuclear element (LINE-1 or L1) retrotransposon insertion on chromosome 11. Strikingly, the deletion of this L1 retrotransposon insertion from chromosome 11 restored the hearing of 299 mutant mice. In summary, we characterize a mouse model that enables the study of stria vascularis development and fluid homeostasis in the scala media.

Tall, dwarf green and San Ramon are three phenotypically distinct forms of coconut, currently used for production of hybrid seeds in Sri Lanka. Development of molecular markers for further improvement of important economic traits of these hybrids was thought important due to various constraints faced by conventional breeders. Sixty random primers (OPERON) were used for generation of RAPD profiles using DNA from 3 individuals of each form. All the sixty primers except one (OPC3) yielded RAPD-PCR products with coconut DNA. They generated a total of 326 consistently amplified bands of which 54 distinguished tall, dwarf green and San Ramon. Among RAPDs detected 16 and 8 were specific to tall and dwarf respectively, 18 distinguished dwarf and San Ramon from tall, 9 distinguished tall and San Ramon from dwarf and 3 distinguished tall and dwarf green from San Ramon. It is expected to study the segregation of these markers in F2 populations derived from tall x dwarf green and tall x San Ramon F1 palms.

Die Einführung von immunhisto-/zytochemischen und molekularbiologischen bzw. zytogenetischen Methoden in der onkologische Forschung hat trotz vieler nach wie vor bestehender offener Fragen insgesamt zu einem besseren Verständnis der genetischen Ursachen der Tumorentstehung geführt. Darüber hinaus ergaben sich in verschiedenen Studien Hinweise für eine prognostische Relevanz von bestimmten (zyto)genetischen Veränderungen bzw. residualen Tumorzellen in Lymphknoten oder Knochenmark. Bevor diese Untersuchungen jedoch in den klinischen Alltag Einzug halten und therapeutische Konsequenzen abgeleitet werden können, sind weitere prospektive Studien mit groáen Patientenfallzahlen sowie vereinheitlichte und methodologisch praktikable Untersuchungstechniken zu fordern.
