The mass propagation of Lilium in vitro by stimulation of multiple adventitious bulb-scale formation and by shake culture

Multiple adventitious bulb scales of Lilium were investigated as a source for bulblet production. Addition of kinetin to the medium markedly increased the number of adventitious bulb scales differentiated from bulblets grown in vitro. The segments with numerous bulb scales were too small to separate into individual bulb scales, but the use of a shake culture technique markedly stimulated their growth. Such bulb scales were severed and each individual scale was transferred to fresh medium without kinetin to induce bulblet formation. The use of multiple adventitious bulb scales grown by shake culture seems to be advantageous for the mass propagation of Lilium bulblets.

The aim of this investigation was produced micro tubers of four potato cultivars Premiere, Bintje, Estima and Escort in vitro. Apical meristems (0.2-0.4 mm) of potato cultivars were excised and cultured on nutrient medium and incubated at 24±2 Cº and 1000 lux light intensity for 16 hrs per day. The developing plantlets were examined serological by using ELISA technique to eliminate the viral infected plantlets. The virus-free plantlets were chopped into pieces with single bud and re cultured on fresh medium for mass propagation. For micro tubers formation in test tubes, the cultures were transferred to another medium containing a high percent of sucrose (60g/L) with different concentrations of kinetin; the cultures were incubated under 16±2 Cº and 8 hrs photoperiod. The plantlets formed micro tubers after 8-10 weeks from culturing. The results showed significant differences among cultivar’s in their response to in vitro culture and micro tubers formation. The results also showed that the kinetin concentration had significant effect on micro tubers, and 1mg/l kinetin concentration was the best. The micro tubers were stored for 10 week at 4Cº to break down the dormancy period, and gave 100% germination under nursery condition. Numbers of tubers derived from micro tubers and normal tubers of these cultivars were compared at the end of season.

Nauclea diderrichii is a tree species of economic importance. However, its plantation establishment is limited by inadequate seedling production. Hence, there is ample scope of tissue culture for its mass propagation. Its in vitro plantlets development as affected by media strengths indicated that 100 % seed germination was obtained in full MS basal medium while the least (3.35 %) was from quarter-strength at 8 Weeks after inoculation (WAI). The effects of BAP and NAA assessed on the growth of its sub-cultured plantlets showed that highest number of leaves (17) and adventitious shoots (3) were obtained from MS basal medium supplemented with 0.1 mg/l BAP only. Whereas, highest shoot length (3.61 cm) and average number of roots (5/plantlet) were obtained from the same medium without hormone(s) at 8 WAI. Further sub-culturing into MS with 0.05 mg/l NAA resulted into plantlets having optimum shoot and massive root growth ready for acclimatization in 6 WAI. The plantlets were successfully acclimatized using coconuthusk/ topsoil mixture with 90 % survival. Plant Tissue Cult. & Biotech. 31(1): 51-60, 2021 (June)

Ashwagandha (Withania somnifera L.) Dunal] is an important medicinal plant and a major source of alkaloids and steroids (withanolids), which is regularly used in pharmaceutical industries. Various vegetative parts were studied for its mass propagation through tissue culture technique. Seeds were pretreated with GA3 (50 and 100 mgl-1) for 24 h and 80% germination was achieved. All the explants were taken from in-vitro germinated plant. Among the different explants tested, multiple shoot formation was achieved from shoot-tip and nodal explants in MS medium + 0.25, 0.5, and 1.0 mgl-1 kinetin. Nodal explants were selected for mass propagation protocol because it formed maximum number of shoots (16.25 shoots per explant) on MS medium + 1mgl-1 kinetin after eight weeks of culture. Increase in concentration of kinetin was most effective for callus formation. For further multiplication these shoots were sub-cultured on MS +0.5 mgl-1 kinetin. Presence of IAA at 0.5 mgl-1 was most effective medium for rooting of in-vitro propagated shoots. However, hardening was not achieved for these propagated plants. SummaryIt is shown that the supernatant of unstirred whole blood at 37° C, stimulated by 1 μg/ml of collagen for 10 sec, produces a rapid generation of pro and antiaggregatory compounds with a final proaggregatory activity which can be detected for more than 60 min on a platelet rich plasma (PRP) by turbidometric aggregometry. A reversible aggregation wave that we have called BASIC wave (for Blood Aggregation Stimulatory and Inhibitory Compounds) is recorded. The collagen stimulation of unstirred PRP produces a similar but smaller BASIC wave. BASIC’s intensity increases if erythrocytes are added to PRP but decreases if white blood cells are added instead. Aspirin abolishes “ex vivo” the ability of whole blood and PRP to generate BASIC waves and dipyridamole “in vitro” significantly reduces BASIC’s intensity in whole blood in every tested sample, but shows little effect in PRP.

ABSTRACT 1. Reserpine does not inhibit the tissue respiration of liver in normal male rats (in vitro). 2. The decrease of tissue respiration of the liver with simultaneous morphological stimulation of the thyroid gland after long administration of reserpine is due to a minute inhibition of the hormone synthesis in the thyroid gland. 3. The morphological alterations of the thyroid in experimental hypothyroidism due to perchlorate can not be prevented with reserpine.

ABSTRACT Whether the short-term regulation of thyroidal protein synthesis by TSH occurs at the transcriptional or the translational level was tested by measuring the effect of actinomycin D (act D) on the TSH-induced stimulation of L-14C-leucine incorporation into the thyroidal proteins of rats. TSH was injected 6 h before the rats were killed. The thyroid glands were then removed and incubated in vitro in the presence of L-14C-leucine for 2 h. The pronounced stimulation of leucine incorporation in the TSH-treated animals was depressed as compared with controls but still significant even when the animals had been pre-treated with 100 μg act D 24 and 7 h before sacrifice. On the other hand, act D strongly decreased incorporation of 3H-uridine into RNA. Short-term regulation of thyroidal protein synthesis by TSH appears to be partly but not wholly dependent on neosynthesis of RNA. Hence regulation may partly occur at the translation level of protein synthesis.
