Characterization of Spanish grapevine cultivar diversity using sequence-tagged microsatellite site markers

A broad germplasm bank collection containing most of the autochthonous Spanish grapevine cultivars was analyzed using six sequence-tagged microsatellite site (STMS) loci: VVS2, VVMD5, VVMD7, ssrVrZAG47, ssrVrZAG62, and ssrVrZAG79. The number of alleles obtained ranged from 9 in ssrVrZAG47 to 13 in VVS2, and the observed genotypes per locus varied between 24 (ssrVrZAG47) and 41 (VVSS2). A total of 57 unique genotypes were obtained considering all 6 loci, and 40 varieties presented at least 1 of these specific genotypes. The genotypic combinations for the 6 loci have generated 163 different profiles in the 176 cultivars. Ten pairs of accessions and one group of four Garnacha's cultivars can not be differentiated. The observed heterozygosity varied between 75.6 (VVMD7) and 90.9% (VVMD5), without significant differences from the expected values for any loci. The VVMD5 locus was the most informative, and also showed the highest discrimination power. The cumulative discrimination power for all six loci was practically 1; however, in fact, these STMS loci have differentiated only about 93% of the accessions, probably owing to high relatedness of the plant material. Usefulness of this STMS set for characterization of a Spanish grapevine collection is emphasized, as well as the elaboration of databases with these molecular markers.Key words: Vitis vinifera L., STMS markers, genotype identification, grapevine synonymies, germplasm bank.

Twenty-one grapevine varieties grown all over Greece and belonging to ‘Mavroudia’ group were ampelographically described and genotyped by AFLP molecular analysis in order to discriminate the varieties, synonyms, homonyms and variations of the group. In most cases, the molecular findings confirmed the results of the ampelographic description. In general, and despite the high degree of genetic similarity between certain pairs of the studied cultivars, the group of ‘Mavroudia’ was characterized as being heterogeneous. From the studied cultivars, ‘Kountoura mavri’, ‘Mavro Spetson’ and ‘Pappoudes’ showed very high degree of genetic similarity, sustaining the hypothesis that the last two are clones of the first. Grapevine cultivar ‘Pappoudes’ was for the first time ampelographically described and identified as being closely related to ‘Kountoura mavri’. High degree of genetic similarity was observed between cultivars ‘Gaidouricha’ and ‘Agiomavritiko’, suggesting that they probably originated from the same parent variety through the accumulation of mutations. This may also be true for cultivars ‘Mavrokorakas’ and ‘Kartsiotis’. Also, the results from the statistical analysis showed that ‘Mavro Arachovis’, ‘Mavroudi Voulgarias’ and ‘Voulgaroudes’, despite the relatively high genetic similarity between them, are different. The same applies for the rest of the cultivars studied, while ‘Mavro Kalavriton’, the most widespread variety of the ‘Mavroudia’ group, showed the lowest degree of genetic similarity within the all the cultivars studied. The ampelographic description in combination with the molecular method AFLP are effective for the study of the between and within genetic diversity of grapevine cultivars as well as for their identification and discrimination.

Vitis vinífera L. cv. Maturana Blanca is an autochthonous minor variety recently recovered in the Rioja Qualified Denomination of Origin (D.O.Ca Rioja, Spain) for the production of monovarietal white wines with singular and differentiated characteristics. In this paper, Maturana Blanca wines made with different technologies were analyzed by sensory analysis and aromatic profile by gas chromatography-mass detector. Maturana Blanca wines were characterized by low pH, high acidity, and yellowish tonalities. The compounds that most influenced the aroma of Maturana Blanca wines were those related to fruity (acetates and ethyl esters), floral aromas (2-phenylethanol), and spicy notes (γ-decalactone). These wines were mainly characterized by volatile compounds of fruity aromas of banana and apple. The use of pre-fermentative maceration increased the concentration of ethyl esters and acetates and produced wines with higher odor activity values, indicating a greater aromatic intensity. The aromatic profile of Maturana Blanca wines fermented in oak barrels showed a greater complexity as they were also characterized by the presence of important amounts of furfural, whiskey lactone, and eugenol. The sensory analysis confirmed the results obtained in the aromatic analysis, and described the wines as fresh and balanced in mouth, with notes of acidity and medium to high persistence. These results will contribute to a better knowledge of this white variety.

Abstract: In Palestine, grape culture consists of ecotypes and cultivars (also called local varieties), for which a large number of homonymous and synonymous designations exist as well as misnaming of cultivars. The present study is the first report using detailed ampelographic characterizations (39 informative traits) to assess genetic diversity and detect similarities among sixteen accessions collected from putative diverse grape genotypes In general, 30 descriptors presented highly and satisfactory divergent genotypes, whereas the remaining traits showed no or very little ampelographic variation. Based on the similarity matrix and the resulting dendrogram of these ampelographic data, distinguishable genotypes as well as some cases of synonymies and homonymies clearly exist. A synonymy case seemed to be in four genotypes including Jandali-Mfarad, Jan-dali-Mrazraz, Jandali, and Hamadani-Mattar, which indeed showed genetic distances of less than 0.5, sug-gesting their relatedness, and the possibility that they are the same genotype, but with different names. In addition, homonym cases also occur in the following pairs of “Marawi’s, Hamadani’s, and Zaini’s genotypes, in which each pair seems to be two distinctive genotypes. Finally, among the 16 examined genotypes, the Zaini-Baladi genotype tended to show the highest genetic distance values from the others and thus could be potentially incorporated into any further local or regional breeding programs as well as germplasm conservation.

The efficacy of extracts and essential oils fromAllium tuberosum, Coriandrum sativum, Cymbopogon martini, Cymbopogon winterianus,andSantolina chamaecyparissuswas evaluated againstCandidaspp. isolates from the oral cavity of patients with periodontal disease. The most active oil was fractionated and tested againstC. albicansbiofilm formation. The oils were obtained by water-distillation and the extracts were prepared with macerated dried plant material. The Minimal Inhibitory Concentration—MIC was determined by the microdilution method. Chemical characterization of oil constituents was performed using Gas Chromatography and Mass Spectrometry (GC-MS). C. sativum activity oil upon cell and biofilm morphology was evaluated by Scanning Electron Microscopy (SEM). The best activities against planktonicCandidaspp. were observed for the essential oil and the grouped F8–10fractions fromC. sativum. The crude oil also affected the biofilm formation inC. albicanscausing a decrease in the biofilm growth. Chemical analysis of the F8–10fractions detected as major active compounds, 2-hexen-1-ol, 3-hexen-1-ol and cyclodecane. Standards of these compounds tested grouped provided a stronger activity than the oil suggesting a synergistic action from the major oil constituents. The activity ofC. sativumoil demonstrates its potential for a new natural antifungal formulation.
