The first stage of follicle development was due chiefly to hydration; during the second (deutoplasmic) stage 60 mg of solid were taken up with each 100 mg increase in follicle weight. Plasma calcium and protein P rose near end of hydration stage, remained elevated during deutoplasmic stage, reached extreme levels (max. Ca = 90 mm/l.; protein P = 86 mm/l.) near ovulation, and generally fell to anestrous levels while eggs were in early cleavage. Calcium increased in proportion to protein bound P of both plasma and follicles. During deutoplasmic stage a phospho-lipoprotein, of similar gross composition to yolk protein, appeared in plasma. Liver weight increased during hydration stage, remained elevated throughout deutoplasmic stage and decreased near ovulation. Fat body weight increased with onset of estrus, reached maximum during hydration stage and progressively decreased during deutoplasmic stage. Plasma and liver changes characteristic of estrus were reproduced in fasted male snakes with estradiol injections.

SummaryInDrosophila, the threeyolk protein(yp) genes are transcribed in a sex-, tissue- and developmentally specific manner, providing an ideal system in which to investigate the factors involved in their regulation. The yolk proteins are synthesized in the fat body of adult females, and in the ovarian follicle cells surrounding the developing oocyte during stages 8–10 of oogenesis. We report here an analysis of theyolk protein 3(yp3) gene and its flanking sequences by means of P-element mediated germ-line transformation and demonstrate that a 747 bp promoter region is sufficient to direct sex-specific expression in the female fat body and both the temporal- and cell-type-specificity of expression during oogenesis. Two elements that independently governyp3transcription in these tissues have been separated and no other sequences in the upstream, downstream or coding regions have been identified that are autonomously involved inyp3expression.

The divergently transcribed yolk protein genes (Yp1 and Yp2) of Drosophila melanogaster are expressed only in adult females, in fat body tissue and in ovarian follicle cells. Using an in vitro transcription assay, we have identified a single 12-bp DNA element that activates transcription from the promoters of both Yp genes. In vivo, this regulatory element is tissue specific: it activates transcription of Yp1 and Yp2 reporter genes in follicle cells but has no detectable effect in fat body or other tissues. The sequence of the element consists of two recognition sites for the GATA family of transcription factors. We show that among the Drosophila genes known to encode GATA factors, only dGATAb is expressed in ovaries. The single transcript that we detect in ovaries is alternatively spliced or initiated to produce an ovary-specific isoform of the protein. Bacterially expressed dGATAb binds to the 12-bp element; a similar binding activity is also present in the Kc0 nuclear extracts used for in vitro transcription assays. These in vitro and in vivo results lead us to propose that dGATAb makes several developmentally regulated products, one of which is a follicle cell-specific protein activating transcription of Yp1 and Yp2 from a known regulatory element.

In rats fasted after the removal of about seven-tenths of the liver the remaining fragment undergoes ordinarily a marked simple hypertrophy and attains the weight reached through a process of simple atrophy by the entire liver of fasting controls. Under circumstances of exceptionally severe inanition, the hypertrophy may not occur, the fragment remaining unchanged or even undergoing a slight atrophy. But since in comparable controls the hepatic atrophy is extreme, the duplication in the liver weights still manifests itself. It is difficult to avoid the conclusion that the liver changes which take place in fasting animal are essentially conditioned on functional demands made upon the organ. Yet great as is the hepatic atrophy then occurring, this does not necessarily mean that these demands are much lessened during inanition. For a comparison of the liver weight with that of the soft body (net body weight less the weight of the ligamentous skeleton) brings out the fact that the organ is, relatively speaking, three-fourths as large at extreme inanition as in the well nourished individual. Of the missing fourth a part may be foodstuffs on temporary deposit. And the rest is perhaps to be accounted for by the existence of special conditions during inanition which favor the vicarious assumption of a part of the usual liver work by the other tissues. While the findings show that the liver size is essentially dependent on functional demands, they do not enable one to say whether the special demands in question come normally to the organ by way of the portal stream.

Changes in gonadal condition and plasma levels of gonadotropin and sex steroids were measured in goldeye (Hiodon alosoides) captured from the North Saskatchewan River, Edmonton, Alberta, from May to October in both 1983 and 1984. Mature females and spermiated males were captured in May and June and spent fish were captured in June. Gonadal recrudescence occurred over the summer and by October, relative gonad weights had reached 70% (females) and 90% (males) of the prespawning maxima. In females, plasma calcium was significantly elevated in spent fish in June, and in August there was a decrease in liver weight. Plasma levels of gonadotropin were highest in females during June, and in males during May. 17α,20β-Dihydroxy-4-pregnen-3-one was elevated in preovulatory or spent females and prespawning males, then decreased throughout the summer, whereas testosterone was only detectable in the plasma of prespawning fish. Estradiol-17β and estrone were not detectable in the plasma of females at any stage, including the period of vitellogenesis.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

This study was initiated on 24 prepubertal Holstein x Kankrej crossbred heifers of nearly identical age (7-9 months) and body weight (130-140 kg) at University farm to evaluate the effect of high plane of nutrition on blood biochemical and minerals profile and the age at puberty. Twelve heifers were managed under routine farm feeding (control) and the rest 12 under ideal optimum feeding regime (treatment) that included extra 1 kg concentrate, 30 g min mix and ad-lib dry fodder. The body weight and ovarian ultrasonography together with blood sampling was carried out at monthly interval from 10 to 18 months of age to study the ovarian dynamics and blood biochemical changes. High plane of nutrition to growing heifers was beneficial in reducing the age of onset of puberty (by 2-3 months) compared to routine farm fed group. The mean plasma total protein and cholesterol concentrations showed a rising trend with significant variations from 10 to 16 months of age, where it got mostly stabilized indicating adult profile. The activity of enzymes GOT and GPT also rose gradually and significantly from 10 months till 14-15 months of age, and thereafter it remained more or less static till 18 months of age. The levels of both these enzymes were higher, with lower protein and cholesterol, in control than the treatment group from 15-16 months of age onwards. The mean plasma levels of both calcium and phosphorus increased gradually and significantly with advancing age till 16-17 months of age, with little higher values in supplemented than a control group. The plasma levels of zinc, iron, copper, and cobalt also showed rising trend with significant differences between 10th and 12th-14th months of age, and from 15th to 18th months of age the levels were statistically the same in all the groups with slightly higher values in the treatment group.
