Recent advances in molecular genetics have made it possible to approach the study of complex polygenic multifactorial diseases such as hypertension. Two major approaches, the candidate gene and genetic mapping, have been used to investigate models of genetic hypertension in the rat.

BackgroundA robust body of evidence from family, twin and adoption studies demonstrates the importance of genes in the pathogenesis of bipolar disorder. Recent advances in molecular genetics have made it possible to identify these susceptibility genes.AimsTo present an overview for clinical psychiatrists.MethodReview of current molecular genetics approaches and emerging findings.ResultsOccasional families may exist in which a single gene plays a major role in determining susceptibility, but the majority of bipolar disorder involves more complex genetic mechanisms such as the interaction of multiple genes and environmental factors. Molecular genetic positional and candidate gene approaches are being used for the genetic dissection of bipolar disorder. No gene has yet been identified but promising findings are emerging. Regions of interest include chromosomes 4p16, 12q23–q24, 16p13, 21q22, and Xq24–q26. Candidate gene association studies are in progress but no robust positive findings have yet emerged.ConclusionIt is almost certain that over the next few years the identification of bipolar susceptiblity genes will have a major impact on our understanding of disease pathophysiology. This is likely to lead to major improvements and treatment in patient care, but will also raise important ethical issues.

Phytophthora root rot (PRR) causes serious annual soybean yield losses worldwide. The most effective method to prevent PRR involves growing cultivars that possess genes conferring resistance to Phytophthora sojae (Rps). In this study, QTL-sequencing combined with genetic mapping was used to identify RpsX in soybean cultivar Xiu94-11 resistance to all P. sojae isolates tested, exhibiting broad-spectrum PRR resistance. Subsequent analysis revealed RpsX was located in the 242-kb genomic region spanning the RpsQ locus. However, a phylogenetic investigation indicated Xiu94-11 carrying RpsX is distantly related to the cultivars containing RpsQ, implying RpsX and RpsQ have different origins. An examination of candidate genes revealed RpsX and RpsQ share common nonsynonymous SNP and a 144-bp insertion in the Glyma.03g027200 sequence encoding a leucine-rich repeat (LRR) region. Glyma.03g027200 was considered to be the likely candidate gene of RpsQ and RpsX. Sequence analyses confirmed that the 144-bp insertion caused by an unequal exchange resulted in two additional LRR-encoding fragments in the candidate gene. A marker developed based on the 144-bp insertion was used to analyze the genetic population and germplasm, and proved to be useful for identifying the RpsX and RpsQ alleles. This study implies that the number of LRR units in the LRR domain may be important for PRR resistance in soybean.

The issue of ‘knowability’ in relation to the origins and distribution of the language families of the world is addressed, and recent advances in historical linguistics and molecular genetics reviewed. While the much-debated problem of the validity of the concept of the language ‘macrofamily’ cannot yet be resolved, it is argued that a time depth for the origins of language families greater than the conventional received figure of c. 6000 years may in some cases be appropriate, allowing the possibility of a correlation between language dispersals and demographic processes following the end of the Pleistocene period. The effects of these processes may still be visible in the linguistic ‘spread zones’, here seen as often the result of farming dispersals, contrasting with the linguistic ‘mosaic zones’ whose early origins may sometimes go back to initial colonization episodes during the late Pleistocene period. If further work in historical linguistics as well as in archaeology and molecular genetics upholds these correlations a ‘new synthesis’, whose outlines may already be discerned, is likely to emerge. This would have important consequences for prehistoric archaeology, and would be of interest also to historical linguists and molecular geneticists. If, however, the proposed recognition of such patterning proves illusory the prospects for ‘knowability’ appear to be less favourable.
