Mentalizing Functions Provide a Conceptual Link of Brain Function and Social Cognition in Major Mental Disorders

Defining synaesthesia has proven to be a challenging task as the number of synaesthesia variants and associated phenomena reported by synaesthetes has increased over the past decade or so. This chapter discusses the inclusion of non-sensory concurrents in the category of synaesthesia. For example, many grapheme-colour synaesthetes also attribute gender and personality to letters and numbers consistently and involuntarily. Here we assess the question of including synaesthetic personification as a type of synaesthesia. We also discuss the relationship between synaesthetic personification and other instances of personification and mentalizing. We hope to convince readers that whether or not they embrace atypical forms of personification as a synaesthesia variant, studying the phenomenon is a worthwhile effort that could yield novel insights into human cognition and brain function.

After decades of neuroscientific research and taxonomic endeavour in psychiatry the quest for biologic markers specific enough to accommodate nosologic categories has not succeeded. Yet, neurophysiology and neuroimaging have developed pwoerful tools to investigate brain function. An immense amount of data has been accumulated regarding normal and pathologic information processing, cognition, emotion and other domains. Some have been correlated with genes underpinning diseases and are candidate endophenotypes. These stand at an intermediate level between genes and phenotype. They encompass several kinds of dysfunctions or abnormalities in brain structure. Rather than matching to singular diagnostic categories, as we devise them today, the same endophenotype is usually shared by distinct pathologic entities. Assuming tha they reflect specif dysfunctions this raises critical questions regarding the DSM way of classifying mental disorders and to the understanding of the neurobiologic phenomena underlying them. It is the purpose of this presentation to discuss these questions and review some of the data, including our own, concerning event-related potentials endophenotypes of psychosis with special focus on the schizophrenia-bipolar dichotomy and present.

AbstractOxytocin is a neuromodulator and hormone that is typically associated with social cognition and behavior. In light of its purported effects on social cognition and behavior, research has investigated its potential as a treatment for psychiatric illnesses characterized by social dysfunction, such as schizophrenia and bipolar disorder. While the results of these trials have been mixed, more recent evidence suggests that the oxytocin system is also linked with cardiometabolic conditions for which individuals with severe mental disorders are at a higher risk for developing. To investigate whether the oxytocin system has a pleiotropic effect on the etiology of severe mental illness and cardiometabolic conditions, we explored oxytocin’s role in the shared genetic liability of schizophrenia, bipolar disorder, type-2 diabetes, and several phenotypes linked with cardiovascular disease and type 2 diabetes risk using a polygenic pathway-specific approach. Analysis of a large sample with about 480,000 individuals (UK Biobank) revealed statistically significant associations across the range of phenotypes analyzed. By comparing these effects to those of polygenic scores calculated from 100 random gene sets, we also demonstrated the specificity of many of these significant results. Altogether, our results suggest that the shared effect of oxytocin-system dysfunction could help partially explain the co-occurrence of social and cardiometabolic dysfunction in severe mental illnesses.

In this paper, we review studies using functional neuroimaging to examine cognition in neuropsychiatric disorders. The focus is on social cognition, which is a topic that has received increasing attention over the past few years. A network of brain regions is proposed for social cognition that includes regions involved in processes relevant to social functioning (for example, self reference and emotion). We discuss the alterations of activity in these areas in patients with autism, depression, schizophrenia, and posttraumatic stress disorder in relation to deficits in social behaviour and symptoms. The evidence to date suggests that there may be some specificity of the brain regions involved in these 4 disorders, but all are associated with dysfunction in the amygdala and dorsal cingulate gyrus. Although there is much work remaining in this area, we are beginning to understand the complex interactions of brain function and behaviour that lead to disruptions of social abilities.

Introduction: There has long been an interest in the effects of diet on mental health, and the interaction of the two with stress; however, the nature of these relationships is not well understood. Although associations between diet, obesity and the related metabolic syndrome (MetS), stress, and mental disorders exist, causal pathways have not been established. Methods: We reviewed the literature on the relationship between diet, stress, obesity and psychiatric disorders related to stress. Results: Diet and obesity can affect mood through direct effects, or stress-related mental disorders could lead to changes in diet habits that affect weight. Alternatively, common factors such as stress or predisposition could lead to both obesity and stress-related mental disorders, such as depression and posttraumatic stress disorder (PTSD). Specific aspects of diet can lead to acute changes in mood as well as stimulate inflammation, which has led to efforts to assess polyunsaturated fats (PUFA) as a treatment for depression. Bidirectional relationships between these different factors are also likely. Finally, there has been increased attention recently on the relationship between the gut and the brain, with the realization that the gut microbiome has an influence on brain function and probably also mood and behavior, introducing another way diet can influence mental health and disorders. Brain areas and neurotransmitters and neuropeptides that are involved in both mood and appetite likely play a role in mediating this relationship. Conclusions: Understanding the relationship between diet, stress and mood and behavior could have important implications for the treatment of both stress-related mental disorders and obesity.

This manuscript has been published in the Journal of Abnormal Psychology (https://psycnet.apa.org/doi/10.1037/abn0000540).Biased social cognition towards an enhanced processing of negative social information might contribute to instability in interpersonal relationships. Such interpersonal dysfunctions are important for the understanding of several mental disorders, among them borderline personality disorder (BPD). To experimentally test enhanced memory retrieval of negative social information, using a newly developed variant of a looking-at-nothing paradigm, 45 BPD patients and 36 healthy women learned positive and negative personality traits of different target persons. In a translational memory test, participants were asked to use the learned information to evaluate statements about the target person. In addition to behavioral measures of memory performance, we investigated eye gaze patterns to decompose memory retrieval processes. We hypothesized that BPD patients would retrieve negative as compared to positive person information more accurately than healthy controls, and show increased eye gaze towards spatial locations where negative information was provided during the learning phase. Results pointed to a more accurate retrieval of negative person attributes in the patient group as compared with healthy controls (HC), thereby corroborating a negativity bias in social cognition in an exemplary sample of patients with interpersonal problems. Interestingly, the observed negativity bias for person memory was associated with BPD severity, stronger expectancies to be rejected by others, and social detachment. No group differences regarding eye fixation behaviour were found. We propose that enhanced retrieval of negative person information might be associated with dysfunctional cognitive schemas as well as reduced behavioral trust, and be of relevance for mental disorders characterized by interpersonal difficulties.

I briefly outline how computational models of cognitive brain function may be used as a resource to further develop and experimentally test hypotheses concerning “autism-psychosis spectrum disorders.”1
