Molecular phylogenetic identification of two new brown-rot corticioid fungi in Dacryobolus from southwestern China

Two new brown-rot corticioid fungi, Dacryobolus gracilis and D. montanus, are described from Chongqing Municipality and Guangxi Autonomous Region, southwestern China. Dacryobolus gracilis is characterized by fragile, ceraceous basidiocarps, odontioid hymenophore with slender aculei, presence of long aculeal cystidia in the central part of the aculei and narrowly allantoid basidiospores; D. montanus is characterized by resupinate, ceraceous basidiocarps, odontioid hymenophore with thicker aculei, absence of cystidia or cystidioles, and narrowly allantoid basidiospores. Molecular phylogeny inferred from internal transcribed spacer (ITS) and ribosomal large subunit (nLSU) sequences data indicated that these two new species were clustered with the other species of Dacryobolus with a strong support. These two new species are described and illustrated in this paper. A key to known species of Dacryobolus is provided.

AbstractThe present paper describes the taxonomy of two new monogeneans, namely, Thaparocleidus longiphallus sp. n. and T. siloniansis sp. n., based on morphological, morphometric and molecular biological analysis, collected from the fish Wallago attu (Bloch & Schn.) and Silonia silondia (Ham.), respectively, at Meerut, UP, India. Genetic characterization of the two new species is based on sequence analyses of the rDNA 28S gene using neighbour-joining and maximum-parsimony techniques. These methods are congruent in depicting T. longiphallus sp. n. and T. siloniansis sp. n. as closely related species, but distinct from each other and forming a subclade with other species of the genus Thaparocleidus Jain, 1952. Secondary-structure models of the large subunit rDNA of the two species were also predicted using a combined comparative and thermodynamic approach. Molecular morphometric and phylogenetic relationships of the isolates of the Thaparocleidus species are discussed in detail.

Two new truffle species Barssia guozigouensis and B. luyashanensis are described and illustrated from North China based on morphological and molecular evidences. Morphologically, B. guozigouensis is diagnosed by its distinctly warty ascomata and solid gleba with small and irregular chambers, and B. luyashanensis is by its red brown ascomata with fine warts and gleba without chambers. Molecular phylogenetic analyses based on the ribosomal internal transcribed spacer region and 28S large subunit nrDNA supported the placement of the new species.

Two fresh collections of Discosia were made from dead leaves of Fagus sylvatica in Italy. As these collections could not be cultured, the fruiting bodies were directly used for sequencing using a Forensic DNA Extraction Kit. Based on analyses of the concatenated internal transcribed spacer regions of the nrDNA operon (ITS) and large subunit rDNA (LSU) gene sequences, as well as morphological characters, the fresh collections are introduced as two new species, namely D. italica and D. fagi. Phylogenetically, these two species are distinct from all other Discosia species. Morphologically, D. italica is somewhat similar with D. fagi, but can be distinguished using dimension of conidiomata and conidiogenous cells. Descriptions and illustrations of the new taxa are provided herein.

Laetiporus is a cosmopolitan genus of brown rot fungi. In this study, L.medogensis and L.xinjiangensis are described as new species from western China, based on morphological and molecular evidence. L.medogensis has only been found on gymnosperms so far and is distinguished by pinkish-buff to clay-buff pileal surface and buff-yellow pore surface, azonate to faintly zonate pileus and ellipsoid to ovoid basidiospores (5–6.2 × 4.2–5.2 μm). L.xinjiangensis is found on angiosperms and is characterised by pale-buff to clay-pink pileal surface, cream to light yellow pore surface, azonate to faintly zonate pileus, large pores (2–3 per mm) and small basidiospores (4.5–5 × 3–4.2 μm). The phylogeny of Laetiporus is reconstructed with multi-gene sequences including the internal transcribed spacer regions (ITS), the large subunit (nrLSU) and small subunit (nrSSU) of the nuclear ribosomal RNA gene, the small subunit of the mitochondrial rRNA gene (mtSSU), the translation elongation factor 1-α gene (EF-1α) and the second subunit of RNA polymerase II (RPB2). The results show that L.medogensis and L.xinjiangensis formed two distinct lineages belonging to Laetiporus. Illustrated descriptions of the two new species are presented. An identification key to species of L.sulphureus complex is provided.

Two leuctrid species of the Rhopalopsole magnicerca group are described as new, R. brevicula sp. nov. and R. jizushana sp. nov. from the Guangxi Autonomous Region of southern China, bordering Vietnam and Yunnan Province of southwestern China, respectively. The new species are compared with related taxa.

AbstractPissodes punctatussp.nov. and Pissodes yunnanensissp.nov., reared from Pinus armandii Franchet and Pinus yunnanensis Franchet (Pinaceae) in southwestern China, are described and illustrated. Both species attack the upper portions of the stems of young trees, and larvae feed in the phloem and cambium.

Species in the genus Tomentella are distributed throughout the temperate and tropical regions worldwide, but few studies associated with the taxonomy and phylogeny of this genus had been reported from Northwest China. In this paper, molecular phylogenetic analyses of the nuclear ribosomal ITS (internal transcribed spacer: ITS1-5.8S-ITS2) and LSU (large subunit: 28S) sequences combined with morphological characteristics identified three new species from Xinjiang Autonomous Region in Northwest China, which were named T. aurantispora, T. kanasensis, and T. schrenkiana. Similar macromorphological and anatomical characteristics are shared by these new species: arachnoid basidiocarps; byssoid sterile margins; utriform basidia with a clamp connection at the base; the absence of rhizomorphs and cystidia; and slightly thick-walled, subglobose to globose basidiospores. Among these new species, the color of the hymenophoral surface, the size of the basidiospores, and some other features can be used for species delimitation. The new species and closely related species in the phylogenetic tree were discussed, and a key to the identified species of Tomentella from China was provided.

A new poroid wood-inhabiting fungal species, Atraporiella yunnanensis sp. nov., is proposed based on morphological and molecular characters. The species is characterized by cream pore surface when dry, which is easy to separate from substrate and very rapidly stained dark brown to black when bruised; hyphal system monomitic with generative hyphae hyaline to pale brown, thin-walled, unbranched, interwoven; slightly allantoid basidiospores, 2.2–3 × 0.8–1.5 µm. The internal transcribed spacer (ITS) and the large subunit (LSU) regions of nuclear ribosomal RNA gene sequences of the studied samples were generated, and phylogenetic analyses were performed with maximum likelihood, maximum parsimony and bayesian inference methods. The phylogenetic analysis based on molecular data of ITS+nLSU sequences showed that Atraporiella yunnanensis belonged to the residual polyporoid clade, formed a monophyletic lineage with a strong support (100% BS, 100% BP, 1.00 BPP) and was closely related to A. neotropica, and then grouped with other related genera: Antrodiella, Pouzaroporia, Steccherinum and Xanthoporus. Both morphological and molecular characters confirmed the placement of the new species in Atraporiella.

Amalothrips bifurcus sp.n. is described from Southwestern China as the third species of Amalothrips, characterized by the forked sense cone on antennal segments III–IV. Rhamphothrips yunnanensis sp.n. is described as the second known species of Rhamphothrips in which the male has laterally on the mesosternum one thorn-like seta and one stout shaded seta. Two species, Amalothrips noorazlani Ng and Parexothrips palumai Mound, are newly recorded from China.
