Bacteria associated with tunicate, Polycarpa aurata, from Lease Sea, Maluku, Indonesia exhibiting anti-multidrug resistant bacteria. Biodiversitas 20: 956-964. Tunicate is a rich secondary metabolites producer with various biological activities whether as an original producer or produced by the associated microorganisms. In this study, a total of 11 tunicate specimens were identified as Polycarpa aurata with four color variations based on morphological characteristic and COI gene identification and BLAST analysis. The P. aurata associated-bacteria were isolated and tested for antimicrobial activity against multi-drug resistant (MDR) bacteria. A total of 86 axenic isolates were successfully purified. Furthermore, nine isolates (10.5%) exhibited antibacterial activity on preliminary screening. Nine prospective isolates were fermented in respective medium (Zobell 2216, modified M1 or modified ISP2 media) then extracted using ethyl acetate. The ethyl acetate extracts from liquid fermentation were tested against MDR Escherichia coli, MDR Bacillus cereus, Methicillin-Resistant Staphylococcus aureus (MRSA) and Methicillin-Sensitive and Staphylococcus aureus (MSSA). As a result, seven isolates (8.1%) still retained the activity at the extract concentration 150 µg/disk. Molecular analysis based on 16S rDNA sequencing revealed the most active isolates, TSB 47, TSC 10 and TSB 34 identified as Bacillus tropicus, Vibrio alginolyticus and Virgibacillus massiliensis, with BLAST homology 99%.

Both tetrahydroisoquinolines (THIQs) and oxindoles (OXs) display a broad range of biological activities including anti-cancer activity, and are therefore recognized as two privileged scaffolds in drug discovery. In the present study, 24 3′,4′-dihydro-2′H-spiro[indoline-3,1′-isoquinolin]-2-ones, designed as molecular hybrids of THIQ and OX, were synthesized and screened in vitro against 59 cell lines in the NCI-60 screen. Twenty compounds displayed weak to moderate inhibition of cell proliferation; among them, three compounds displayed at least 50% inhibition of cell proliferation. The compounds appeared to target primarily renal cell cancer lines; however, leukaemia, melanoma, non-small cell lung cancer, prostate, ovarian and even breast cancer cell lines were also affected. Therefore, this class of spirooxindoles may provide useful leads in the search for new anti-cancer agents.

Synthesis of 3,5-diaryltetrahydro-N- [(4’-nitroanilino)methyl-thiazine-1,1-dioxide andN-[(4’-methylanilino)methyl-]-1,4-thiazine-1,1-dioxides by condensing 3,5-diaryl-tetrahydro-1,4-thiazine-1,1-dioxide with formaldehyde and 4-nitroaniline/4-methylaniline in the presence of hydrochloric acid is reported. The structures of the synthesized compounds have been confirmed by elemental and spectral analysis. The preliminary screening of the compounds for their biological activities gives significant results

Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.
