The anticancer and free radical scavenging potential of the Unani medicine Agaricus albus L.

Abstract Objectives Traditional Unani medicine plays a major role in maintaining health in developing countries. For ages, Unani physicians have described many Classical Unani Formulations (CUF) for their anti-cancerous action (Dāfiʻ-i-saraṭān) in the human population. These formulations contain various individual drugs which also have anticancer activity. The present study was designed to screen and compare cytotoxic and antioxidant properties of different extracts of Agaricus albus L. (A. albus), a drug used in the Unani system of medicine on human breast cancer cell lines (MCF-7). Methods Cytotoxic effect was assessed using MTT assay kit and free radical scavenging activity was done by using DPPH assay. Aqueous, hydro-ethanol and methanolic extracts were used at different concentrations. Results All extracts were shown good antioxidant and cytotoxic activity. Among all extracts, maximum cytotoxic effect was observed in methanolic extract and aqueous extract at the concentration of 1,000 μg against MCF-7 cell line in comparison to paclitaxel. The maximum antioxidant activity was observed in the hydroethanolic extract at the concentration of 1,000 μg against the MCF-7 cell line in comparison to ascorbic acid. Conclusions The present study advocates, the anticancer and antioxidant property of A. albus and suggests that this drug may be used as a potential drug for cancer treatment after a successful and detailed preclinical study and clinical trial.

Heterogeneous method for grafting of syringaldehyde (SD) onto chitosan by laccase oxidation, yielding a product with improved antioxidant activities. The results showed that about 1.1% amino groups participated in this reaction. The FTIR showed that the absorption of amino groups were decreased, the absorption of benzene ring structures were disappeared and a new band appeared at 1637 cm-1 corresponding to C=N vibrations that were due to the Schiff base reaction between the chitosan and syringaldehyde. Additionally, the chitosan had insignificant reducing power and DPPH free radical scavenging activity, SD-chitosan had reducing power of A700=0.684, DPPH free radical scavenging activity of 53.2%. SD-chitosan presented improved antioxidant properties.

Objective: The main focus of this study is to screen the marine sponges for potent free radical scavenging activity. Methods: Various methods such as 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assay are employed to ascertain the antioxidant properties of marine sponges namely Dysidea herbacea and Sigmadocia pumila. Results: On analyzing, the result of ABTS assay D. herbacea and S. pumila exhibited almost equal antioxidant properties. While calculating the inhibitory concentration 50% value for DPPH assay, the Sample 1 and 2 has an IC of 655.49 and 826.739 μl, respectively, and in FRAP assay, the Sample 1 and 2 has an IC of 67.587 and 74.57 μg, respectively. Conclusion: Overall from this assay, D. herbacea revealed slightly better antioxidant activity when compared to S. pumila, also which in future may serve as a better source to fight against various diseases

<b><i>Introduction:</i></b> Peptic lesions usually develop when there is an imbalance between aggressive drivers and gastro-protective mediators that guard the lining of the gastrointestinal tract. The most crucial of these mediators are antioxidants, whose loss may predispose to oxidative stress, which is believed to be the main aggravator of several diseases including peptic ulcer. Proton pump inhibitors (PPIs) are drugs that are highly effective and widely used for therapeutic management of peptic disorders through inhibition of gastric acid secretion. In spite of this, oxidative damage may continue to be a major issue that can predispose to future lesions. <b><i>Objective:</i></b> The present study is designed to explore the possible antioxidant capability of different PPIs, including omeprazole, esomeprazole, lansoprazole, pantoprazole, and rabeprazole, in an aim to suggest an agent that, in addition to its acid-suppression properties, can provide antioxidant profit. <b><i>Methods:</i></b> The antioxidant activity of different PPIs was evaluated calorimetrically to test the ability of each drug to quench oxygen free radical, using the well-known stable free radical α,α-diphenyl-β-picrylhydrazyl (DPPH), and compared to ascorbic acid (AA; vitamin C). The measurements were performed using a spectrophotometer at 517 nm. <b><i>Results:</i></b> All the studied drugs reduced DPPH, but to different extents. However, omeprazole and esomeprazole showed the highest ability to scavenge free radicals (50% inhibitory concentrations [IC<sub>50</sub>s] of the percentage for free radical scavenging activity are 18.7 ± 5.7 and 18.7 ± 5.7, respectively, and the AA equivalents are 83,772 ± 11,887 and 81,732 ± 8,523 mg AA/100 g, respectively). Conversely, lansoprazole, pantoprazole, and rabeprazole might be having no role in this story (IC<sub>50</sub>s of the percentage for free radical scavenging activity are 49.3 ± 3.1, 49 ± 9.4, and 40.7 ± 7.2, respectively, and the AA equivalents are 30,458 ± 3,884, 32,222 ± 10,377, and 37,876 ± 8,816 mg AA/100 g, respectively). <b><i>Conclusion:</i></b> Thus, omeprazole and esomeprazole may confer a significant dual action in gastrointestinal protection by providing potent antioxidant properties in addition to their major role as acid-suppression agents. However, further studies are essential to elucidate the mechanism behind the difference between the drugs of the same class.

AbstractNine derivatives of three natural diarylheptanoids, curcumin, demethoxycurcumin and bisdemethoxycurcumin, were prepared. Their antioxidant, free radical scavenging, nitric oxide (NO) inhibitory and cytotoxic activities were evaluated and compared with those of the respective natural compounds. Curcumin (1), demethoxycurcumin (2), demethyldemethoxycurcumin (C3), diacetyldemethoxycurcumin (AC2) and triacetyldemethylcurcumin (AC5) exhibited higher antioxidant activity than quercetin while products from demethylation of 1 and 2 exhibited higher free radical scavenging activity. Compounds AC2 and AC5 were found to be most active in inhibiting breast cancer cells (MCF-7) proliferation with IC50 values of 6.7 and 3.6 μм, respectively. The activity of AC2 is almost doubled and of AC5 almost tripled as compared to curcumin. Their selectivity towards different cell lines is also more noticeable. Compounds AC2 and AC5 also showed increased activity against a human prostate cancer cell line (DU-145) and non-small lung cancer cell line (NCI-H460) with IC50 values of 20.4, 16.3 and 18.3, 10.7 μм, respectively.

Ethanol extracts from Hohenbuehelia serotina and Armillaria mellea fruit bodies were prepared and investigated for their antioxidant capacities in three different assays, namely, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, reducing power and chelating ability on ferrous ions. Among the two mushrooms, ethanol extracts from A. mellea was more effective than that from H. serotina in scavenging DPPH free radical ability, while ethanol extracts from H. serotina was some higher than that from A. mellea in reducing power. With regard to chelating effect on ferrous ions, at 5 mg/mL, chelating ability of ethanol extracts from H. serotina were 43.17 %, while that from A. mellea was 46.54%. It can be concluded that ethanol extracts from A. mellea and H. serotina were good in antioxidant properties.

To investigate the fractions of the chloroform extract of Chrysopogon zizanioides L by column and thin layer chromatography and antioxidant activity of fractions of chloroform extract of Chrysopogon zizanioides L. The extract was screened for possible antioxidant activities by free radical scavenging activity (DPPH) method. The results showed that all the plant fractions possessed antioxidant properties including radical scavenging activities. The anti-oxidative activities of four compounds were compared with the standard ascorbic acid. The compound four of Chrysopogon zizanioides L was more effective than that of other three compounds. This study suggests that the four compounds from Chrysopogon zizanioides L chloroform extract exhibit great potential for antioxidant activity and may be useful for their nutritional and medicinal functions.

ABSTRACTObjectives: To analyze the phytochemicals quantitatively and screening the antioxidant efficacy of methanol extracts of Elytraria acaulis plant extractby in vitro.Methods: The total phenols, flavonoids, and tannin contents analyzed by Folin-Ciocalteu method, aluminum chloride colorimetric and calculated asgallic acid equivalents/g (GAE/g) of dry weight. Free radical scavenging activity was screened using 1,1-diphenyl-2-picrylhydrazyl (DPPH), radicalscavenging activities, and ferric reducing power assay was also analyzed.Results: The total phenolic content of the aqueous leaf extract was 46.84 µg GAE/g of extract powder. Significantly similar levels of total flavonoid andtannin contents of the plant were 41.72 and 39.50 µg GAE/g, respectively. The plant extracts showed appreciable free radical scavenging activities atthe highest concentration of 400 µg/mL superoxide anion radical with IC values (107.7±1.081 µg/mL), and for DPPH and considerably high amountof radical scavenging activity was found with very low IC5050 values (4.3±0.88 µg/mL) compared with quercetin.Conclusion: The phenolic and flavonoid compounds provide substantial antioxidant properties which could be effectively used for pharmaceutical,nutraceutical as well as anti-inflammatory applications.Keywords: Phenols, Flavonoids, 1,1-diphenyl-2-picrylhydrazyl, Pharmaceuticals.

A comparison of total polyphenols (TP) and flavonoids (TF) contents, antioxidant activity, and silver nanoparticles (AgNPs) biosynthesis potentiality of 35 common edible fruits in Bangladesh were studied. Among the fruits, Emblica officinalis showed the highest content of TP (89 mg gallic acid equivalent (GAE)/g powder), reducing power (OD, 1.98 at 1 mg powder/ml), and 2,2-diphenyl-1-picrylhydrazyl (DPPH)-free radical scavenging activity (95.6% at 0.5 mg powder/ml) whereas the highest content (17.3 mg (+)-catechin equivalent (CE)/g powder) of TF was recorded in Garcinia cowa. The inhibitory concentration 50 (IC50) for DPPH-free radical scavenging of Annona reticulata, E. officinalis, G. cowa, Phoenix sylvestris, Psidium guajava and Syzygium cumini were 180, 21.8, 172, 188.9, 162.4 and 176.2 μg powder/ml, respectively. Litchi chinensis showed the highest total antioxidant capacity (79 mg GAE/g powder) followed by E. officinalis (69 mg GAE/g powder). The fruits showed a very strong correlation (r2 = 0.99) between TP and reducing power. P. guajava showed the highest potentiality (OD, 0.74 at 10 mg powder/ml) in the biosynthesis of AgNPs followed by S. cumini, A. reticulata and P. sylvestris. Therefore, most potential five fruits should be graded as E. officinalis > S. cumini > P. guajava > A. reticulata > P. sylvestris. Bangladesh J. Bot. 50(3): 445-451, 2021 (September)
