AbstractThe insecticidal efficacy of the experimental bioinoculants (Natur Nova, Natur Agro Hungary Ltd., Hungary) containing Beauveria bassiana (Bals.-Criv). Vuill. and Metarhizium anisopliae Metschnikoff against stored product pests modelled by granary weevil, Sitophilus granarius L. was evaluated on wheat in laboratory tests. In order to comparability, two applications were also set up: one purely ash-treated grain treatment against wireworms and a soil-injected one. Our results confirmed that some biopesticides can exert unexpected effect on non-target arthropods. However, the examined biopesticides were as empirically effected against the target arthropods regarding wireworms. Nevertheless, the application exempt from ash of the examined bioinoculants Natur Nova containing B. bassiana and M. anisopliae were not suitable for the elimination of the stored product pests modelled by S. granarius. In contrast, the putative plant protection effect of these biopesticides was observed in the course of applying the agents in the recommended environment and pest species as well as when applying them with ash carrier.

Abstract Laboratory bioassays were conducted to assess the insecticidal efficacy of the formulation SilicoSec® used alone or in combination with isolates of entomopathogenic fungi, Metarhizium anisopliae (Metschinkoff) Sorokin and Beauveria bassiana (Balsamo) Vuillemin. SilicoSec® is a commercial diatomaceous earth (DE) formulation. Wheat was treated with 200 mg/kg of DE, 400 mg/kg of each isolates alone or a combination of them, against Tribolium castaneum Herbst, Rhyzopertha dominica (F.), and Oryzaephilus surinamensis L. The experiments were carried out at 27±1°C and 65±5% relative humidity (RH) in continuous darkness. The pathogenicity of all isolates was significantly low even after 7 days of exposure, with the exception of R. dominica. The isolates were virulent to the beetles, but the efficacy of the isolates was enhanced in combination with the DE. Tribolium castaneum was the most resistant species, followed by R. dominica. The findings indicated that the addition of the DE to the isolates increased the pathogenicity especially at the highest exposure interval. The addition of DE may provide satisfactory control of the insect-pests of stored products.

Laboratory tests were carried out to evaluate the insecticidal efficacy of different natural inert dusts (diatomaceous earth, wood ash, quartz sand) and the leaf powder of Azadirachta indica A. Juss. against granary weevil (Sitophilus granarius [L.]) adults. The efficacy of the substances was tested individually and in combination with each other. The substances were applied at different concentrations, and bioassays were carried out at four different temperatures (20, 25, 30 and 35°C) and two different relative humidity (RH) levels (55% and 75%). The adult mortality was recorded after the 7th, 14th and 21st days of exposure. The progeny production of individuals exposed to different combinations was also assessed. Wood ash proved to be the most efficient inert dust in our research. We detected 100% mortality in the treatment exposed to a higher concentration (5 w%) of wood ash at 35°C and 55% RH after 7 days of exposure. A lower RH level had also a negative impact on the progeny production. We can conclude that wood ash can be efficient in controlling granary weevil adults as a single substance or in combination with other substances. Further surveys should focus on the impact of the wood ash dose rates. Due to the high percentage of area covered with forest in some European countries, the main ingredient is present locally, but additional surveys are needed to help improve the practical use of wood ash.

Sitophilus granarius (L.) is an important pest of stored grain worldwide. In recent years, sustainable methods against it have received attention as grain stock protective means. Our aim was to obtain information about the efficacy of infrared irradiation (IR) against S. granarius in laboratory conditions. The change in adult-activity and median lethal dose (LD50) triggered by IR in S. granarius was examined. The insecticidal efficacy in the infested grains was also analyzed at 12, 24, 48, and 72h following exposure to IR (250W), and the progeny-production was assessed 45 days upon the treatment. Based on our findings, total mortality ensued in a grain stock of 50 g at 412 s and a for 100 g grain at 256 s. A significant increase in S. granarius mortality could be observed in the higher grain weight regime, which can be accounted for by the higher heat-absorbance of objects with higher weight. The activity of pests immediately after the beginning of IR increased and subsequently became moderated. The observation of activity-peak brought about by irradiation contribute to the optimization of chemical intervention. This treatment could provide an effective and sustainable technique in integrated pest management.

The insecticidal virulence of various entomopathogenic fungal isolates retrieved from soil samples was tested on adults of the granary weevil Sitophilus granarius (L.) (Coleoptera: Curculionidae). Bioassays were carried out in the laboratory where experimental adults were sprayed with 1 mL of conidial suspension (108 conidia/mL) from each isolate. Mortality was recorded at 7, 14, and 21 days after exposure. Mean mortality, mean lethal time, survival, and hazard effect were estimated for each isolate. Two isolates of Beauveria bassiana (Balsamo) Vuillemin (Hypocreales: Cordycipitaceae), one isolate of Aspergillus insuetus (Bainier) Thom & Church (Eurotiales: Trichocomaceae) and Metarhizium anisopliae (Metschinkoff) Sorokin (Hypocreales: Clavicipitaceae) resulted in the highest mortality (97–100%). The isolates with both the highest hazard effect and the lowest survival rate were Aspergillus sp. and M. anisopliae. Our results indicate that entomopathogenic fungi have the potential to become a very useful tool in reducing chemical applications in storage facilities.

The granary weevil Sitophilus granarius is a stored product pest found worldwide. Environmental damages, human health issues and the emergence of resistance are driving scientists to seeks alternatives to synthetic insecticides for its control. With low mammal toxicity and low persistence, essential oils are more and more being considered a potential alternative. In this study, we compare the toxicity of 25 essential oils, representing a large array of chemical compositions, on adult granary weevils. Bioassays indicated that Allium sativum was the most toxic essential oil, with the lowest calculated lethal concentration 90 (LC90) both after 24 h and 7 days. Gaultheria procumbens, Mentha arvensis and Eucalyptus dives oils appeared to have a good potential in terms of toxicity/cost ratio for further development of a plant-derived biocide. Low influence of exposure time was observed for most of essential oils. The methodology developed here offers the possibility to test a large array of essential oils in the same experimental bioassay and in a standardized way. It is a first step to the development of new biocide for alternative management strategies of stored product pests.

En busca de una alternativa de control del mosquito Culex quinquefasciatus en áreas urbanas se desarrolló el proyecto: “Evaluación de microorganismos biocontroladores sobre el mosquito Culex quinquefasciatus” cuyo objetivo fue evaluar la capacidad biocontroladora de diferentes hongos entomopatógenos sobre larvas del mosquito a nivel de laboratorio. Se evaluaron 16 aislamientos de hongos en diferentes concentraciones. Se constataron diferencias en el porcentaje de mortalidad producido en larvas de Cx. quinquefasciatus dependientes del tipo de aislamiento, de la concentración utilizada y del tiempo de exposición. Los aislamientos que presentaron el nivel de mortalidad más alto pertenecen a la especie Metarhizium anisopliae, seguidos por aislamientos de la especie Beauveria bassiana. El aislamiento que presentó el mayor nivel de mortalidad (100%) en menor tiempo de exposición (24 horas) fue Metarhizium anisopliae 99068 en una concentración de 1x106 e/ml. Por lo tanto este aislamiento es recomendado para su evaluación en campo y desarrollo de un producto comercial.

Abstract Background The diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), is a noxious pest of cruciferous crops all over the world causing serious economic damage. Management of insect pest generally depends on chemical control; however, due to development of resistance against all types of insecticides, alternative approaches especially utilization of a microbial agent is inevitable. Results Potential of 2 entomopathogenic fungi (EPF), viz., Beauveria bassiana and Metarhizium anisopliae, was evaluated against 2nd and 3rd larval instars of P. xylostella by adopting leaf dip and direct spraying methods under laboratory conditions. Significant mortality rate was achieved by each fungus under adopted methodologies. However, B. bassiana was found to be more effective in both conditions than M. anisopliae. Highest mean corrected mortality (77.80%) was recorded, when spores of B. bassiana were sprayed on the 2nd instar larvae (LC50=1.78×104/ml) after the 6th day of treatment. Similarly, incase of M. anisopliae LC50 for the 2nd instar at the same methodology was 2.78×104/ml with a mortality percentage of 70.0%. Offspring sex ratio was non-significantly related to treatment concentration and methodology, except for the control. Conclusion Beauveria bassiana and M. anisopliae had potential to suppress P. xylostella infestations when applied appropriately. Present findings suggested that B. bassiana and M. anisopliae when sprayed on immatures of host insect had more effect as compared to leaf dip procedure. Furthermore, no significant effect of concentrations was observed on sex ratio.
