Hematological parameters, intraerythrocytic phosphates, hemoglobin, and whole blood Bohr effect of the South American armored catfish Hoplostenum littorale were studied during different seasons of the year. In addition, the degree of dependence on air breathing was determined for this species. The hematological parameters presented seasonal variations, which were not correlated to oxygen, temperature, and water level oscillations. Five anodic hemoglobin fractions were detected in starch gel electrophoresis. In addition to ATP, GTP and Fe-GTP being detected, 2,3-DPG was also detected in red blood cells of H. littorale. The latter is an intraerythrocytic phosphate characteristic to red blood cells of mammalians. The increased production of 2,3-DPG could be associated with decreasing Hb-O2 affinity and both features could be related to environmental temperature increase. Whole blood Bohr effect was influenced by water temperature. This study confirms H. littorale to be continuous and not obligate air breather, under all dissolved oxygen level conditions.

Hemoglobin, hematological parameters, intraerythrocytic phosphates, and whole blood Bohr effect of Mylossoma duriventris captured in the Amazon river were studied during five different periods of the year. Some important variations of these parameters are described and can be correlated to oxygen and water level oscillations. Four anodal hemoglobin fractions were detected on starch gel electrophoreses. ATP was the major intraerythrocytic phosphate during February, April, and August while GTP was the major phosphate during October and November. These two periods differ also in terms of water level and available oxygen. Whole blood Bohr effect is directly influenced by ATP and GTP concentrations.

Whole blood was tapped from the embryos of anaesthetized pregnant New Zealand rats using a micropipette of approximately 250 μm diameter, inserted through the uterine wall via a 5-mm incision in the anterior abdominal wall. The procedure was repeated 3 times (days 15, 17 and 19 of fetal life) in the same pregnant rats, and thereafter, twice in the surviving newborns (days 14 and 30 of neonatal life). Osmotic fragility of the red blood cells (RBC), reticulocyte counts and haemoglobin values of each sample were analysed. The fragility curve of the fetuses showed the presence of 2 types of cell population: a fragile and a more resistant type. This was correlated with the number of immature RBC (reticulocytes) in the samples. Starch gel electrophoresis of the haemoglobin revealed 4 fractions in both the fetuses and newborn animals.

Background: The hypoxic environment stimulates the human body to increase the levels of hemoglobin (HGB) and hematocrit and the number of red blood cells. Such enhancements have individual differences, leading to a wide range of HGB in Tibetans’ whole blood (WB). Study Design: WB of male Tibetans was divided into 3 groups according to different HGB (i.e., A: >120 but ≤185 g/L, B: >185 but ≤210 g/L, and C: >210 g/L). Suspended red blood cells (SRBC) processed by collected WB and stored in standard conditions were examined aseptically on days 1, 14, 21, and 35 after storage. The routine biochemical indexes, deformability, cell morphology, and membrane proteins were tested. Results: Mean corpuscular volume, adenosine triphosphate, pH, and deformability were not different in group A vs. those in storage (p > 0.05). The increased rate of irreversible morphology of red blood cells was different among the 3 groups, but there was no difference in the percentage of red blood cells with an irreversible morphology after 35 days of storage. Group C performed better in terms of osmotic fragility and showed a lower rigid index than group A. Furthermore, SDS-PAGE revealed similar cross-linking degrees of cell membrane protein but the band 3 protein of group C seemed to experience weaker clustering than that of group A as detected by Western Blot analysis after 35 days of storage. Conclusions: There was no difference in deformability or morphological changes in the 3 groups over the 35 days of storage. High HGB levels of plateau SRBC did not accelerate the RBC change from a biconcave disc into a spherical shape and it did not cause a reduction in deformability during 35 days of preservation in bank conditions.

AbstractObjectivesAnalysis of platelet glycoprotein (GP) expression by flow cytometry is applied for diagnostic confirmation of GP-associated thrombocytopathies. While platelet-rich plasma may be used for distinct identification of target events, this strategy is not feasible for small sample volumes or for patients showing low platelet counts and/or giant platelets. However, also the use of whole blood (WB) is hampered by the difficulty to discriminate platelets from red blood cells (RBC) in such patients. To circumvent these limitations, we evaluated the feasibility of a RBC gating-out strategy.MethodsIn addition to platelet GPIb, GPIIa/IIIa, as well as P-selectin (CD62P), citrated whole blood (CWB) samples were stained for RBC-specific glycophorin A (CD235a). CD235a-negative platelet events were further discriminated by forward-/side-scatter characteristics and platelet GP expressions analyzed relative to that of a healthy control sample processed in parallel.ResultsEstablished reference intervals allowed for clear identification of decreased GPIIb/IIIa- or GPIb expression pattern in samples of patients with confirmed Glanzmann thrombasthenia or Bernard–Soulier syndrome, respectively. It could be shown that the analysis of 2,500 platelet events is sufficient for reliable GP expression analysis, rendering the proposed method applicable to samples with low platelet counts.ConclusionsThis study demonstrates the feasibility of CD235a-based exclusion of RBC for platelet GP expression analysis in CWB. In contrast to direct staining of platelet-specific antigens for target identification, this indirect gating out approach is generally applicable independent of any underlying platelet GP expression deficiency.

Purpose: to study hematological parameters of blood in bulls-producers of dairy breeds on the day of semen collection in the Udmurt Republic after a long winter period of operation, depending on the breeds, age and place of selection.Materials and methods. The work was performed at the Federal Research Center for Animal Husbandry named after Academy Member L. K. Ernst on the basis of AO "Udmurtplem" of the Udmurt Republic in the period from 2020 to 2021 on dairy bulls (n=20) aged 15-69 months, including a purebred Holstein breed of domestic selection (n=6), a Holstein black-and-white breed of European selection (the Netherlands, n=6), a black-and-white breed with blood transfusion on Holsteins on at the level of 94-98% (n=8). The content of white blood cells, red blood cells, hemoglobin and hematocrit, depending on age and breed, was studied on the ABC VET hematological analyzer on the day of taking the seed.Results. It was found that on average, in 20 bulls aged 15-69 months, the level of white blood cells was at the level of reference values of 8.8±0.25 x 109/l, red blood cells 10.3 x 1012/l, which is 50% more than the reference values. The concentration of hemoglobin is 128.0±2.92 g/l, hematocrit is on average 54.3%, with a norm of 24-46%. Breeding bulls at a reliable level, differing in age, showed that some animals had high indicators for red blood cells of 10.3±0.26 x 102, for hematocrit of 54.3% against 24-46% of reference values. Also, according to hematological indicators, there was a tendency to increase the concentration of hemoglobin and hematocrit in European-bred bulls.Conclusion. The study of the variability in hematological parameters depending on the selection showed that there is no significant difference in leukocytes and all indicators are at the level of reference values, and in erythrocytes more than 50% than the highest indicators. The highest hemoglobin values were at the level of 141-156 g/l instead of 128 g/l according to the highest reference values. Hematocrit also showed high max values in all groups in comparison with the reference values of 59-66% versus 46% in the norm. It is necessary when analyzing hematological blood parameters in addition to the average statistical indicators (M+m) also, monitor the variability (min-max) in order to obtain more objective information.
