Ubiquitin is required under both normal and stress conditions. Under stress conditions, upregulation of the polyubiquitin gene UBC is essential to meet the requirement of increased ubiquitin levels to confer stress resistance. However, UBC upregulation is usually observed only under stress conditions and not under normal conditions. Therefore, it has not been possible to upregulate UBC under normal conditions to study the effect of excess ubiquitin on cellular machinery. Recently, the CRISPR/Cas9 system has been widely used in biological research as a useful tool to study gene disruption effects. In this study, using an inducible CRISPR/Cas9 variant, a dCas9–VP64 fusion protein, combined with a single guide RNA (sgRNA) containing MS2 aptamer loops and MS2-p65-HSF1, we developed a system to increase the ubiquitin pool via upregulation of UBC. Although it is challenging to upregulate the expression of a gene that is already expressed at high levels, the significance of our system is that UBC upregulation can be induced in an efficient, reversible manner that is compatible with cellular processes, even under normal conditions. This system can be used to study ubiquitin pool dynamics and it will be a useful tool in identifying the role of ubiquitin under normal and stress conditions.

BackgroundThe rapid increase in research endeavour has not kept pace with the advent of well-publicised theories and treatments for autism.AimsTo explore some of the newer developments in biological research into autism.MethodA review of recent publications and presentations.ResultsThe concept is shifting from the narrow perception of aloof autism, described by Kanner, to a wider one that includes a spectrum extending to a broader, subclinical phenotype. The genetic basis has been established; now we need to discover the location and interaction of the relevant sites. There is considerable interest in the bowel as a pathogenetic agent, particularly in the effects of exogenous opioids and multiple viral infection (the latter posing a public health problem). Also of concern is the role of (potentially treatable) epilepsy, analogous to the Laudau-Kleffner syndrome.ConclusionsIn the absence of a cure, the implementation of ideas will continue to outstrip factual evidence. Clinicians are challenged by the availability of information (and misinformation), particularly on the internet.

In this article, we evaluate an initiative recently launched by the national government in Mexico to create ‘Patenting Centers’ in various universities and research institutions in diverse regions of the country. We focus particularly on elucidating how the installation of these Patenting Centers has augmented the number of national filings for intellectual property (IP) protection, and how the Centers have contributed to increasing the quality of IP applications. Furthermore, we analyze how the Mexican Patenting Centers have qualitatively contributed to fostering local cultures of innovation, for example through capacity-building activities directed towards scientific researchers. We also attempt to understand how the Patenting Centers have supported processes of technology transfer and commercialization, which we evaluate by examining a case study from the Northwest Biological Research Center (CIBNOR). Our findings indicate that the Mexican Patenting Centers have contributed to increasing IP protection activity in various regions of the country, and that they have augmented interactions between public research institutions and the productive sector. We conclude with suggestions for how the Patenting Center model may be further assessed in the future, to ensure that the government's mission of fostering endogenous innovation and the creation of a knowledge-based economy may continue to be realized.

The article discusses the organization of an academic institution in 1938 in Borok to conduct biological research and the role of Aleksandr Petrovich Shennikov, corresponding member of the USSR Academy of Sciences, in this process.

Methylglyoxal (MG) – is a high reactive α- oxoaldehyde, which can be synthesized through various biochemical processes in vivo. MG is capable of interacting with nucleophilic groups of proteins, lipids and nucleic acids which leads to their glycation. Covalent compounds that formed in this process were named advanced glycation end products. Advanced glycation end products play an important role in the pathogenesis of diabetes, chronic inflammation, cancer and Alzheimer's disease. One of the most stable compounds, which is formed by reacting of methylglyoxal with the nitrogenous bases of the DNA molecule is N2- carboxyethyl -2' - deoxyguanosine (CEdG). This compound can be used as a marker for monitoring of various diseases, investigation of the role of glycation end products in their pathogenesis, as well as to search for the therapeutic targets. To perform our molecular-biological investigations by mass spectrometry, we synthesized N2- carboxyethyl -2' - deoxyguanosine, and have proposed a new method for purifying compounds by reverse - phase HPLC. As a result, we achieved the optimal purity of the sample, while substantially reducing the cost of the purification procedure.
