Abstract Although oral cancer is multifactorial in origin only few had evaluated the diagnostic parameters for candidal infection in oral squamous cell carcinoma (OSCC). We aims to compare and assess the accuracy of Calcofluor Whilte (CFW) to that of conventional staining methods to identify the presence of candidal infection in OSCC. Archival collection of 43 OSCC were retrieved for this investigation. Standard staining protocol was followed for the index test (CFW) and reference standard (Periodic Acid Schiff). Two examiners were blinded for either one of the staining techniques. Diagnostic test evaluation and Kappa statistic was calculated using MedCalc software. The study comprised of 37 males, six females, and their mean age at the time of presentation was 51 (range 23 to 75 years). The sensitivity and specificity of CFW was 75.8% (CI 57.7 to 88.9%) and 10.0% (CI 0.2 to 44.5%). While the positive predictive and negative predictive values were 63.1% (CI − 50.6% − 74.0%) and 67.0% (CI − 49.2% − 81.0%). Our results show minimal agreement with PAS with a Kappa score of 0.148 (95% CI = 0.39 to 0.09). Overall detection rate was similar for both the index test and reference standard although there was considerable number of intermediate test results. Sensitivity of CFW was comparable to that of earlier studies but specificity was low and the degree of concordance of 60.4%. Although CFW staining can come with high false positive they are an useful test for ruling out candida infection when the test result is negative.

Objectives: The aim of the study is to compare the efficiency of Ki-67 with special stains in the stroma of different grades of Oral Squamous Cell Carcinoma (OSCC) and to evaluate the influence of these changes in predicting the prognosis of these tumors. Materials and Methods: A total of 30 cases of different grades of Oral Squamous Cell Carcinoma and 6 cases of control were sections and stained with Picrosirus red (PR), combined Alcian blue-periodic acid Schiff reagent (AB-PAS) and Immunohistochemical (IHC) marker Ki-67. Results: Collagen fibre nature using PR stain and proliferative activity of malignant epithelial cells using IHC marker Ki-67 was found to be statistically significant. Mucin presence using AB-PAS was statistically insignificant. Conclusion: Prognosis in different grades of oral squamous cell carcinoma can be accessed by change in collagen fibres birefringence, as the tumour progresses there is change from mature to immature collagen. Ki-67 is a good proliferative marker and shows that there is positive correlation with histological grading of oral squamous cell carcinoma. Presence of acidic or neutral mucin in OSCC needed to be further studied. For assessing the prognosis in different grades of OSCC, special stains can also be used. AbstractPurposeOral Squamous Cell Carcinoma (OSCC) is one of the most prevalent cancers in the world with maximum number of cases reported from India. Poor survival rate associated with OSCC can be attributed to non-availability of a biomarker, as one of the major reasons, leading to late presentation. Identification of an early diagnostic biomarker, which can also be used as a screening tool, will be helpful in reducing the disease morbidity and mortality.Experimental DesignIn this article we report Parallel Reaction Monitoring (PRM) based validation of 12 candidate proteins, identified initially by TMT tag based relative quantification of salivary proteins on LC-MS, in the saliva of Oral Squamous Cell Carcinoma (OSCC) cases (N=50) and healthy controls (N=49), AZGP1, AHSG, KRT6C, S100A7, S100A9, KLK1, BPIFB2, IGLL5, CORO1A, LACRT, LCN2 and PSAP. Heavy isotope labelled reference peptides were used to produce calibration curve and absolute quantification of proteins and resulting data was analyzed statistically using R.ResultsSalivary AHSG (p=0.0041**) and KRT6C (p=0.002**) were significantly upregulated in OSCC cases while AZGP1 (p=<0.0001***), KLK1 (p=0.006**) and BPIFB2 (p=0.0061**) were significantly downregulated. Multivariate logistic regression modelling resulted in a risk prediction model consisting of AZGP1, AHSG and KRT6C with p value <0.0001***. Using this model ROC with area under the curve of 82.2% was produced and sensitivity and specificity observed for this model was 78% and 73.5%. Positive and negative predictive values for the model were 76% and 75% respectively.ConclusionWe report a potential biomarker panel consisting of proteins AZGP1, AHSG and KRT6C for early diagnosis of OSCC.

A cross sectional study of 29 cases of oral squamous cell carcinoma with or without  cervical lymph node metastasis was done among Bangladeshi patients from January 2006 to December 2007. Majority of the study subjects (34.5%) belonged to the age group of 40-49 years. 58.6% of the study subjects were male, while remaining 41.4% of them were female. 51.7% of the lesions were located in the alveolar ridge where the other common sites were buccal mucosa (27.6%) and retro molar area (13.8%). Half of the study subjects (51.7%) were habituated to betel quid chewing followed by 37.9% and 10.3% were habituated to smoking and betel quid-smoking respectively. Grade I lesions was most prevalent (75.9%) in the study subjects.  Majority of cases presented with Stage IV lesions (55.2%). The sensitivity, specificity, positive predictive value, negative predictive value & accuracy of clinical palpation method for determining metastatic cervical lymph nodes were 93.33%, 64.29%, 73.68%, 90% and 79.3% respectively. Careful and repeated clinical palpation plays important role in evaluation of cervical lymph nodes though several modern techniques may help additionally in the management of oral cancer.DOI: http://dx.doi.org/10.3329/updcj.v1i2.13978 Update Dent. Squamous cell carcinoma characterized by poor prognosis due to aggressive tumor growth and dissemination high rate of tumor cell . age ranged of patient case included in the study 40-62 years and mean age 55±99. The sex distribution male/female ratio 1:1. Male case 15 and female 15 of the present study The results of clinical forums showed in the current study was endophytic 10(33.3%) in the same time Exophytic were presented in 20 cases (76.7%). Regarding distribution of the tumors site, the preponderance of them 19 cases 73.3% were located alveolar mucosa, followed by in the tongue 11 cases(36.7%) Tumor stage was analyzed and recorded in Oral squamous cell carcinoma included cases, the preponderance of them were Stage II 11 cases 36.7% followed by stage III 10 cases 33.3% , 9 cases 30.0% were stage I. While Concerning tumor grade, majority of them 15 cases 50% had grade II moderately differentiated SCC, while 11 cases 36.7% had grade III poorly differentiated SCC and 4 cases 13.3% had grade I well differentiated SCC Positive TGF-β3 immunostaining was detected as cell with staining brown color, all tissues sections included show Positive expression based on IHC teqnique. Positive Transforming Growth Factor TGF-β3 Immuno staining was found in all case results and display that 4 samples with percentage 13.3% expressed strong positive 87.67 ± 1.45 expression , 11cases 36.7% showed 51.33 ±0.88 positive expression moderate at the same time 15 samples 50.0% showed positive weak expression.
