Abstract Five various chemometric methods were established for the simultaneous determination of azilsartan medoxomil (AZM) and chlorthalidone in the presence of azilsartan which is the core impurity of AZM. The full spectrum-based chemometric techniques, namely partial least squares (PLS), principal component regression, and artificial neural networks (ANN), were among the applied methods. Besides, the ANN and PLS were the other two methods that were extended by genetic algorithm procedure (GA-PLS and GA-ANN) as a wavelength selection procedure. The models were developed by applying a multilevel multifactor experimental design. The predictive power of the suggested models was evaluated through a validation set containing nine mixtures with different ratios of the three analytes. For the analysis of Edarbyclor® tablets, all the proposed procedures were applied and the best results were achieved in the case of ANN, GA-ANN, and GA-PLS methods. The findings of the three methods were revealed as the quantitative tool for the analysis of the three components without any intrusion from the co-formulated excipient and without prior separation procedures. Moreover, the GA impact on strengthening the predictive power of ANN- and PLS-based models was also highlighted.

A comparative study for the validation and advancement of two analytical approaches applied for the simultaneous determination of Mometasone Furoate (MF), Miconazole Nitrate (MIC) and Gentamicin (GM) formulated in Momenta® cream. The first approach was TLC-spectrodensitometric method, which was advanced by separating the three components on TLC aluminum plates coated with silica gel 60 F254 using chloroform: methanol: formic acid (4:0.3:0.15, v/v/v) as a mobile phase, then scanned at 254nm using Camage TLC scanner 3 operated in reflectance-absorbance mode. The second approach was the chemometric method using two models: Partial Least Squares (PLS) and Principle Component Regression Model (PCR). The proposed approaches were validated according to ICH guidelines and were applied for the determination of the ternary mixtures in their analytical mixtures and pharmaceutical preparation.

An asymmetrical FIA merging-zones manifold based on the dual injection of two sample microvolumes was developed for the simultaneous determination of salicylic acid and acetylsalicylic acid in pharmaceutical preparations at a sampling frequency of 30/h. The complex formed between the Fe(III) reagent continuously introduced in the system and salicylic acid was monitored photometrically at 520 nm. One of the sample plugs was prehydrolysed on injection into an NaOH stream and was circulated through a longer channel than the other plug. This yielded two FIA peaks corresponding to salicylic acid and the overall content, respectively. The proposed manifold was successfully used to control the dissolution test of a pharmaceutical preparation.
