The effect of bisphenol A and chlorinated derivatives of bisphenol A on the level of serum vitellogenin in Japanese medaka (Oryzias latipes)

2,2-bis (4-hydroxyphenyl) propane or Bisphenol A (BPA), has been reported to behave as an endocrine disrupter below acute toxic levels, and is widely present in the water environment. Although BPA is easily chlorinated, very little is reported on the effect of chlorinated BPA to the aquatic organisms. In this study, the estrogenic activities of BPA and its chlorinated derivatives were evaluated by the induction of vitellogenin (VTG) in the serum of mature male Japanese medaka. In addition, the effect of sodium hypochlorite on the decomposition of BPA was tested. The relative potencies of estrogenic activities of chlorinated BPA descended in the order 3,3′-diClBPA>BPA≥3-ClBPA>3,3′,5-triClBPA, and no estrogenic activity was observed in 3,3′,5,5′-tetraClBPA. Lowest Observed Effect Concentration (LOEC) and No Observed Effect Concentration (NOEC) for both 3-ClBPA and 3,3′-diClBPA were 500 μg/L and 200 μg/L, respectively. LOEC for 3,3′,5-triClBPA was >500 μg/L. When BPA was reacted with sodium hypochlorite (24 hours; residual chlorine at 1 ppm), however, complete decomposition of BPA and its chlorinated derivatives was observed. The decrease in BPA and its chlorinated derivatives paralleled the decrease in estrogenic potency evaluated by the induction of vitellogenin (VTG) in the serum of mature male Japanese medaka.

Abstract As one of the alternatives of 2,2-bis(4-hydroxyphenyl)propane (bisphenol A, BPA), 2,2-bis(4-hydroxyphenyl)butane (bisphenol B, BPB) has not gained sufficient concerns so far, due to the limited concentration and toxicity data available. In this study, the acute toxicity of BPB to three aquatic organisms, i.e., Tetradesmus obliquus, Daphnia magna and Danio rerio, was investigated, and it showed that Daphnia magna was the most sensitive organism with the half effective concentration (EC50) of 3.93 mg/L. Thereout, the screened Daphnia magna was exposed to BPB for 21 days to explore the chronic toxicity. Results indicated that BPB restricted the body length of parent Daphnia magna and reduced the total number of broods and neonates. The no-observed effect concentration of BPB to Daphnia magna was as low as 0.01 mg/L, which was two orders of magnitude lower than that reported 0.86–5.00 mg/L of BPA. Furthermore, the ecological risk of BPB was quantitatively assessed using the risk quotient (RQ) method. Obviously, although the environmental concentrations and detectable rate of BPB were much lower than that of BPA, its ecological risk was not necessarily lower. Hence, BPB should not be ignored in the future environmental monitoring and management.

The objective of this work has been to characterize the estrogenic activity of bisphenol-A (BPA) and the adverse effects on the endocannabinoid system (ECS) in modulating germ cell progression. Male offspring exposed to BPA during the foetal-perinatal period at doses below the no-observed-adverse-effect-level were used to investigate the exposure effects in adulthood. Results showed that BPA accumulates specifically in epididymal fat rather than in abdominal fat and targets testicular expression of 3β-hydroxysteroid dehydrogenase and cytochrome P450 aromatase, thus promoting sustained increase of estrogens and a decrease of testosterone. The exposure to BPA affects the expression levels of some ECS components, namely type-1 (CB1) and type-2 cannabinoid (CB2) receptor and monoacylglycerol-lipase (MAGL). Furthermore, it affects the temporal progression of germ cells reported to be responsive to ECS and promotes epithelial germ cell exfoliation. In particular, it increases the germ cell content (i.e., spermatogonia while reducing spermatocytes and spermatids), accelerates progression of spermatocytes and spermatids, promotes epithelial detachment of round and condensed spermatids and interferes with expression of cell–cell junction genes (i.e., zonula occcludens protein-1, vimentin and β-catenin). Altogether, our study provides evidence that early exposure to BPA produces in adulthood sustained and site-specific BPA accumulation in epididymal fat, becoming a risk factor for the reproductive endocrine pathways associated to ECS.

Mature male medaka were continuously exposed to 0.005, 0.0–5 or 1.0 ppb of estradiol-17β (E2 or 0.1, 10 or 100 ppb of p-nonylphenol (NP) or bis-phenol-A (BPA). Female-specific proteins (Fsp) were induced in medaka exposed to 0.005 ppb of E2, 0.1 ppb of NP, or 10 ppb of BPA. Concentrations of 0.005 pbb of E2 and 0.1 ppb of NP corresponded to concentrations of these chemicals detected in river water in Japan. The abilities of the 3 chemicals to induce Fsp were E2> NP> BPA. Embryonic medaka were exposed to E2, NP and BPA under conditions of static-renewal for 200–230 days until pre-maturity. Survival ratios of medaka exposed to E2 and NP declined in concentrations more than 25 ppb and 50 ppb, respectively. The groups of medaka exposed to E2 had individuals with testis-ova or abnormal gonad. There was no male in exposure to 1.0 ppb E2. When exposed to 100 ppb of NP or BPA, abnormal gonad was also detected. Abnormal anal fin (female-like) was observed in male exposed to 100 ppb of NP. The LC50 values for each of the 3 chemicals were much higher than the concentrations detected in water in the environment—the 3 chemicals were considered to have no lethal effect on medaka in aquatic environments. However, exposures to E2 or NP at environmental concentrations induced Fsp. BPA also had the ability to affect medaka as an environmental estrogen, although its extrogenic activity was weaker than that of E2 or NP.

<p>A Lake Multi-biotic Integrity Index (LMII) for the China’s second largest interior lake (Dongting Lake) was developed to assess the water quality status using algal and macroinvertebrate metrics. Algae and benthic macroinvertebrate assemblages were sampled at 10 sections across 3 subregions of Dongting Lake. We used a stepwise process to evaluate properties of candidate metrics and selected ten for the LMII: Pampean diatom index, diatom quotient, trophic diatom index, relative abundance diatoms, Margalef index of algae, percent sensitive diatoms, % facultative individuals, % Chironomidae individuals, % predators individuals, and total number of macroinvertebrate taxa. We then tested the accuracy and feasibility of the LMII by comparing the correlation with physical-chemical parameters. Evaluation of the LMII showed that it discriminated well between reference and impaired sections and was strongly related to the major chemical and physical stressors (r = 0.766, P&lt;0.001). The re-scored results from the 10 sections showed that the water quality of western Dongting Lake was good, while that of southern Dongting Lake was relatively good and whereas that of eastern Dongting Lake was poor. The discriminatory biocriteria of the LMII are suitable for the assessment of the water quality of Dongting Lake. Additionally, more metrics belonging to habitat, hydrology, physics and chemistry should be considered into the LMII, so as to establish comprehensive assessment system which can reflect the community structure of aquatic organisms, physical and chemical characteristics of water environment, human activities, and so on.</p>
