Oil palm OG hybrid genetic potentials are high in term of vegetative and generative traits, such as: low height increment, compact palm, high free fatty acid and high carotenes. However, OG hybrid reproduction is hindered due to its low germination rate. So, alternative method for germination is needed, which is in vitro embryo zygotic culture. This study objective was to reveal the best medium composition for OG hybrid embryo culture by modificated several oil palm basic medium from oil palm tissue culture protocol and to understand the role of plant growth regulators, activated charcoal, and sucrose on in vitro growth and development of OG hybrid embryo. The design used was Completely Randomized Design (CRD) with 16 replication and 4 embryos for each replicates, while the treatments were: A1 = M129 (control); A2 = M129 + activated charcoal; A3 = M129+ activated charcoal + sucrose; A4 = ½ M129 + activated charcoal + sucrose; A5 = M129 + GA3 ; and A6 = M129 + GA3 + NAA. From the study we found that the best medium for OG hybrid open pollinated clone's embryo culture was M129 with GA3 and NAA (A6). This medium gave percentage of in vitro germination up to 14.06%, and later on the plantlets from this medium gave the best vegetative growth performance such as high number of leaves and primer roots.

The oil palm (Elaeis guineennsis Jacq.) is the plant with major oil yield in tropical areas over the world. Pará state is the major producer of Brazil. However, the production of resistant hybrids (Elaeis oleifera x Elaeis guinennsis) to several diseases is necessary, in spite of less productive, is a viable alternative for areas where occur more aggressive diseases. The aim of this research was to rescue in vitro embryos of four varieties of interspecific hybrids of oil palm (Elaeis oleifera x Elaeis guineensis) in different media. Four varieties were tested: (1) Cj-2141; (2) CI-2061; (3) Cj-502 and (4) Cj-494. Four media were tested: (T1) half strength MS medium (½ MS), (T2) MS, (T3) MS + 0.5mg L-1 1-naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP) and (T4) ½ MS+ 0.5mg.L-1 NAA and BAP. All media were supplemented with 0.17g.L-1 NaH2PO4, activated charcoal (0.25%), sucrose (3%) and solidified with phytagel (0.2%). The most germination rate of interspecific hybrids embryos was obtained on ½ MS medium, supplemented with 0.17g.L-1 NaH2PO4, activated charcoal (0.25%), sucrose (3%) and solidified with phytagel (0.2%). The variety CI-2061 presents the best capacity to develop normal seedlings.

Jurnal Lahan Suboptimal : Journal of Suboptimal Lands 9(2): 149-159.Embryo culture is a widely used and effective way of overcoming the dormancy nature of seeds in oil palm.  At the germination stage, the influence of gowth regulators play an important role although in relatively lower concentrations in the germination media. This aim research determined the effect of adding various concentrations of gowth regulator on the three types of oil palm fruit based on the thickness of the shell. This research carried out at the Tissue Culture Laboratory of PT. Sampoerna Ago Tbk. from September 2018 to January 2019. The gowth regulator substances used in this study were NAA, BAP and GA3. The composition of the gowth regulators used were H1 = 0.05 mg/L NAA + 0.1 mg/L BAP + 0.1 mg/L GA3, H2 = 0.05 mg/L NAA + 0.3 mg/L BAP + 0.1 mg/L GA3, H3 = 0.05 mg/L NAA + 0.5 mg/L BAP + 0.1 mg/L GA3 and H4 = No gowth regulator. While the types of oil palm fruit used were C1 = Dura, C2 = Tenera and C3 = Pisifera. This research used a factorial complete randomized design with three replications. The results showed that in general the use of gowth regulator substances H1 = 0.05 mg/L NAA + 0.1 mg/L BAP + 0.1 mg/L GA3 and the type of dura fruit showed positive results based on viability, scoring, survival and height of plantlets when compared with other treatments.

Oil palm (Elaeis guineensis Jacq.) has become the largest traded vegetable oil in the world due to its innumerable uses and its huge economic importance. Which increases the requirement of quality seedling but the conventional seed production limits the commercial seed production with its long germination period and low germination rate. In this context in vitro germination of zygotic embryo could be a promising alternative. Hence in this study the effect of culturing media, plant growth regulators and genotypes were assessed on in vitro regeneration and plantlet development of oil palm zygotic embryos. Zygotic embryos of four elite genotypes were cultured on MS, N6 and Y3 media, with or without plant growth regulators (NAA, BA and GA3-0.2mg/lt). There was no effect of culture media, plant growth regulators and genotypes on germination of zygotic embryos. Whereas on the growth and development of in vitro plantlets, there was a significant effect of culture media and genotypes. Where culture media N6 performs well and Y3 has shown more abnormality. In case of genotypes, genotypes G1 and G2 shown better performance when compared to genotypes G3 and G4. The effect of plant growth regulators on germination and plant growth was non significant.

Aim: This study evaluated efficient culture media for the regeneration of elite material through somatic embryogenesis from oil palm zygotic embryos. Methodology: For callus induction, zygotic embryos of four elite genotypes (G1-264T, G2-238DX17P, G3-37DX17P and G4-237T) were cultured on three basal media (Y3, MS and N6) with different auxin 2 mg l-1 (Picloram, 2,4-D and Dicamba) combinations. Subculture was made every month for three passages. It evaluated various callus characters. The embryogenic calli from callus induction media were transferred to the embryo maturation medium and subcultured until the polyembryoids formed. For shoot and root formation, somatic embryo clumps were transferred into regeneration media. In-vitro plantlets with well-grown roots were hardened in pots for six weeks and assessed clonal fidelity using polymorphic SSR primers. Results: Among the treatments, calli from N6+2,4-D, Y3+2,4-D and N6+Picloram showed the highest embryogenic callus potential. G4-237T induced more embryogenic calli (32.982) among genotypes, which was on par with G1-264T (24.196). Embryogenic calli grown on N6 media with Dicamba showed the highest proliferation rate (1.141). After 60 days of culture on regeneration media, the highest number of plantlets per somatic embryogenic clump was obtained from G1-264T on N6 media supplemented with Dicamba. Interpretation: Culture media salt concentration showed a significant difference among media by causing perturbations of auxin flow during somatic embryogenesis affecting callus induction, proliferation and plantlet regeneration. This may be useful for standardizing the genotype-specific regeneration media in oil palm.

<p>ABSTRACT</p><p>Molecular genetic analysis of hard bunch phenomenon in oil palm was done in order to elucidate the role of genetic factor underlying hard bunch in oil palm plantation. The aim of this study was to identify the AFLP primer combination that co-segregates with hard bunch phenotype related gene in oil palm. Molecular analysis was done by bulk segregant analysis approach. DNA was isolated from leaves of the normal and hard bunch palm. DNA from ten individual palms from each category were pooled and used as a template. A total of 56 AFLP primer combinations were selected for selection of polymorphic primer, and as a result it was found that 22 AFLP primer combinations (39.28%) were polymorphic. A total of 48 individual of palm DNA containing 24 individual for each group were further genotyped by those 22 polymorphic markers. Of these, one AFLP primer combination (E-ACC/M-CTG) was obtained as a co-segregated marker that distinguished the hard bunch DNA from the normal one. Based on the analysis of the target sequence aligned to the oil palm DNA sequences available in database, we found that our sequence has similarity with Ty-1 copia retrotransposon.
