Tubers from 7 potato varieties were analyzed for their rates of glycoalkaloid accumulation in response to stresses of three types of mechanical injury and low temperature storage. Mechanical injuries were found to greatly stimulate glycoalkaloid accumulation in both peel and flesh of tubers. The extent of glycoalkaloid accumulation appears to depend on variety, type of mechanical injury, and storage period. Most of the injury-stimulated glycoalkaloid accumulation occurred within 7 and 14 days after treatment. Cutting the tubers resulted in the highest content of glycoalkaloids both in flesh and peel up to levels that exceeded the upper safety limit of 200 mg/kg FW. Injury stimulated α-solanine accumulation in stored potato tubers is more than α-chaconine, resulting in a decrease in the α-chaconine: α-solanine ratio. When tubers were stored at low temperature, the rate of glycoalkaloid accumulation was found to be independent of the glycoalkaloid level at harvest. The greatest increase in total glycoalkaloid content of the seven varieties was found after two weeks of storage at both 4 ºC and 10 ºC. Further storage at these temperatures resulted in a decrease in the rate of glycoalkaloid accumulation in most of them. At 10 ºC glycoalkaloid content tended to increase more rapidly than at 4 ºC. The α-solanine content of the tubers showed an increase following low temperature storage.

This study developed a method for furosemide quantification through high performance liquid chromatographic technique. Special attention was given to solute loss and storage stability due to furosemide’s low solubility and photosensitivity, respectively. The performance of Nylon and PVDF filters was tested in a 2 mg.L-1 furosemide solution. PVDF filters showed better recovery capacity and therefore are more suitable for furosemide filtration.  Over eight days, three different storage conditions were studied to access furosemide degradation susceptibility: (i) exposure to light at room temperature, (ii) storage at room temperature without exposure to light, and (iii) storage at 4ºC without exposure to light. The study demonstrated that after 48 h under natural light exposure furosemide was completely degraded. Furosemide solution stored in the dark was stable. Storage temperature did not seem to affect furosemide concentration. The study shows that the selection of more suitable filter and storage conditions for furosemide determination is crucial to avoid underestimation errors.

The influence of light exposure, bottle color and storage temperature on the quality parameters of Malvasia delle Lipari (MdL) sweet wine were investigated. Wine samples bottled in clear-colored (colorless, green and amber) glass were stored under different artificial lighting conditions, in order to simulate the retail environment (one cool-white, fluorescent lamp) and to perform an accelerated test (four and six cool-white, fluorescent lamps). The storage temperature was kept constant (25 °C) for the first 90 days of the experiment and then samples were monitored for up to 180 days at higher temperatures (30, 35 and 40 °C). The principal enological parameters, total phenols, color, 5-hydroxymethylfurfural (HMF) and 2-furaldehyde (2F) contents were studied. The shelf-life test pointed out minimum variations of the basic chemical parameters, while the quality attributes most affected by lighting were color, together with HMF and 2F levels which, hence, can be considered as indicators of the severity of storage conditions.

Wounding stress is an effective strategy to increase the content of bioactive compounds in horticultural crops. Potato tubers subjected to wounding stress accumulate chlorogenic acid (CGA) and CGA isomers (neo-CGA and crypto-CGA), which are phenolics that prevent and treat different chronic and degenerative diseases. In this study, the effects of wounding stress and storage temperature (10 °C and 20 °C for 168 h) on the accumulation of CGA isomers in potatoes were evaluated. Results indicated that CGA accumulation was favored when wounded potatoes were stored at 20 °C for 120 h, obtaining a 1923.1% higher concentration when compared with samples before storage. Furthermore, wounded potatoes stored at 10 °C for 120 h showed the highest neo-CGA increase in concentration (712.2%). Likewise, the highest crypto-CGA concentration (84.9% higher than control samples) was quantified in wounded potatoes stored at 20 °C for 144 h. Based on the results from both the present study and previous reports, a strategy that summarizes effective postharvest stress conditions that induce the accumulation of specific CGA isomers in potatoes is presented. The tissue with an increased content of bioactive compounds could be used as raw material to produce functional foods or could be subjected to downstream processing to produce dietary supplements.
