Effect of Rhizoctonia spp. on Polyphenol Oxidase Enzyme Activity in Alfalfa Seedling

Soil borne diseases cause significant losses on quantity and quality of many crop species annually. Rhizoctonia is a widespread and ecologically diverse soilborne fungus causing different types of diseases in many plant species including alfalfa (Medicago sativa). Rhizoctonia species have been traditionally identified based on the cell nuclear condition. Polyphenol oxidases (PPOs) are ubiquitous copper-containing enzymes that are widely occurring enzymes among plants. PPOs are involved in the oxidation of polyphenols into quinones (antimicrobial compounds) and lignification of plant cells that contribute to the formation of defense barriers against pathogens. The study was conducted with the aim to determine the effect of indigenous isolates of a multinucleate (Rhizoctonia solani AG-4) and nineteen isolates of binucleate Rhizoctonia on PPO activity in alfalfa (cv. Gea) seedling under in vitro conditions. The activity of PPO enzyme was determined in inoculated and uninoculated control alfalfa plants after ten days from inoculation. There was a significant increase in the activity of PPO after treatment of alfalfa seedling with isolates of Rhizoctonia. Among Rhizoctonia isolates, highest induction of PPO activity was recorded with pathogenic R. solani AG-4. In present study, increased amounts of PPO were also observed in plants that were challenged with Rhizoctonia spp. PPO has a role in catalyzing phenolic oxidation in limiting disease development. PPO may therefore be involved in induction of defense resistance against plant diseases.

The role of autoantibodies in in vitro fertilization (IVF) has been discussed for almost three decades. Nonetheless, studies are still scarce and widely controversial. The aim of this study is to provide a comprehensive systematic review on the possible complications associated to autoantibodies (AA) impeding the chances of a successful IVF cycle. An Embase, PubMed/Medline and Cochrane Central Database search was performed on 1 December 2018, from 2006 until that date. From the 598 articles yielded in the search only 44 relevant articles ultimately fulfilled the inclusion criteria and were qualitatively analyzed. Five subsets of results were identified, namely, thyroid related AA, anti-phospholipid antibodies, anti-nuclear antibodies, AA affecting the reproductive system and AA related to celiac disease. It may be implied that the majority of auto-antibodies exert a statistically significant effect on miscarriage rates, whereas the effects on clinical pregnancy and live birth rates differ according to the type of auto-antibodies. While significant research is performed in the field, the quality of evidence provided is still low. The conduction of well-designed prospective cohort studies is an absolute necessity in order to define the impact of the different types of autoantibodies on IVF outcome.

Because of the great amount of hard dental tissues loss, most of non-vital teeth must be prosthetically restored using a post. The anchorage of these posts is based on enlarging the root canal, so the question is what type of endodontic post is more appropriate to use in order to ensure a favorable prognosis of these teeth, avoiding as long as possible their extraction. Loss of teeth is affecting not only the oral cavity and the dento-maxillary system, but also the quality of life of the patients, with both psychological and general health consequences. Even treated, the edentulous state determines major changes in diet and can be a cause of gastrointestinal disorders. Thisin vitroexperiment tested the resistance to fracture of non vital teeth restored with different types of posts, in order to find out which type is best to use for their restoration. From the of this in vitro experiment it has been observed that in the case of teeth restored with metal posts, regardless of the diameter of the post, the failure of the sample was represented by root fracture. In the case of the teeth restored with fiberglass posts, the failure was caused by the fracture of the post or the fracture of the coronary part of the tooth.

Abstract Background Glistenings describe small, refractile microvacuoles that may arise within the intraocular lens (IOL) material and reduce the patients’ quality of vision. Lenses composed of hydrophobic acrylic material are particularly affected by glistening formation. In this study, we compared the tendency of glistening formation in six different types of hydrophobic acrylic intraocular lenses (IOLs). Methods We used a well-established accelerated laboratory method to develop glistenings in the following IOLs: Vivinex XY1 (Hoya), AcrySof SN60WF (Alcon), Tecnis ZCB00 (AMO), Avansee PN6A (Kowa), Aktis SP NS-60YG (Nidek), and CT Lucia 601P (Zeiss). IOLs were first immersed in saline at 45 °C for 24 h and then at 37 °C for 2.5 h in a water bath. Microvacuole (MV) density and size (Miyata grading) were documented and calculated using an image analysis program. Results The mean glistening density [MV/mm2] and mean Miyata grading (in brackets) were: Vivinex: 11.6 ± 5.7 (0), SN60WF: 264.4 ± 110.3 (2.6), Tecnis: 6.0 ± 2.8 (0), Avansee: 2.2 ± 0.7 (0), Aktis: 851.4 ± 59.4 (3+) and CT Lucia: 71.0 ± 71.6 (1). Conclusions While all tested IOLs showed glistenings with the accelerated laboratory method, the Aktis and SN60WF showed the highest microvacuole density, followed by the CT Lucia. In comparison, the Vivinex, Tecnis, and Avansee IOLs showed far fewer number of glistenings.

Peripheral nerve injuries (PNI) can have several etiologies, such as trauma and iatrogenic interventions, that can lead to the loss of structure and/or function impairment. These changes can cause partial or complete loss of motor and sensory functions, physical disability, and neuropathic pain, which in turn can affect the quality of life. This review aims to revisit the concepts associated with the PNI and the anatomy of the peripheral nerve is detailed to explain the different types of injury. Then, some of the available therapeutic strategies are explained, including surgical methods, pharmacological therapies, and the use of cell-based therapies alone or in combination with biomaterials in the form of tube guides. Nevertheless, even with the various available treatments, it is difficult to achieve a perfect outcome with complete functional recovery. This review aims to enhance the importance of new therapies, especially in severe lesions, to overcome limitations and achieve better outcomes. The urge for new approaches and the understanding of the different methods to evaluate nerve regeneration is fundamental from a One Health perspective. In vitro models followed by in vivo models are very important to be able to translate the achievements to human medicine.

This study was aimed to measure the decolorization of tattoo dyes by different chemicals and polyphenol oxidases from several plant sources. The tattoo inks removal market has burgeoned over the years, due to increased spread of tattooed persons about the world. Laser and surgery are presently the gold standards for removing of the tattoo. However, both of them have blemishes. Consequently, lots of persons were preferring easier, faster and cheaper procedures for tattoo remove. In this study polyphenol oxidases enzyme from many plant sources and different chemicals were used for decolorization of tattoo dyes in vitro. The polyphenol oxidase enzyme was used for removing of tattoo dyes (brown and blue) in order to demonstrate their potential in the treatment and decolorization of the tattoo, which is hazardous when removing by laser. The results show that 89 and 82 % of the brown and blue tattoo dyes respectively, were removed after 24 hours by enzyme extracted from Malva parviflora leaves, whereas the decolorization efficiency of polyphenol oxidase from other plant sources given less than 16% of the same dyes. The results for tattoo dyes decolorization by different chemicals revealed that Bimethylbenzylamine was the best chemical used with decolorization ratio 36 and 38 % for brown and blue tattoo dye, respectively.

Previous freeze fracture results on the intact giant, amoeba Chaos carolinensis indicated the presence of a fibrillar arrangement of filaments within the cytoplasm. A complete interpretation of the three dimensional ultrastructure of these structures, and their possible role in amoeboid movement was not possible, since comparable results could not be obtained with conventional fixation of intact amoebae. Progress in interpreting the freeze fracture images of amoebae required a more thorough understanding of the different types of filaments present in amoebae, and of the ways in which they could be organized while remaining functional.The recent development of a calcium sensitive, demembranated, amoeboid model of Chaos carolinensis has made it possible to achieve a better understanding of such functional arrangements of amoeboid filaments. In these models the motility of demembranated cytoplasm can be controlled in vitro, and the chemical conditions necessary for contractility, and cytoplasmic streaming can be investigated. It is clear from these studies that “fibrils” exist in amoeboid models, and that they are capable of contracting along their length under conditions similar to those which cause contraction in vertebrate muscles.

The utilization of the ChemScan® RDI was tested for different types of water concentrates. Concentrates were prepared by cartridge filtration or flocculation, and analysed either without purification, or after Immunomagnetic separation (IMS) or flotation on percoll-sucrose gradients. Theenumeration of the oocysts was subsequently performed using the ChemScan® RDI Cryptosporidium application. Enumeration by direct microscopic observation of the entire surface of the membrane was carried out as a control, and recoveries were calculated as a ratio between the ChemScan® RDI result and the result obtained with direct microscopic enumeration. The Chemscan enumeration technique proved reliable, with recoveries yielding close to 100% in most cases (average 125%, range from 86 to 467%) for all the concentration/purification techniques tested. The quality of the antibodies was shown to be critical, with antibodies from some suppliers yielding recoveries a low as 10% in some cases. This difficulty could, however, be overcome by the utilization of the antibody provided by Chemunex. These data conclusively prove that laser scanning cytometry, which greatly facilitates the microscopic enumeration of Cryptosporidium oocysts from water samples and decreases the time of observation by four to six times, can be successfully applied to water concentrates prepared from a variety of concentration/purification techniques.
