The presented work contains an analysis of seroprevalence, tension, and duration of post-vaccination immunity to the SARS-CoV-2 coronavirus in the residents of the Republic of Belarus after immunization with two vaccine preparations available in the country: Sputnik V and Vero Cell.It was found that seroconversion after the injection of the Vero Cell vaccine occurred significantly later than after the Sputnik V vaccine (p < 0.001). Nevertheless, two-stage immunization with the vaccines provided a sufficiently high efficiency of the inoculated antibodies to the S protein of the COVID-19 pathogen: the proportion of seropositive persons reached 99.19 [96.89; 99.97] % for Sputnik V and 96.03 [90.81; 98.53] % for Vero Cell. The efficiency of antibody formation after immunization with the Vero Cell vaccine was lower in older persons (in the group > 65 years). The proportion of individuals with the highest IgG score for the SARS-CoV-2 S protein was higher after the Sputnik V administration compared to that in response to the Vero Cell vaccine (p < 0.05), and gradually decreased over time. However, no significant decrease in the level of seropositive individuals after 90 days from the start of immunization with the both vaccine preparations was detected. In COVID-19 survivors immunized with the Sputnik V vaccine, the quantitative indicators of post-vaccination antibodies reached their peak values after 1 dose of the vaccine. The obtained results complement the accumulated world science and practical information on the problem of the postvaccination immunity formation in the context of the use of different drugs against COVID-19.>< 0.001). AbstractCoxiella burnetii is the bacterial causative agent of the zoonosis Q fever. The current human Q fever vaccine, Q-VAX®, is a fixed, whole cell vaccine (WCV) licensed solely for use in Australia. C. burnetii WCV administration is associated with a dermal hypersensitivity reaction in people with pre-existing immunity to C. burnetii, limiting wider use. Consequently, a less reactogenic vaccine is needed. Here, we investigated contributions of the C. burnetii Dot/Icm type IVB secretion system (T4BSS) and lipopolysaccharide (LPS) in protection and reactogenicity of fixed WCVs. A 32.5 kb region containing 23 dot/icm genes was deleted in the virulent Nine Mile phase I (NMI) strain and the resulting mutant was evaluated in guinea pig models of C. burnetii infection, vaccination-challenge, and post-vaccination hypersensitivity. The NMI ∆dot/icm strain was avirulent, protective as a WCV against a robust C. burnetii challenge, and displayed potentially altered reactogenicity compared to NMI. Nine Mile phase II (NMII) strains of C. burnetii that produce rough LPS, were similarly tested. NMI was significantly more protective than NMII as a WCV; however, both vaccines exhibited similar reactogenicity. Collectively, our results indicate that, like phase I LPS, the T4BSS is required for full virulence by C. burnetii. Conversely, unlike phase I LPS, the T4BSS is not required for vaccine-induced protection. LPS length does not appear to contribute to reactogenicity while the T4BSS may contribute to this response. NMI ∆dot/icm represents an avirulent phase I strain with full vaccine efficacy, illustrating the potential of genetically modified C. burnetii as improved WCVs.

ABSTRACTPorcine epidemic diarrhea virus (PEDV) causes high mortality in neonatal piglets. The PEDV spike (S) protein contains two intracellular sorting motifs, YxxΦ (tyrosine-based motif YEVF or YEAF) and KVHVQ at the cytoplasmic tail, yet their functions have not been fully elucidated. Some Vero cell-adapted and/or attenuated PEDV variants contain ablations in these two motifs. We hypothesized that these motifs contribute to viral pathogenicity. By transiently expressing PEDV S proteins with mutations in the motifs, we confirmed that the motif KVHVQ is involved in retention of the S proteins in the endoplasmic reticulum (ER)-Golgi intermediate compartment (ERGIC). In addition, we showed that the YxxΦ motif triggers endocytosis of S proteins. These two motifs synergistically regulate the level of S expressed on the cell surface. To investigate their role in viral pathogenicity, we generated three recombinant PEDVs by introducing deletions or a mutation in the two motifs of the infectious clone of PEDV PC22A strain (icPC22A): (i) icΔ10aa (ΔYxxΦEKVHVQ), (ii) icΔ5aa (ΔKVHVQ), and (iii) icYA (Y1378A, to an inactivated motif, AEVF). Infection of Vero cells with icΔ10aa resulted in larger syncytia and more virions, with reduced numbers of S protein projections on the surface compared with icPC22A. Furthermore, we orally inoculated five groups of 5-day-old gnotobiotic piglets with the three mutants, icPC22A, or a mock treatment. Mutant icΔ10aa caused less severe diarrhea rate and significantly milder intestinal lesions than icPC22A, icΔ5aa, and icYA. These data suggest that the deletion of both motifs can reduce the virulence of PEDV in piglets.IMPORTANCEMany coronaviruses (CoVs) possess conserved motifs YxxΦ and/or KxHxx/KKxx in the cytoplasmic tail of the S protein. The KxHxx/KKxx motif has been identified as the ER retrieval signal, but the function of the YxxΦ motif in the intracellular sorting of CoV S proteins remains controversial. In this study, we showed that the YxxΦ of PEDV S protein is an endocytosis signal. Furthermore, using reverse genetics technology, we evaluated its role in PEDV pathogenicity in neonatal piglets. Our results explain one attenuation mechanism of Vero cell-adapted PEDV variants lacking functional YxxΦ and KVHVQ motifs. Knowledge from this study may aid in the design of efficacious live attenuated vaccines against PEDV, as well as other CoVs bearing the same motif in their S protein.

<p class="NoParagraphStyle" align="center"><strong>ABSTRACT</strong></p><p class="NoParagraphStyle" align="center"><strong> </strong></p><p>The study was aimed to evaluate the efficacy of formalin-killed non-hemolytic <em>Streptococcus agalactiae </em>N14G and NK1 isolates whole-killed vaccine to prevent streptococcosis in tilapia. Ten fishes were reared in a tank 60x30x35 cm3 with an average body weight at 10.79±0.99 g. Fish was vaccinated through bath immersion at a concentration of 109 cfu/mL. Fish was subsequently challenged by intraperitonial injection of <em>Streptococcus agalactiae </em>105 cfu/mL at 11 days post-vaccination. Parameters observed were survival, relative percent survival (RPS), total leukocyte, phagocytic activity, antibody titer, total erythrocyte, haemoglobin level, haematocrit level, dan water quality. Samplings were performed in day-0, 20, and 30 after vaccination. Both vaccines have shown higher survival (60%) and RPS (40%) when challenged with pathogenic <em>Streptococcus </em>N14G isolates than other treatments. Ikan nila yang digunakan memiliki bobot 10,79±0,99 g, dipelihara sebanyak sepuluh ekor dalam akuarium ukuran 60x30x35 cm3. Ikan divaksinasi dengan metode perendaman dengan dosis 109 cfu/mL. Uji tantang dilakukan pada hari ke-11 pascavaksinasi dengan dosis 105 cfu/mL. Parameter yang diamati meliputi sintasan (SR), sintasan relatif/<em>relative percent surviva</em>l (RPS), total leukosit, aktivitas fagositik, titer antibodi, total eritrosit, kadar hemoglobin, kadar hematokrit, dan kualitas air. Pengamatan parameter dilakukan pada hari ke-0, ke-10, ke-20, dan ke-30. Hasil penelitian menunjukkan perlakuan kedua vaksin yang diinfeksi bakteri patogen isolat N14G memberikan nilai sintasan dan nilai RPS tertinggi dibanding perlakuan lainnya. Nilai sintasan dan RPS kedua perlakuan tersebut adalah 60% dan 40%. Nilai RPS yang cukup kecil menunjukkan vaksin yang diberikan masih kurang efektif untuk mencegah infeksi bakteri <em>S. agalactiae</em>.</p><p> </p><p>Kata kunci: ikan nila, perendaman, <em>Streptococcus agalactiae</em>, vaksin sel utuh</p>
