Empirical evidence of complementary medicine from one community in Kalimantan often uses Ulin fruit (Eusideroxylon zwageri) for medical therapy or as herbal drinks by people with diabetes mellitus. This research aims to identify the antidiabetic compound of Ulin fruit extract against Streptozotocin (STZ) induced diabetic rats. Extraction was performed by maceration method using 96% ethanol, phytochemical test of extracts, extract toxicity test on mice (50 mg/BW to 200 mg/BW), test antidiabetic extract in diabetic rats induced by STZ as much as 70 mg/kgBW with each extract dose of 150, 300, and 600 mg given for 14 days, positive control is Glibenclamide 0.25 mg/kg/day, and the negative control is CMC 1%. Phytochemical test results showed that the extract of Ulin contains alkaloids, tannins, flavonoids, phenolics, terpenoids, and steroids. Ulin fruit extract was not toxic to rats at 50, 150, and 200 mg for three days. Based on the Mann-Whitney test, there was an effect of giving the extract on changes in the average body weight and body temperature of rats during three days of administration (p = 0.000). The results showed novelty that the extract of Ulin fruit affected reducing blood glucose levels in diabetic rats during 14 days of administration (p=0.000).Keywords: Antidiabetic, Eusideroxylon zwageri, flavonoids, streprozotocin, diabetic rats

Abstract Synbiotics are a combination of probiotics and prebiotics that have a positive effect on livestock performance. The method of giving synbiotics as a growth supplement needs to be studied to get the best results. This study aimed to investigate the effect of administering synbiotic (a mixture of Lactobacillus casei and porang tuber extracts (LCPE) in water or as feed supplement on performance and nutrient digestibility of Tegal duck. The research used a total number of 240-day old ducks with an average body weight of 47.84 ± 6.32 g. The ducks were weighed and distributed into 24 groups. Two experiments (I and II) were carried out simultaneously in the same cage and time. Each experiment used 120 ducks. Ducks were adapted from the age of 1 to 14 days. Parameter data were measure in ducks aged 15 to 42 days. Trial I included 3 groups of dietary synbiotic (0, 10, 20 ml/kg) through feed, while In trial II, the ducks were fed synbiotics (0; 10; 20 ml/L) through water drinking for four weeks. The observed variables included growth performance and nutrient digestibility. The mixture of Lactobacillus casei and glucomannan from porang tuber extract improved nutrient digestibility and duck performance. The best performance and nutrient digestibility is LCPE 20 ml/L drinking water.

Purpose. To study the possibility of obtaining highly productive intergeneric salmonid hybrids between rainbow trout and brook trout; brown trout and brook trout; rainbow trout and brown trout, as well as to develop methodological approaches and determine of optimal variants of hybrid crosses. Findings. In order to obtain intergeneric hybrids, we used six variants of hybrid crosses with brood fish of three salmonids belonging to three families (Salmo, Oncorhynhus, Salvelinus). The study used age-4 female rainbow trout with average body weight of 3296.8 g, Fork length was 62.6 cm, and the average working fecundity was 7420 eggs. Age-3 rainbow trout males had an average body weight of 1613 g and an average body length of 49.8 cm; age-3 brown trout females had an average body weight of 453.8 g and average working fecundity of 1540 eggs, and males had an average weight of 458.7 g; age-3 brook trout females had an average weight of 809.7 g and a length of 38.9 cm with working fecundity of 1732 eggs, and age-4 males had an average weight of 1212.8 g and an average body length of 46.0 cm. Twelve variants of fertilization were used: six variants at normal water temperature and six variants after a temperature shock. Under natural conditions, the creation of intergeneric hybrids is almost impossible, except for variants between brown trout and brook trout, which is due to the similarity of their biology. However, the efficiency of this cross is low and economically impractical for fish farmers. When applying the temperature shock during fertilization, hybrids proved to be the most effective, where females were rainbow trout, and males were brook trout and brown trout. The average weight of young-of-the-year intergeneric hybrids was, depending on the species of fish, from 8 to 54 g. The highest results were obtained for the creation of hybrids where following broodstock was used: ♂brook trout Х ♀brown trout; ♂brown trout Х ♀rainbow trout. In these variants of crossbreeding, the survival rate of young-of-the-year during the period of cultivation was 94.8 and 92.8%, respectively. In particular, the above hybrids did not suffer from infectious diseases during the growing period. Originality. New data on the development of methods for obtaining viable offspring of newly created hybrids were obtained, and the optimal variants of crossing between females and males of these salmonids were determined. Practical value. The results can be used for artificial breeding of salmonids in specialized farms that will allow obtaining high quality products and reducing their costs. Key words:rainbow trout, brown trout, brook trout, incubation, free embryos, larvae, fry, young-of-the-year.

Human environmental exposures to toxicants, such as polychlorinated biphenyls (PCBs), increase oxidative stress and disease susceptibility. Such exposures during pregnancy and/or nursing have been demonstrated to adversely affect offspring health outcomes. Nuclear factor erythroid-2-related factor 2 (Nrf2) regulates the antioxidant response and is involved in the detoxification of coplanar PCBs, like PCB126. The purpose of this study was to investigate glucose tolerance and body composition in PCB-exposed offspring expressing or lacking Nrf2. We hypothesized that offspring lacking Nrf2 expression would be more susceptible to the long-term health detriments associated with perinatal PCB exposure. During gestation, whole-body Nrf2 heterozygous (Het) and whole-body Nrf2 knockout (KO) mice were exposed to vehicle or PCB126. Shortly after birth, litters were cross-fostered to unexposed dams to prevent PCB exposure during nursing. Offspring were weaned, and their body weight, body composition, and glucose tolerance were recorded. At two months of age, PCB exposure resulted in a significant reduction in the average body weight of offspring born to Nrf2 Het dams (p < 0.001) that primarily arose from the decrease in average lean body mass in offspring (p < 0.001). There were no differences in average body weight of PCB-exposed offspring born to Nrf2 KO dams (p > 0.05), and this was because offspring of Nrf2 KO dams exposed to PCB126 during pregnancy experienced a significant elevation in fat mass (p = 0.002) that offset the significant reduction in average lean mass (p < 0.001). Regardless, the lack of Nrf2 expression in the offspring themselves did not enhance the differences observed. After an oral glucose challenge, PCB-exposed offspring exhibited significant impairments in glucose disposal and uptake (p < 0.05). Offspring born to Nrf2 Het dams exhibited these impairments at 30 min and 120 min, while offspring born to Nrf2 KO dams exhibited these impairments at zero, 15, 30, 60 and 120 min after the glucose challenge. Again, the interactions between offspring genotype and PCB exposure were not significant. These findings were largely consistent as the offspring reached four months of age and demonstrate that the lack of offspring Nrf2 expression does not worsen the metabolic derangements caused by in utero PCB exposure as we expected. Future directions will focus on understanding how the observed maternal Nrf2 genotypic differences can influence offspring metabolic responses to in utero PCB exposure.

Abstract Single and simultaneous toxic effects of glyphosate (Amega Up, 360 g L−1, 4%) and copper sulphate (0.01%) were studied in avian embryos treated either with injection directly into the air chamber or by immersion application for 30 min on day 0 of incubation. Alterations of the chicken embryos were evaluated during necropsy performed on day 19 of incubation, together with mortality, body weight and the type of developmental abnormalities. Based on the results, the injection application appeared to be more toxic than the immersion method, as it induced increased mortality and reduced the average body weight, and resulted in a higher incidence of congenital anomalies. Supposedly, a toxicodynamic interaction occurs between copper sulphate and glyphosate, which may reduce the vitality of embryos and thus decrease the number of offspring in wild birds.

The present study was conducted to investigate the effects of diet with betaine supplementation on the growth performance, carcass quality and fat deposition in finishing Ningxiang pigs. A total of 24 Ningxiang pigs (43.6 ± 5.34 kg of average body weight) was randomly divided into two groups, with 6 replicates per treatment and 2 pigs per replicate. The treatments included a control group (basal diet) and a test group (basal diet + 0.2% betaine). The whole trial lasted 81 days. At the end of the experiment, one pig (close to the average body weight of all experimental pigs) per replicate was slaughtered to determine the carcass traits, meat quality and the mRNA expression levels of genes relate to fat deposition (one pig per replicate was randomly selected and fasted for 12 h, n = 6). Results indicated that growth performance was not changed with betaine supplementation. However, dietary with betaine supplementation decreased back fat thickness and fat percentage, and increased the lean meat percentage as well (p < 0.05). In addition, diet with betaine supplementation reduced drip loss, water loss, cooking loss, shear force and b×24 h value of meat (p < 0.05). There was no difference in total moisture, ether extract and crude protein of longissimus thoracis between the control and test group. Dietary with betaine supplementation decreased ether extract and total cholesterol (p < 0.05) in liver. Dietary with betaine supplementation upregulated the mRNA expression levels of adipose triglyceride lipase (ATGL) and sirtuin 1 (Sirt1), while downregulated the mRNA expression levels of fatty acid synthase (FAS) and acetyl CoA carboxylase (ACC) in subcutaneous fat of back (p < 0.05). Besides, dietary with betaine supplementation upregulated the fatty acid binding protein 4 (FABP4) mRNA expression of longissimus thoracis in finishing Ningxiang pigs (p < 0.05). These results showed that diet supplemented with betaine could improve the slaughtering performance and meat quality, and regulate the genes expression to affect the fat deposition in finishing Ningxiang pigs.

The main cause of obesity is an imbalance in consumption and caloric intake. Obesity is described as an excessive and abnormal accumulation of more than 20 percent of body fat in an individual's average body weight. The average body weight refers to an “individual's maximum healthy weight,” which is determined primarily by their height, age, and muscular growth. Obesity, however, is diagnosed by calculating the BMI which is a measure of individual’s body weight and height.  Additionally, according to the National Institutes of Health, a person with a maximum limit of BMI of 30 kg/m2 is declared obese. Moreover, despite World Health Organization concerns, the prevalence of obesity is increasing among children and adults worldwide. Excessive overweight continues to the most underestimated public health problems on a global scale, including the fact that obesity is linked with an increased risk of sickness, disability, and death.  Cardiovascular disorders, especially diabetes and hypertension have become a leading cause of death globally and these are the primary illnesses linked with obesity. However, the mechanisms behind obesity-related hypertension and other metabolic disorders remain little understood. In this present review, we discussed the correlation between cardiovascular disease and obesity, focusing on the biological processes underlying the relationship between hypertension and obesity.

AbstractA total of 360-day-old broiler chicks were allocated into six groups in 2 (Coccidial challenge or not) × 3 (dietary treatments) factorial design. Three dietary treatments including: basic diet, basic diet plus organic acids (OAs) in drinking water, and basic diet plus OAs in the feed with and without coccidial challenge. The OAs in water or feed improved (P < 0.01) average body weight (ABW), average body weight gain (ABWG), and feed conversion ratio (FCR) as compared with the control diet during starter, grower, and whole experimental period. Coccidial challenge decreased BW, ABWG, and average feed intake (AFI), as well as resulted in poor FCR during the starter and whole experimental period (P < 0.05). Though there was no interaction between OAs supplementation and coccidial challenge, the OAs supplementation improved the overall performance with and without coccidial challenge birds on 21 d and 35 d. IgG was found higher (P = 0.03) in broilers fed OAs in feed without the coccidial challenge group. On 18 d, OAs supplementation in feed increased TNF-γ (P = 0.006), whereas the coccidial challenge decreases TNF-γ (P = 0.01) and IL-10 (P =  < .0001), and increases IgM (P = 0.03), IgG (P = 0.04) and IgA (P = 0.02). On 29 d, the coccidial challenge increases IgM and IgA. On 18 d, jejunal lesion score was found significantly higher in the coccidial challenged group as compared to OAs supplementation with coccidial challenged groups on 18 d (P < 0.0001) and 29 d (P = 0.03). Crypt depth was higher, and Villus-height to Crypt depth ratio was lower in the coccidial challenge group on 18 and 29 d. The Goblet cells were found higher in the non-coccidial challenge on 18 d. After 18 d, 16S rDNA gene sequence analysis of ileal chyme has shown that coccidial challenge decreases Lactobacillus_reuteri species as compared to the non-challenged group (P = 0.02). After 29, Cyanobacteria abundance reduced (P = 0.014) in the challenged group than the non-challenged group at the phylum level. At the genus level, Lactobacillus (P = 0.036) and unidentified Cyanobacteria (P = 0.01) were found higher in the non-challenged group than the coccidial challenge group. The results indicate that the OAs supplementation showed improved responses in a pattern similar to the non-challenged control group by neutralizing the negative effects of the coccidial challenge.
