Three different kinetic models – Monod’s model, Monod’s model with substrate inhibition, and Monod's model with substrate and product inhibition were developed for studying of beer fermentation with free and immobilized cells at different main fermentation and maturation temperatures. The most accurate model was Monod's model with substrate and product inhibition. It showed that maturation temperature had no effect on primary metabolism but it affected significantly the secondary metabolites production. In regard to carbonyl compounds and esters, the increase in maturation temperature led to different trends for free and immobilized cells. Regarding the higher alcohols, the increase in maturation temperature resulted in increase in their yield coefficients for both immobilized and free cells. A sensory evaluation of beers produced with free and immobilized cells were also carried out and the results showed similar results for two beer types.

A novel strain, Bacillus fusiformis CGMCC1347, was utilized successfully to transform isoeugenol to vanillin and the product inhibition could be well avoided by using immobilized cells in the isoeugenol/aqueous biphasic system. The Bacillus fusiformis CGMCC1347 cells were entrapped into sodium alginate under conditions of 2.5% sodium alginate, 10% cells and 0.1 mol l-1 CaCl2. The optimum pH for free cells and immobilized cells were 4.0 and 3.5, respectively, and the optimum reaction temperature for both free and immobilized cells was 37 °C. The pH stabilities of free cells and immobilized cells were good at all investigated pH levels. For thermal stability, free cells were stable at 50°C60°C and the immobilized cells were stable at 50°C 80°C. The half-life of the immobilized cells was more than 25 d, comparing with less than 14 d for the free cells at 4°C.

Plant cells and tissue cultures hold great promise for controlled production of a myriad of useful secondary metabolites on demand. The current yield and productivity cannot fulfill the commercial goal of a plant cell-based bioprocess for the production of most secondary metabolites. In order to stretch the boundary, recent advances, new directions and opportunities in plant cell-based bioprocessing, have been critically examined for the 10 years from 1992 to 2002. A review of the literature indicated that most of the R&D work was devoted predominantly to studies at an empirical level. A rational approach to molecular plant cell bioprocessing based on the fundamental understanding of metabolic pathways and their regulations is urgently required to stimulate further advances; however, the strategies and technical framework are still being developed. It is the aim of this review to take a step forward in framing workable strategies and technologies for molecular plant cell-based bioprocessing. Using anthocyanin biosynthesis as a case study, an integrated postgenomic approach has been proposed. This combines the functional analysis of metabolic pathways for biosynthesis of a particular metabolite from profiling of gene expression and protein expression to metabolic profiling. A global correlation not only can thus be established at the three molecular levels, but also places emphasis on the interactions between primary metabolism and secondary metabolism; between competing and/or complimentary pathways; and between biosynthetic and post-biosynthetic events.

The present study describes the development of a novel functional beverage through the application of probiotic Lactobacillus plantarum ATCC (American Type Culture Collection) 14917 in Cornelian cherry juice fermentation. The probiotic was employed in free and immobilized in a delignified wheat bran carrier (DWB) form. Cornelian cherry juice was fermented for 24 h and then it was stored at 4 °C for 4 weeks. Several parameters were evaluated such as residual sugar, organic acid and alcohol levels, total phenolics content, and cell viability as well as consumers acceptance. Regarding sugar and organic acids analyses, it was proved that the probiotic free or immobilized biocatalyst was effective. The concentration of ethanol was maintained at low levels (0.3–0.9% v/v). The total phenolic content of fermented Cornelian cherry juice with immobilized cells was recorded in higher levels (214–264 mg GAE/100 mL) for all the cold storage time compared to fermented juice with free cells (165–199 mg GAE/100 mL) and non-fermented juice (135–169 mg GAE/100 mL). Immobilized cells retained their viability in higher levels (9.95 log cfu/mL at the 4th week) compared to free cells (7.36 log cfu/mL at the 4th week). No significant sensory differences were observed among the fermented and the non-fermented samples.

The present work was conducted in order to investigate the effect of auxin type (2,4-D and NAA) and concentration (0.00, 0.25, 0.50, 1.00 and 2.00 mg/l) on Balanites aegyptiaca callus cultures growth and production of secondary metabolites. Obtained results demonstrated that supplementation MS medium with 2,4-D at 2.0 mg/l could enhanced and recorded the ultimate values of callus fresh weight, antioxidant activity (%), total flavonoids, total phenolic compounds and total saponins contents and yields of Balanites aegyptiaca L. callus.

Abstract The citric acid excretion of Ca-alginate-immobilized cells of Aspergillus niger in batch culture decreased with a half-time of approximately 19 days. Reactivation of the biocatalysts by regeneration in growth medium was possible, but it was followed by a submerged sporulation of the fungus, and medium was highly contaminated with free cells. Citric acid production could better be prolonged by semicontinuous cultivation with medium exchange every 7 or 14 days, respectively. After 32 days the remaining activity in semicontinuous culture was 1.4-fold higher than in comparable batch experiments. Similar improvements were obtained with a continuous process at a dilution rate of 0.125 v/v · d, whereby medium efflux completely free of detaching mycelia.
