The aim of the study was to characterize the population structure and assess the genetic diversity of warmblood horses used in the show jumping discipline. Pedigree data of 1,048 horses participating in the Polish Championships for Young Horses were analyzed. The pedigree of these animals included 12 863 individuals. The study consisted in analysis of the pedigree structure of the horses and characterization of the homozygosity and genetic diversity in the population. It was found that pedigree completeness and depth were sufficient for reliable assessment of the genetic diversity in the analyzed population. Although the average inbreeding coefficient exhibited at an acceptable level (approx. 1.01%), the increasing percentage of inbred animals seems disturbing. The results have shown that modern sport horses are derived from a small number of high-quality sires whose offspring were intensively used for breeding—bottleneck effect. In consequence, a greater part of the genetic variation reduction was observed in the non-founder generations. Given the changes in the studied population, the level of inbreeding in modern sport horses should be monitored, and pedigree data should be effectively used in selection for mating.

The Slovak Pinzgau breed faces the bottleneck effect and the loss of diversity due to unequal use of founders and significant population decline. Further population size reduction can lead to serious problems. High-throughput genotypes of 179 individuals were used to characterise genetic diversity and differentiation of Slovak Pinzgau, Austrian Pinzgau, Cika and Piedmontese cattle by Bayesian clustering algorithm. A gene flow network for the clusters estimated from admixture results was produced. The low estimate of genetic differentiation (FST) in Pinzgau cattle populations indicated that differentiation among these populations is low, particularly owing to a common historical origin and high gene flow. The changes in the log marginal likelihood indicated Austrian Pinzgau as the most similar breed to Slovak Pinzgau. All populations except Piedmontese showed two ways of gene flow among populations indicating that Piedmontese was involved in formation of analysed breeds while these breeds were not involved in creation of Piedmontese. Genetic evaluation represents an important tool in breeding and cattle selection. It is more strategically important than ever to preserve as much of the livestock diversity as possible, to ensure a prompt and proper response to the needs of future generations. The information provided by the fine-scale genetic characterization of this study clearly shows that there is a difference in genetic composition of Slovak and Austrian populations as well as Cika and Piedmontese. Slovak Pinzgau cattle despite its population size has the potential to serve as a basic gene reserve of this breed with the European and World importance.

Background: The genetic variability in a population is the raw material for selection, because the estimation of genetic parameters depends on the variability present within the population. The pedigree analysis is a method to assess population genetic variability. An increase in the level of inbreeding disturbs the production performance of the animals. Hence, it is essential to assess the effect of inbreeding on production performance of the animals at regular intervals. The present study was conducted on data of Bharat Merino (BM) sheep with twin objectives of evaluating the population structure by pedigree analysis and possible effect of inbreeding on lamb growth and heritability estimates. Methods: The study was conducted on data consisting of a total of 9688 pedigree records of BM sheep born from 1975 to 2018 (43 years), out of which 9050 formed population reference (with both the parents known). ENDOG ver 4.8 program was used to generate different measures of genetic diversity. General Linear Model of SPSS 25.0 was used to ascertain the effect of inbreeding (Fi) or change in inbreeding (ÄFi) on the lamb live weights. Using animal model with software WOMBAT, single trait linear mixed model analyses were performed. The heritability estimates and breeding values were obtained by including or excluding the inbreeding coefficient in the model to observe how the estimates of heritability varied with inclusion or exclusion of the inbreeding coefficient. Result: Effective number of founders (fe) was 56, constituting 11.39% founders in the population reference, while the effective number of ancestors (fa) was 43.The genetic contribution of the 15 most influent ancestors explained 50% of the genetic variability in the dataset. The ratio fe/fa, representing the effect of population bottleneck, was 1.302. The average inbreeding coefficients for the whole pedigree was 2.36%, while it was 3.84% for inbred animals. It was found that the inbreeding coefficient (Fi) increased with the addition of each generation to the pedigree. The average relatedness coefficient was 4.53% between members of the population.The effect of individual inbreeding (Fi) or the change in inbreeding (DFi) was not significant on the lamb live weights, except the effect of individual inbreeding (Fi) on three-month body weight and average daily gain (0-3month) and of change in inbreeding (DFi) on three-month body weight. From the analysis of the pedigree data of Bharat Merino sheep, it was found that the most of the measures of genetic diversity were within acceptable limits and the pedigree data was reasonably well maintained. When inbreeding was accounted for in the model, there were reductions in h2 estimates as well as the estimates of breeding values for both 3WT and ADG1 and consequently there were changes in ranking of animals for both 3WT and ADG1.

Cryptosporidiumfelis is an important cause of feline and human cryptosporidiosis. However, the transmission of this pathogen between humans and cats remains controversial, partially due to a lack of genetic characterization of isolates from cats. The present study was conducted to examine the genetic diversity of C. felis in cats in China and to assess their potential zoonotic transmission. A newly developed subtyping tool based on a sequence analysis of the 60-kDa glycoprotein (gp60) gene was employed to identify the subtypes of 30 cat-derived C. felis isolates from Guangdong and Shanghai. Altogether, 20 C. felis isolates were successfully subtyped. The results of the sequence alignment showed a high genetic diversity, with 13 novel subtypes and 2 known subtypes of the XIXa subtype family being identified. The known subtypes were previously detected in humans, while some of the subtypes formed well-supported subclusters with human-derived subtypes from other countries in a phylogenetic analysis of the gp60 sequences. The results of this study confirmed the high genetic diversity of the XIXa subtype family of C. felis. The common occurrence of this subtype family in both humans and cats suggests that there could be cross-species transmission of C. felis.

Scrub typhus is a vector-borne, acute febrile illness caused by Orientia tsutsugamushi. Scrub typhus continues to be an important but neglected tropical disease in Nepal. Information on this pathogen in Nepal is limited to serological surveys with little information available on molecular methods to detect O. tsutsugamushi. Limited information exists on the genetic diversity of this pathogen. A total of 282 blood samples were obtained from patients with suspected scrub typhus from central Nepal and 84 (30%) were positive for O. tsutsugamushi by 16S rRNA qPCR. Positive samples were further subjected to 56 kDa and 47 kDa molecular typing and molecularly compared to other O. tsutsugamushi strains. Phylogenetic analysis revealed that Nepalese O. tsutsugamushi strains largely cluster together and cluster away from other O. tsutsugamushi strains from Asia and elsewhere. One exception was the sample of Nepal_1, with its partial 56 kDa sequence clustering more closely with non-Nepalese O. tsutsugamushi 56 kDa sequences, potentially indicating that homologous recombination may influence the genetic diversity of strains in this region. Knowledge on the circulating strains in Nepal is important to the development of diagnostic tests and vaccines to support public health measures to control scrub typhus in this country.

In many dairy products, Leuconostoc spp. is a natural part of non-starter lactic acid bacteria (NSLAB) accounting for flavor development. However, data on the genomic diversity of Leuconostoc spp. isolates obtained from cheese are still scarce. The focus of this study was the genomic characterization of Leuconostoc spp. obtained from different traditional Montenegrin brine cheeses with the aim to explore their diversity and provide genetic information as a basis for the selection of strains for future cheese production. In 2019, sixteen Leuconostoc spp. isolates were obtained from white brine cheeses from nine different producers located in three municipalities in the northern region of Montenegro. All isolates were identified as Ln. mesenteroides. Classical multilocus sequence tying (MLST) and core genome (cg) MLST revealed a high diversity of the Montenegrin Ln. mesenteroides cheese isolates. All isolates carried genes of the bacteriocin biosynthetic gene clusters, eight out of 16 strains carried the citCDEFG operon, 14 carried butA, and all 16 isolates carried alsS and ilv, genes involved in forming important aromas and flavor compounds. Safety evaluation indicated that isolates carried no pathogenic factors and no virulence factors. In conclusion, Ln. mesenteroides isolates from Montenegrin traditional cheeses displayed a high genetic diversity and were unrelated to strains deposited in GenBank.
