Ten secondary metabolites, including a new grifolin analog, grifolin B (1); a new homovalencic acid derivative, 12-hydroxyhomovalencic acid (7); and a compound isolated from a natural source for the first time (9), along with seven known compounds, grifolin (2), averantin (3), 7-chloroaverantin (4), 1′-O-methylaverantin (5), 7-hydroxy-2-(2-hydroxypropyl)-5-pentylchromone (6), homovalencic acid (8), and bekeleylactone E (10), were isolated from two fungal strains. The structures of 1–10 were identified by detailed analysis and comparison of their spectroscopic data with literature values. Compounds 9 and 10 showed moderate cytotoxic activity against a panel of cancer cell lines (PC-3, HCT-15, MDA-MB-231, ACHN, NCI-H23, NUGC-3), with the GI50 values ranging from 1.1 µM to 3.6 µM, whereas 1 displayed a weak 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity without cytotoxicity against all tested cell lines.

Abstract Objectives Cacao(Theobroma cacao) has been widely consumed since more than 4000 years ago which was regarded as sacred food. Cacao has been considered as a strong antioxidant due to its abundant phenolic and especially flavonoid contents. In this experiment, different extraction solvents and methods were used to analyze whether these factors affect the antioxidant activities and phytochemical compounds of cacao powder extracts. Methods Four different cacao extract samples were examined to identify their anti-oxidative activities; raw cacao powder extracted in 70% methanol solution(RM), raw cacao powder extracted in 80% ethanol solution(RE), freeze dried cacao powder extracted in 70% methanol solution(FM), freeze dried cacao powder extracted in 80% ethanol solution(FE). Total phenolic compound content was calculated based on gallic acid standard curve and total flavonoid content was calculated based on the standard curve of quercetin. ABTS scavenging activity was experimented with ABTS reagent diluted by PBS(pH7.4) to make the O.D. value 0.7 ± 0.02 at 734 nm before using 2.5 mM ABTS. The DPPH radical scavenging activity was assessed by 1 mM DPPH methanol solution. Results The contents of total phenol compounds were 45.4 mg GAE/g(RM), 37.3 mg GAE/g(RE), 50.6 mg GAE/g(FM), and 52.7 mg GAE/g(FE). The contents of total flavonoid compounds were 34.3 mg QE/g(RM), 29.7 mg QE/g(RE), 85.1 mg QE/g(FM), and 89.7 mg QE/g(FE). FM group showed the highest ABTS radical scavenging activity which is a marker of antioxidant activity but the other sample groups represented almost as high ABTS radical scavenging activity as FM group. RM group showed the highest DPPH radical scavenging activity which is also a marker of antioxidant activity. Conclusions The results show that different extract solvents and sample preparation methods affect DPPH radical scavenging activity, total phenol and flavonoid contents of cacao powder extracts. Antibacterial and antioxidant activities of different extracts of saxicolous lichen Xanthoparmelia stenophylla sampled from Norashen, Gegharkunik Province of Armenia were studied. Methanol, ethanol and acetone extracts of lichen thalli were demonstrated to have activity against only tested gram-positive bacteria. Methanol extract of the lichen showed the highest amount of DPPH radical scavenging activity (~68%). Our studies did not reveal any significant antibacterial and antioxidant activities of aqueous extract.

Abstract Ginger (Zingiber officinale) is a popular spice which used for the treatment of different gastrointestinal and inflammatory discomfort. In the present study, the total phenolic content (TPC) and antioxidant activity of ginger extract using four solvents (ethanol, methanol, acetone and ethyl acetate) were determined. Among the four solvents, methanol extract showed that the maximum phenolic (1183.813 mg GAE/100 g at Ayikel and 1022.409 mg GAE/100 g at Mandura) and the least were found in acetone extract (748.865 mg GAE/100 g at Ayikel and 690.152 mg GAE/100 g at Mandura). In addition, the highest DPPH radical scavenging activity (84.868% at Ayikel and 82.883% at Mandura) was observed in methanol. However, acetone showed the least DPPH radical scavenging activity (73.864% at Ayikel and 70.597% at Mandura). Antioxidant activities of ginger extracts were also expressed as IC50 values and acetone extract has maximum IC50 value (0.654 and 0.812 mg/mL) followed by ethyl acetate and ethanol, while the lowest for methanol extracts (0.481 and 0.525 mg/mL). The result of this study showed that extraction solvents significantly affected the total phenolic content and antioxidant activities of ginger. Thus, ginger can be regarded as promising candidates for natural sources of antioxidants with high value of phenolic contents.

Tea and ginger are plants with high antioxidant potential. Combinations of antioxidants from different sources could also produce synergistic antioxidant effects. This study investigated the influence of solvent on antioxidant content of tea, ginger, and tea + ginger blends. Under the investigated extraction conditions, water was the most effective extraction solvent to maximise peroxide scavenging and iron chelating activity of tea, ginger, and their blends. Aqueous ethanol was the most effective solvent to maximise ABTS radical scavenging activity and ethanol was the best solvent to maximise DPPH radical scavenging activity. A good multivariate regression model that explains the relationship between the total flavonoid content of the extracts and their antioxidant activities was obtained (R2 and Q2 of 0.93 and 0.83, respectively). Extracts of tea-ginger blends exhibited synergistic effects in their ABTS and DPPH radical scavenging activity.

This research investigated the microbial quality and the protein contents of Thai commercial frozen products of silkworm (Bombyx mori), bamboo caterpillar (Omphisa fuscidentalis), and field cricket (Gryllus bimaculatus). Total mesophilic aerobic counts were about 8-8.4 log cfu/g, while lactic acid bacteria numbers were up to 5.2 log cfu/g samples. Yeasts and mold as well as Enterobacteriaceae numbers were found to be no more than 6.3 and 6.6 log cfu/g, respectively, while Clostridium spp. enumeration detected 3.2-3.6 cfu per gram frozen insect samples. The protein content in cases of cricket and bamboo caterpillar after the Soxhlet defatting method and the Folch lipid extraction combined with isoelectric point precipitation were similar when compared at pH 3.5 or pH 4.5. In contrast, the protein yield from silkworm was higher at pH 3.5 in the Soxhlet defatted sample, as opposed to the Folch method treated sample where higher protein yield was obtained at pH 4.5. Furthermore, 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP) of field cricket subcritical water (121 or 135°C for 15 or 30 minutes) hydrolysates were also measured on a pilot basis. These tests revealed higher antioxidant activities in treated samples than in the untreated control. The highest DPPH radical scavenging activity and FRAP values were detected in samples treated at 135°C, while the protein content of these samples was lower than that of the untreated control. These results indicate that hydrolytic compounds of proteins and probably other types of cricket materials are possibly involved in the antioxidant activities of the treated defatted cricket samples.

Banana is one of the most important crops, providing multiple benefits. Although it has been widely studied for its health benefits, little information can be found about its proteins. This study determined the antihypertensive and antioxidant activities of the crude, purified, and hydrolyzed protein extracts from ‘Señorita’ banana pulp. Crude proteins were extracted using Tris-HCl buffer and purified through ammonium sulfate precipitation, dialysis, and gel filtration chromatography. The protein content of the crude, partially purified, and purified extracts were found to be 167.32, 120.45, and 28.51 μg·mL−1, respectively, with major protein having an approximate molecular weight of 15 kDa. These extracts were then subjected to enzymatic hydrolysis for release of bioactive peptides prior to ACE inhibitory and antioxidant activities determination. Among these samples, the undigested crude extract had the highest ACE inhibitory activity (85.20%). There was also an observable increase in ACE inhibition of the digested samples with increased digestion time. Meanwhile, the 3-h and 4-h crude digests had the highest DPPH radical scavenging activity with 30.82% and 34.74%, respectively. These were not significantly different from the activity of the standard, ascorbic acid. A general decrease in DPPH radical scavenging activity of the samples was observed with increased digestion time. These observations were coherent with the in silico analysis of the putative major protein, lectin, which showed that its enzymatic hydrolysis releases ACE inhibitor and antioxidant peptides.

Seed coat color is one of the main agronomical traits that determine the chemical quality of soybean seeds and has been used as a parameter during cultivar development. In this study, seeds of yellow (n = 10), greenish-yellow (n = 5), and light-yellow (n = 4) soybean accessions were evaluated for their contents of total protein, total oil, total phenolic (TPC), and five prominent fatty acids including palmitic acid (PA), stearic acid (SA), oleic acid (OA), linoleic acid (LA), and linolenic acid (LLA), relative to a control cultivar, and the effect of seed coat color on each was investigated. Antioxidant activity was also evaluated using 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, Trolox equivalent antioxidant capacity (TEAC), and ferric reducing antioxidant power (FRAP). The results showed significant variations of metabolite contents and antioxidant activities between the soybeans. The average TPC, DPPH-radical scavenging activity, and FRAP were each in the order of greenish-yellow > yellow > light-yellow soybeans. In contrast, light-yellow soybeans contained a high level of OA and low levels of SA, LA, and LLA, each except LA differing significantly from yellow and greenish-yellow soybeans (p < 0.05). Our findings suggest that greenish-yellow and light-yellow soybeans could be good sources of antioxidants and high-quality soybean oil, respectively.

Objective: The objective of the present study was to evaluate the total phenolic (TPC) and flavonoid content (TFC) of five less utilized fruits such as Aegle marmelos, Spondias pinnata, Limonia acidissima, Averhoa carambola, Crescentia cujete and was compared with Phyllanthus emblica, (Amla) well known for its antioxidant activities. Total phenolic and flavonoid contents of samples were correlated with antioxidant activities like 1,1-Diphenyl-2-Picryl-Hydrazyl (DPPH) free radical scavenging assay, Ferric reducing antioxidant potential (FRAP) assay and total antioxidant capacity (TAC).Methods: The total phenolic of each fruit extract were determined by the Folin-Ciocalteu method with some modifications and the total flavonoids were estimated by Aluminum trichloride colourimetric method. The DPPH antioxidant assay, The FRAP assay and TAC were determined spectrophotometrically.Results: The total phenolics were expressed as mg/100g Gallic acid equivalent (mg GAE/100 gm) and the total flavonoids were expressed as mg/100g Quercetin equivalent (mg QE/100 gm). TPC was found to be maximum in Spondias pinnata with 142.16 mg GAE/100 gm where as TFC was maximum in Phyllanthus emblica with 91.1 mgQE/100 gm. DPPH radical scavenging activity was expressed n percentage(%), FRAP values expressed as mg/100g Ascorbic equivalent (AAE) and the total antioxidant activity was expressed as mg/100g Ascorbic equivalent. Maximum DPPH radical scavenging activity was shown by Spondias pinnata (93.75%), FRAP values were maximum in Phyllanthus emblica with 72.6 mg AAE/100 gm and total antioxidant capacity was found to be highest in Spondias pinnata (50.1 mg AAE/100 gm).Conclusion: Spondias pinnata, an underutilized fruit, was found to be promising with antioxidant activities comparable to Phyllanthus emblica.
