This research aims to understand the precise intracellular metabolic processes of how microbes solubilize insoluble phosphorus (Insol-P) to increase bio-available P. Newly isolated Penicillium oxalicum PSF-4 exhibited outstanding tricalcium phosphate (TP) and iron phosphate (IP) solubilization performance—as manifested by microbial growth and the secretion of low-molecular-weight organic acids (LMWOAs). Untargeted metabolomics approach was employed to assess the metabolic alterations of 73 intracellular metabolites induced by TP and IP compared with soluble KH2PO4 in P. oxalicum. Based on the changes of intracellular metabolites, it was concluded that (i) the enhanced intracellular glyoxylate and carbohydrate metabolisms increased the extracellular LMWOAs production; (ii) the exposure of Insol-P poses potential effects to P. oxalicum in destructing essential cellular functions, affecting microbial growth, and disrupting amino acid, lipid, and nucleotide metabolisms; and (iii) the intracellular amino acid utilization played a significant role to stimulate microbial growth and the extracellular LMWOAs biosynthesis.

Abstract Florunner peanut seeds contained five trypsin isoinhibitors. Amino acid profiles of the trypsin inhibitors fraction showed high levels of aspartic acid, half-cystine and serine and low levels of histidine and tyrosine. The molecular weight of the inhibitor was 8.3 KDa. The presence of multiforms of this inhibitor, its low molecular weight and the high amount of half-cystine indicate that peanut trypsin inhibitor is of the Bowman-Birk type.

Pure enamel was prepared using an original microdissection technic. Protein concentration was 375 μg per gram of enamel. Polyacrylamide gel electrophoresis showed a single fast-migrating zone containing a thin double band. Ultracentrifugation studies suggested that the proteins were of low molecular weight or of weak density. Absorption spectra showed a strong absorbance at 260nm. Amino acid analyses yielded a composition of 25% Gly, 13.5% Glu, 11% Ser, 11% Pro, 2% Cys and 2% Hyp. A glucidic content of 15% was estimated and glucose, galactose, mannose and fucose were identified. The organic matrix of enamel seemed to be constituted of two major glycoproteins probably fibrous but different from keratin.

Extracelluar matrix (ECM) proteins create complex networks of macromolecules which fill-in the extracellular spaces of living tissues. They provide structural support and play an important role in maintaining cellular functions. Identification of ECM proteins can play a vital role in studying various types of diseases. Conventional wet lab–based methods are reliable; however, they are expensive and time consuming and are, therefore, not scalable. In this research, we propose a sequence-based novel machine learning approach for the prediction of ECM proteins. In the proposed method, composition of k-spaced amino acid pair (CKSAAP) features are encoded into a classifiable latent space (LS) with the help of deep latent space encoding (LSE). A comprehensive ablation analysis is conducted for performance evaluation of the proposed method. Results are compared with other state-of-the-art methods on the benchmark dataset, and the proposed ECM-LSE approach has shown to comprehensively outperform the contemporary methods.
