Background: Literature data on the effect of methionine on functional activity and, especially, on morphological changes in the liver parenchyma in animals of different ages are sporadic, and research results are often ambiguous. Aim: The purpose of this work was to study and compare the morphofunctional changes in the liver of rats of different ages on prolonged administration of L-methionine. Material and Methods: The experiment was performed on 48 male Wistar rats of 3 and 15 months of age. Animals of the experimental group received L-methionine at a dose of 250 mg/kg body weight in addition to the standard diet, daily for 21 days. Histological preparations were prepared from liver tissue by a standard technique. Morphometry was performed on digital images using the computer program «Image J». Succinate dehydrogenase activity and protein concentration were determined in the suspension of hepatocyte mitochondria. Results: It was revealed that 21-day administration of L-methionine to rats led to hypertrophy of the hepatocyte nucleus, an increase in the nuclear-cytoplasmic ratio, the number of binuclear hepatocytes, and the nucleolus in the cell nucleus. The relative area of ​​the sinusoids network increased by 50% in 3-month-old animals. This indicated a better blood filling of the liver parenchyma. The increase in succinate dehydrogenase activity and protein concentration was revealed in the suspension of hepatocyte mitochondria of the experimental rats. This indicated an increase in the mitochondria energy potential and protein-synthetic activity. Conclusions: The administration of prophylactic doses of methionine to healthy rats leads to the appearance of pronounced morphological and functional signs of increased activity of hepatocytes. The severity of this effect has a distinct age-dependent character. In young rats, it is more pronounced than in mature rats. The results of the study are important for practical medicine when using methionine for therapeutic and prophylactic purposes.

Multiplexed proteomics using isobaric tagging allows for simultaneously comparing the proteomes of multiple samples. In this technique, digested peptides from each sample are labeled with a chemical tag prior to pooling sample for LC-MS/MS with nanoflow chromatography (NanoLC). The isobaric nature of the tag prevents deconvolution of samples until fragmentation liberates the isotopically labeled reporter ions. To ensure efficient peptide labeling, large concentrations of labeling reagents are included in the reagent kits to allow scientists to use high ratios of chemical label per peptide. The increasing speed and sensitivity of mass spectrometers has reduced the peptide concentration required for analysis, leading to most of the label or labeled sample to be discarded. In conjunction, improvements in the speed of sample loading, reliable pump pressure, and stable gradient construction of analytical flow HPLCs has continued to improve the sample delivery process to the mass spectrometer. In this study we describe a method for performing multiplexed proteomics without the use of NanoLC by using offline fractionation of labeled peptides followed by rapid “standard flow” HPLC gradient LC-MS/MS. Standard Flow Multiplexed Proteomics (SFloMPro) enables high coverage quantitative proteomics of up to 16 mammalian samples in about 24 h. In this study, we compare NanoLC and SFloMPro analysis of fractionated samples. Our results demonstrate that comparable data is obtained by injecting 20 µg of labeled peptides per fraction with SFloMPro, compared to 1 µg per fraction with NanoLC. We conclude that, for experiments where protein concentration is not strictly limited, SFloMPro is a competitive approach to traditional NanoLC workflows with improved up-time, reliability and at a lower relative cost per sample.

AAC Richard is a food grade soybean [Glycine max (L.) Merr] cultivar with yellow hilum, high protein concentration, and good processing quality for foreign and domestic soymilk, tofu, and miso markets. It has resistance to SCN (soybean cyst nematode) (Heterodera Glycines Ichinohe). AAC Richard was developed at the Agriculture and Agri-Food Canada (AAFC) Harrow Research and Development Centre (Harrow-RDC), Harrow, Ontario and is adapted to areas of southwest Ontario with 3100 or more crop heat units and has a relative maturity of 2.3 (MG 2.3).

Colostrum provides the newborn with nutrients and immunoglobulins. Immunoglobulins and their intestinal transfer play a major role in the immune system of neonates since they are born agammaglobulinemic. In this study immunoglobulin G (IgG) content was determined in alpaca colostrum and the correlations of the IgG concentration by fat, protein, lactose and minerals were calculated. Colostrum samples were collected daily from 20 multiparous alpaca mares during the first four days after parturition. The IgG concentrations were determined by radial immunodiffusion using a Camelid IgG Test Kit. The IgG concentration decreased significantly from 26,319 mg/dL on day 1 to 3848.8 mg/dL on day 4. There were significant correlations between IgG concentration and the other components of the colostrum. While the correlations between IgG and fat (r = −0.69, p ≤ 0.001) and lactose (r = −0.64, p ≤ 0.001) were negative, the correlations with protein (r = 0.91, p ≤ 0.001), magnesium (r = 0.86, p ≤ 0.001) and cobalt (r = 0.87, p ≤ 0.001) were strongly positive. Due to the strong association, the colostrum protein concentration could be used for a brief estimation of the IgG content.

CDC Hickie, a yellow cotyledon field pea (Pisum sativum L.) cultivar, was released in 2021 by the Crop Development Centre, University of Saskatchewan for distribution to Select seed growers through the Variety Release Committee of the Saskatchewan Pulse Growers. CDC Hickie has good lodging resistance, medium time to maturity, medium-sized, round seeds, mean seed protein concentration of 24.4%, and good yielding ability. It is resistant to powdery mildew and moderately susceptible to mycosphaerella blight and Fusarium root rot. CDC Hickie is adapted to the field pea growing regions of western Canada.

Fourty five local buffaloes naturally infected with A. marginal age of 2-5 years. Clinical signs, present parasireamia haematological and biochemical changes were studied. Thin thick blood smears Stained with Gimsas stain were used to identify the parasite. The results indicated that most of the infected were suffering from mild form of infection and the 1I11p01”l11I1E' clinical signs observed were mild rise of body temperature, increase respiratory and plus rate, weal: ruminal contraction and generalized weakness. Palcness and iteration of mucous membrane, loss appetite, nasal and ocular discharge, coughing, salivation, atarcp, constipation, diarrhea, and rccumbancy. Statistical analysis showc increase (P<0.01) in body temperature, respiratory and pulsera wlflc nigrrificant decrease (P<0.01) in ruminal contraction We. observed. Parasiteamia in infected animals ranged from 1.3- 5.2%. Values of red blood cell, haemoglobin and packed cell volume were significantly low (P<0.01). Total leulcocyric and neutrophils count was significantly increase (P<0.0l). and (P<0.05) respectively. Blood indices values showed a significant increase in (MCHC) (P<0.0l) while (MCV) values showed no difference, reticulocytes were also present mildly biochemical changes indicated higher levels of AST. Total bilirubin (P<0.01). Total protein concentration was less (P<0.01) and no difference was observed inALT and blood urea nitrogen levels between infected and control animals.

AbstractOn-farm soybean yield has increased considerably in the last 50 years in southern Brazil, but there is still little information about how selection and breeding for yield increase has changed the agronomic attributes of cultivars. The objectives of this study were to evaluate the changes in soybean yield, seed oil and protein concentration, and changes in plant attributes that might be associated with yield improvement of 26 soybean cultivars released over the past 50 years in southern Brazil, sown simultaneously in a common field environment for two growing seasons. The average rate of yield gain was 45.9 kg ha−1 yr−1 (2.1% ha−1 yr−1), mainly due increased seed number per area and harvest index. Over year of cultivar release, cultivars became less susceptible to lodging, as well as plant mortality reduced. Meanwhile, the seed oil concentration increased, and seed protein concentration decreased, which could have negative consequences for soybeans use and requires further attention for breeding of future cultivars. Breeders have successfully contributed to the annual rate of soybean yield increase in southern Brazil. By our results, as well as the official on-farm production data, there is no evidence of soybean yield reaching a plateau in the near future in southern Brazil.
