Bovine herpesvirus type-1 (BoHV-1) is a widespread pathogen that provokes infectious rhinotracheitis and polymicrobial infections in cattle, resulting in serious economic losses to the farm animal industry and trade restrictions. To date, non-toxic active drugs against BoHV-1 are not available. The exploitation of bioactive properties of microbial products is of great pharmaceutical interest. In fact, fungi are a promising source of novel drugs with a broad spectrum of activities and functions, including antiviral properties. Hence, the potential antiviral properties of 3-O-methylfunicone (OMF), a secondary metabolite produced by Talaromyces pinophilus, were evaluated on BoHV-1. In this study, during BoHV-1 infection in bovine cells (MDBK), the non-toxic concentration of 5 µM OMF considerably reduced signs of cell death and increased cell proliferation. Furthermore, OMF significantly decreased the virus titer as well as the cytopathic effect and strongly inhibited the expression of bICP0, the major regulatory protein in the BoHV-1 lytic cycle. These findings were accompanied by a considerable up-regulation in the expression of the aryl hydrocarbon receptor (AhR), a multifunctional transcription factor also linked to the host’s response to a herpesvirus infection. Overall, our results suggest that by involving AhR, OMF shows potential against a BoHV-1 infection.

Bovine herpesvirus type 1 (BoHV-1) is recognized as a major cause of economic losses in cattle. Vaccination has been widely applied to minimize losses induced by BoHV-1 infections. We have previously reported the development of a differential BoHV-1 vaccine, based on a recombinant glycoprotein E (gE)-deleted virus (265gE-). In present paper the efficacy of such recombinant was evaluated as an inactivated vaccine. Five BoHV-1 seronegative calves were vaccinated intramuscularly on day 0 and boostered 30 days later with an inactivated, oil adjuvanted vaccine containing an antigenic mass equivalent to 10(7.0) fifty per cent cell culture infectious doses (CCID50) of 265gE-. Three calves were kept as non vaccinated controls. On day 60 post vaccination both vaccinated and controls were challenged with the virulent parental strain. No clinical signs or adverse effects were seen after or during vaccination. After challenge, 2/5 vaccinated calves showed mild clinical signs of infection, whereas all non vaccinated controls displayed intense rhinotracheitis and shed virus for longer and to higher titres than vaccinated calves. Serological responses were detected in all vaccinated animals after the second dose of vaccine, but not on control calves. Following corticosteroid administration in attempting to induce reactivation of the latent infection, no clinical signs were observed in vaccinated calves, whereas non vaccinated controls showed clinical signs of respiratory disease. In view of its immunogenicity and protective effect upon challenge with a virulent BoHV-1, the oil adjuvanted preparation with the inactivated 265gE- recombinant was shown to be suitable for use as a vaccine.

Bovine herpesvirus type 1 infection (IBR/IPV virus) presents the most spread infection in nowadays cattle breeding. Bovine herpesvirus-1 (BHV-1) may be an agent of serious health disorders causing great economic losses. Since the largest number of cattle in Southern Baåka and Srem district are raised in small herds (what may considerably influence effective implementation of program for control and eradication of BHV-1 infection) it was necessary to investigate the prevalence of BHV-1 infection in the cattle population in this area, what was the aim of this work. Detection and estimation of the presence and prevalence of BHV-1 was done by detection of specific antibodies against BHV-1 in sera samples of cattle raised individually or in small herds (up to 20 animals) collected during the State Program of Disease Control in 2005 and 2006. The sampling was randomly carried out taking care that the samples from all the settlements and municipalities in Southern Bačka and Srem were present. A total of 16,610 samples was collected. Animals seropositive for BHV-1 were detected in all municipalities, but not in all settlements. The highest percent of seropositive animals was found in Beočin municipality(27.27%), Titel municipality (27.16%), Žabalj (22.45%) and Stara Pazova municipality (22.15%). The lowest prevalence was in Bački Petrovac (8.16%) and Temerin municipalities (9.68%). In other municipalities the prevalence ranged between 10% and 20%. From the obtained results it can be concluded that BHV-1 infection was not present in 10 settlements in Southern Bačka and 14 settlements in Srem district. In other settlements (151) the percentage of seropositive animals ranged from 1 to 50%. However, if we analyze the obtained results in the area, it may be concluded that the prevalence is almost the same: in Srem district there were 18.42% and in Southern Bačka 18.79% seropositive animals. Low prevalence was detected in the majority of settlements. The data that in 24 settlements in both districts there were no seropositive animals His an encouraging information. This points on a possibility of easier and faster implementation of control measures and eradication of BHV-1 infection. The results obtained in serology examination provide us an insight in immunology status of small herds in Southern Bačka and Srem districts, what is a starting point in designing and implementing necessary measures with the aim to prevent cases of this disease, as well as its spreading and eradication.

The specificity of monoclonal antibodies (mAbs) to desired targets makes these molecules suitable for therapeutic and diagnostic uses against a wide range of pathogens. Phage display antibody libraries offer one method by which mAbs can be selected for, without the use of conventional hybridoma technology. In this work, phage display technology was used to construct, select and characterize a combinatorial single chain fragment variable (scFv) antibody library against bovine herpesvirus type 1 (BoHV-1) from the immune repertoire of chickens immunized with the virus. In silico analysis of the hypervariable domains of the antibody heavy chains revealed a high frequency of scFv fragments with low variability, suggesting that selection had probably been carried out and favored by a few im-munogenic viral antigens. The reactivity of the scFv fragments selected against BoHV-1 was demon-strated by Phage-ELISA. A significant increase in antibody reactivity to the target was observed after six rounds of library selection, showing its potential use as a molecule for BoHV-1 diagnosis. The strategy described here opens up a field for the use of phage display as a tool for selection of mono-clonal antibodies that could be used for theranostic applications against infectious and parasitic dis-eases of veterinary interest.
