Real-time monitoring of product titers during process development and production of biotherapeutics facilitate implementation of quality-by-design principles and enable rapid bioprocess decision and optimization of the production process. Conventional analytical methods are generally performed offline/at-line and, therefore, are not capable of generating real-time data. In this study, a novel fiber optical nanoplasmonic sensor technology was explored for rapid IgG titer measurements. The sensor combines localized surface plasmon resonance transduction and robust single use Protein A-modified sensor chips, housed in a flexible flow cell, for specific IgG detection. The sensor requires small sample volumes (1–150 µL) and shows a reproducibility and sensitivity comparable to Protein G high performance liquid chromatography-ultraviolet (HPLC-UV). The dynamic range of the sensor system can be tuned by varying the sample volume, which enables quantification of IgG samples ranging from 0.0015 to 10 mg/mL, without need for sample dilution. The sensor shows limited interference from the sample matrix and negligible unspecific protein binding. IgG titers can be rapidly determined in samples from filtered unpurified Chinese hamster ovary (CHO) cell cultures and show good correlation with enzyme-linked immunosorbent assay (ELISA).

Determining the current position of objects in logistics processes (products, unit loads, forklifts, etc.) is the basis for new process development and optimization opportunities (e.g., development of storage strategy, forklift route planning, etc.) that significantly affect the competitiveness of companies. Background information systems that provide real-time data using localization, - or called RTLS (Real Time Location System),- are becoming more widespread. In the dissertation I summarize the operation, physical realization, conditions of use and advantages of a real-time technology that can be applied to a warehouse serving its production, as well as the steps of a possible implementation.

The sensor technology is applied to the discus sports, which can be called digital discus, a real-time data acquisition of the moving discus information can be easily gained. The system uses statistical analysis and data mining to do analysis with these collected data. It's solved the problem on getting the accurate kinetic data and kinematics data of the moving discus for the sports like discus, and proved significant data for study athlete's technical movement and achievement.

Holistic energy management in the shipping industry involves reliable data collection, systematic processing and smart analysis. The era of digitisation allows sensor technology to be used on-board vessels, converting different forms of signal into a digital format that can be exported conveniently for further processing. Appropriate sensor selection is important to ensure continuous data collection when vessels sail through harsh conditions. However, without proper processing, this leads to the collection of big data sets but without resulting useful intelligence that benefits the industry. The adoption of digital and computer technology, allows the next phase of fast data processing. This contributes to the growing area of big data analysis, which is now a problem for many technological sectors, including the maritime industry. Enormous databases are often stored without clear goals or suitable uses. Processing of data requires engineering knowledge to ensure suitable filters are applied to raw data. This systematic processing of data leads to transparency in real time data display and contributes to predictive analysis. In addition, the generation of series of raw data when coupled with other external data such as weather information provides a rich database that reflects the true scenario of the vessel. Subsequent processing will then provide improved decision making tools for optimal operations. These advances open the door for different market analyses and the generation of new knowledge. This paper highlights the crucial steps needed and the challenges of sensor installation to obtain accurate data, followed by pre and post processing of data to generate knowledge. With this, big data can now provide information and reveal hidden patterns and trends regarding vessel operations, machinery diagnostics and energy efficient fleet management. A case study was carried out on a tug boat that operates in the North Sea, firstly to demonstrate confidence in the raw data collected and secondly to demonstrate the systematic filtration, aggregation and display of useful information.

ABSTRACT A real-time immunoquantitative PCR (iqPCR) method for detection of Staphylococcus aureus enterotoxin B (SEB) was developed and evaluated using both pure cultures and foods. The assay consisted of immunocapture of SEB and real-time PCR amplification of the DNA probe linked to the detection antibody. iqPCR was compared to an in-house enzyme-linked immunosorbent assay (ELISA) using the same couple of capture-detection antibodies and to commercial kits for detection of S. aureus enterotoxins (SE). The iqPCR was approximately 1,000 times more sensitive (<10 pg ml−1) than the in-house ELISA and had a dynamic range of approximately 10 pg ml−1 to approximately 30,000 pg ml−1. iqPCR was not inhibited by any of the foods tested and was able to detect SEB present in these foods. No cross-reactivity with SE other than SEB was observed. Application of iqPCR for detection of SEB in cultures of S. aureus revealed the onset of SEB production after 4 h of incubation at 22, 37, and 42°C, which was in the first half of the exponential growth phase. The total amounts of SEB produced by the two strains tested were larger at 42°C than at 37°C and were strain dependent.

Convergent experimental data, autobiographical studies, and investigations on daily life have all demonstrated that gathering information retrospectively is a highly dubious methodology. Retrospection is subject to multiple systematic distortions (i.e., affective valence effect, mood congruent memory effect, duration neglect; peak end rule) as it is based on (often biased) storage and recollection of memories of the original experience or the behavior that are of interest. The method of choice to circumvent these biases is the use of electronic diaries to collect self-reported symptoms, behaviors, or physiological processes in real time. Different terms have been used for this kind of methodology: ambulatory assessment, ecological momentary assessment, experience sampling method, and real-time data capture. Even though the terms differ, they have in common the use of computer-assisted methodology to assess self-reported symptoms, behaviors, or physiological processes, while the participant undergoes normal daily activities. In this review we discuss the main features and advantages of ambulatory assessment regarding clinical psychology and psychiatry: (a) the use of realtime assessment to circumvent biased recollection, (b) assessment in real life to enhance generalizability, (c) repeated assessment to investigate within person processes, (d) multimodal assessment, including psychological, physiological and behavioral data, (e) the opportunity to assess and investigate context-specific relationships, and (f) the possibility of giving feedback in real time. Using prototypic examples from the literature of clinical psychology and psychiatry, we demonstrate that ambulatory assessment can answer specific research questions better than laboratory or questionnaire studies.
