The results of efficiency studies by major economic and energetic parameters of pretreatment of soybean with inoculants based on rhizobia Bradyrhizobium japonicum and associative diazotrophs Azospirillum brasilense are provided. It was established that the highest levels of the studied parameters are achieved using mixed cultures of nitrogen fixing microorganisms. The high economic and energetic efficiency was also observed during inoculation of soybean with rhizobia pure culture. However, no significant positive impact of A. brasilense applied separately was found in terms of studied parameters.

Objective. Investigate the activity of biosynthesis of phytohormonal substances with nitrogen-fixing bacteria Bradyrhizobium japonicum and Azospirillum brasilense in pure and mixed culture. Methods. Microbiological, chromatographic, and mathematical. Results It has been established that the simultaneous cultivation of B. japonicum M-8 and A. brasilense 410 increases the content of gibberellins and cytokinins in the culture fluid of the test microorganisms. The content of gibberellic acid and isopentenylidene has increased most intensively in mixed culture compared with the pure culture of rhizobia. In the course of co-cultivation, the studied diazotrophs more intensively produced auxins compared to soybean rhizobia in pure culture, but less compared to pure culture of azospirilla. The highest level of abscisic acid that can inhibit the formation of nodules was found in A. brasilense 410 culture fluid, and it was lower when cultivating B japonicum M-8. However, the smallest amount of this phytohormone was found in the culture liquid of diazotrophs under their co-cultivation. The lowest ratio of auxin/cytokinin was found in B. japonicum M-8 and A. brasilense 410 culture fluid under their co-cultivation, which should positively influence the formation of a symbiotic system when interacting with soybean plants. Conclusion. A combination of cultivating rhizobia and azospirilla showed an increase in the amount of cytokinins and gibberellins in the culture fluid of the microorganisms, a decrease in the amount of abscisic acid and improvement in the auxin/cytokinin ratio compared to the values of the pure cultures of the nitrogen-fixing bacteria studied. An analysis of the quantitative parameters of the content of phytohormones suggests the feasibility of combining B. japonicum and A. brasilense in a mixed culture for the effective introduction of rhizobia in soybean agrocenosis.

The paper presents the research results of pre-sowing seeds bacterization with nitrogen fixing bacteria Bradyrhizobium japonicum and Azospirillum brasilense influence on the loss of moisture, water soluble humus and nutrients. It was shown that seeds bacterization reduces the leaching intensity of nitrates, phosphorus, potassium, calcium and magnesium. Moreover, the least losses of nutrients were observed in a variants with joint application of both studied nitrogen fixing microorganisms. It was noted that seeds bacterization with B. japonicum and A. brasilense had promoted increase of chlorophyll content in the leaves of soybean plants. The highest yield was observed in the variant with the joint use of rhizobia and azospirillum.


Rumen ciliates still have mysterious secrets and influences in ruminants. This study investigated the effect of transfaunation of pure and mixed cultures of rumen ciliates on physical clinical examination, selected serum parameters and milk profile in defaunated lactating dairy goats. A number of 8 Baladi native breed goats were randomly classified into two groups each one containing 4 goats. Pure culture group was transfaunated with 6 ml of pure culture of Holotricha spp., while mixed culture group was transfaunated with 6 ml of mixed culture of 81.85% Holotricha and 18.15% Ophryoscolex spp. once weekly for three consecutive weeks, after defaunation of both groups using 30 ml of 8% SLS for two consecutive days. Serum and milk samples were collected weekly for three successive weeks to study effect of type of ciliate culture, duration of transfaunation and their interaction. Results revealed that transfaunation of pure and mixed cultures of rumen ciliates had no effect on physical examination with minimal non-significant improvement of calcium, inorganic phosphorous, total protein and globulin in serum of defaunated goats. Transfaunation of pure or mixed cultures of rumen ciliates within three weeks could not improve significantly decreased milk fat % of defaunated goats without any effect on other measured milk profile parameters. It is concluded that further investigations on transfaunation without prior defaunation should be performed using different pure and mixed cultures of rumen ciliates for therapeutic and productive purposes.

O tamanho da semente é um importante indicador físico da qualidade fisiológica das sementes, pois pode afetar a taxa de germinação e o crescimento inicial das plântulas. Neste contexto, o presente estudo teve como objetivo avaliar os efeitos do tamanho das sementes e da inoculação de Bradyrhizobium japonicum e Azospirillum brasilense de forma isolada ou combinada na emergência das plântulas, nodulação das raízes e no crescimento inicial das plantas de amendoim (Arachis hypogaea L., cv. IAC Tatu ST). As plantas foram cultivadas em vasos plásticos de 8,0 L preenchidos com um solo arenoso do Cerrado e mantidas sob condições de casa-de-vegetação. O delineamento experimental utilizado foi o de blocos ao acaso em esquema fatorial 3 × 4 com quatro repetições. Os tratamentos foram constituídos por três tamanho de sementes (pequena, média e grande) e por quatro tratamentos de inoculação: i) controle (sem inoculação); ii) inoculação das sementes com Bradyrhizobium japonicum; iii) inoculação das sementes com Azospirillum brasilense; e, iv) coinoculação das sementes com B. japonicum e A. brasilense. Os resultados reportaram que a utilização de sementes grandes melhorou a nodulação das raízes, o crescimento das plantas, o acúmulo e a partição de matéria seca das plantas. Portanto, a implantação da cultura do amendoim deve ser realizada com a utilização de sementes de maior tamanho (grandes), por proporcionar sustentabilidade para o sistema de produção de amendoim, melhorando a fixação biológica de nitrogênio e o crescimento das plantas cultivadas em solos arenosos do Cerrado Sul-mato-grossense. A coinoculação das sementes com B. japonicum e A. brasilense melhorou a nodulação das raízes e a altura das plantas, no entanto, não teve efeito no crescimento, na partição de matéria seca e nos índices morfofisiológicos das plantas de amendoim, quando cultivadas em solos arenosos do Cerrado sob condições controladas.

ABSTRACT This study aimed to verify the effect of co-inoculation, association between Azospirillum brasilense and Bradyrhizobium japonicum bacteria, on soybean plants subjected to water deficit at two sowing dates. Two field experiments were conducted at the Universidade Federal de Tocantins, campus of Palmas, Brazil, in 2016. The experimental design was randomized blocks in a split-split-plot arrangement with four repetitions, where the plots consisted of two irrigation depths (100 and 25% of crop evapotranspiration - ETc), the subplots was composed of two methods of inoculant application (inoculation with Bradyrhizobium japonicum and co-inoculation with Azospirillum brasilense + Bradyrhizobium japonicum) and the sub-subplots comprised two soybean cultivars (TMG 132 and ANTA 82). The cultivars responded differently to the sowing dates. Co-inoculation did not influence grain yield under full irrigation conditions (100% ETc), in neither cultivar evaluated. However, under the water deficit condition (25% ETc), the grain yield of the cultivar TMG 132 increased 77.20%, indicating that there are different responses of interaction between Azospirillum brasilense, plant genotype and sowing dates.

ABSTRACT In many bacteria, the ferric uptake regulator (Fur) protein plays a central role in the regulation of iron uptake genes. Because iron figures prominently in the agriculturally important symbiosis between soybean and its nitrogen-fixing endosymbiont Bradyrhizobium japonicum, we wanted to assess the role of Fur in the interaction. We identified a fur mutant by selecting for manganese resistance. Manganese interacts with the Fur protein and represses iron uptake genes. In the presence of high levels of manganese, bacteria with a wild-type copy of the fur gene repress iron uptake systems and starve for iron, whereas fur mutants fail to repress iron uptake systems and survive. The B. japonicum fur mutant, as expected, fails to repress iron-regulated outer membrane proteins in the presence of iron. Unexpectedly, a wild-type copy of the fur gene cannot complement the fur mutant. Expression of the fur mutant allele in wild-type cells leads to a fur phenotype. Unlike a B. japonicum fur-null mutant, the strain carrying the dominant-negative fur mutation is unable to form functional, nitrogen-fixing nodules on soybean, mung bean, or cowpea, suggesting a role for a Fur-regulated protein or proteins in the symbiosis.
