DISORDERS OF THE SKIN’S BARRIER FUNCTION IN PATIENTS WITH ATOPIC DERMATITIS WITH MUTATIONS OF THE FILAGGRIN GENE

Introduction: The paper presents the results of investigations of the skin’s barrier function in patients diagnosed with atopic dermatitis (AD) with the presence of one of the genetic predispositions, namely the filaggrin gene FLG 2284del4 mutation. The aim: to deepen the study of the relationship between the mutations of the gene FLG del2282 and the morphological features of the skin, since the data on its physiological state in patients diagnosed with AD are contradictory that will contribute to the improvement of the modern basis necessary for the development of treatment-diagnostic and preventive measures. Materials and methods: In the given study of patients diagnosed with AD (41 person), the frequency of the FLG 2282del4 gene deletion constituted 19.51% (8 patients) according to the method of Palmer et al. To determine the functional parameters of the skin in patients diagnosed with AD in the carriers of the FLG gene mutation and in patients void of this mutation, they have applied a multifunctional Aramo TS portable analyzer that enables to carry out dermatoscopic diagnosis of the skin surface. Results and conclusions: As a result of the analysis of the obtained physiological parameters of the skin in the patients of both groups during the treatment process, it is noteworthy that the dynamics of these indices after treatment in patients void of the filaggrin gene mutation is significantly better in comparison with the patients in whom the filaggrin 2282 del14 gene mutation was revealed. There is a tendency towards the decrease of skin’s moisture and elasticity in patients with revealed mutation compared with the ones who are void of it. Given this fact, the study of physiological parameters of the skin is important to consider when substantiating the diagnostic-treatment and prophylactic algorithm for patients diagnosed with AD.

Проведена статистическая обработка сформированной базы данных показателей неспецифической резистентности MS «Excel» с целью изучения динамики иммунитета по параметрам бактерицидной, лизоцимной и фагоцитарной активности у крупного рогатого скота черно-пестрой породы разных возрастов. Было установлено, что средние значения показателей лизоцимной и бактерицидной активности у телят в 6 мес были ниже (47,89% и 49,09%, соответственно), чем у животных в 3—4 года (53,75% и 52,81%). Выведено уравнение регрессии, описывающее взаимосвязь между средними показателями фагоцитарной активности и возрастом при высокой степени достоверности (R2=0,90). Изменения лизоцимной и бактерицидной активности в зависимости от возраста были недостоверными (R2=0,58 и R2=0,49, соответственно). Установлена высокая корреляция между бактерицидной и лизоцимной активностью. Отмечена динамика изменения неспецифического иммунитета у коров различных возрастных групп. Создание баз данных и постоянный мониторинг основных показателей естественной резистентности в зависимости от различных факторов (породы, возраста, стадий лактации, кормления, способов содержания), которые влияют на данные показатели, необходимо для оценки физиологического состояния организма животных и их устойчивости к болезням. Statistical processing of the generated MS "Excel" on indicators of non-specific resistance database was carried out with the aim of studying the dynamics of the development of immunity in terms of bactericidal, lysozyme and phagocytic activity in black-and-white cattle of different ages. It was found that the average values of indicators of lysozyme and bactericidal activity in calves at 6 months were lower (47.89% and 49.09%, respectively) than in animals at the age of 3—4 years (53.75% and 52.81%). The equation of regression was derived, it describes the relationship between the average indicators of phagocytic activity and the age with a high degree of reliability (R2=0.90). The changes in lysozyme and bactericidal activity depending on age were dubious (R2=0.58 and R2=0.49, respectively). The high correlation between bactericidal and lysozyme activity was established. It was noticed that the cows of different age groups experienced the dynamics of changes in nonspecific immunity. The creation of databases and constant monitoring of the main indicators of natural resistance, depending on various factors (breed, age, stages of lactation, feeding, methods of keeping) that affect these indicators, is necessary for assessing the physiological state of the animals’ body of and their resistance to diseases.

Cornified cells of the stratum corneum have a monolayer of an unusual lipid covalently attached to the outer surface. This is referred to as the corneocyte lipid envelope (CLE). It consists of a monolayer of ω-hydroxyceramides covalently attached to the outer surface of the cornified envelope. The CLE is essential for proper barrier function of the skin and is derived from linoleate-rich acylglucosylceramides synthesized in the viable epidermis. Biosynthesis of acylglucosylceramide and its conversion to the cornified envelope is complex. Acylglucosylceramide in the bounding membrane of the lamellar granule is the precursor of the CLE. The acylglucosylceramide in the limiting membrane of the lamellar granule may be oriented with the glucosyl moiety on the inside. Conversion of the acylglucosylceramide to the CLE requires removal of the glucose by action of a glucocerebrosidase. The ester-linked fatty acid may be removed by an as yet unidentified esterase, and the resulting ω-hydroxyceramide may become ester linked to the outer surface of the cornified envelope through action of transglutaminase 1. Prior to removal of ester-linked fatty acids, linoleate is oxidized to an epoxy alcohol through action of 2 lipoxygenases. This can be further oxidized to an epoxy-enone, which can spontaneously attach to the cornified envelope through Schiff’s base formation. Mutations of genes coding for enzymes involved in biosynthesis of the CLE result in ichthyosis, often accompanied by neurologic dysfunction. The CLE is recognized as essential for barrier function of skin, but many questions about details of this essentiality remain. What are the relative roles of the 2 mechanisms of lipid attachment? What is the orientation of acylglucosylceramide in the bounding membrane of lamellar granules? Some evidence supports a role for CLE as a scaffold upon which intercellular lamellae unfold, but other evidence does not support this role. There is also controversial evidence for a role in stratum corneum cohesion. Evidence is presented to suggest that covalently bound ω-hydroxyceramides serve as a reservoir for free sphingosine that can serve in communicating with the viable epidermis and act as a potent broad-acting antimicrobial at the skin surface. Many questions remain.

In order to be able to use methods of universal algebra for investigating posets, we assigned to every pseudocomplemented poset, to every relatively pseudocomplemented poset and to every sectionally pseudocomplemented poset, a certain algebra (based on a commutative directoid or on a λ-lattice) which satisfies certain identities and implications. We show that the assigned algebras fully characterize the given corresponding posets. A certain kind of symmetry can be seen in the relationship between the classes of mentioned posets and the classes of directoids and λ-lattices representing these relational structures. As we show in the paper, this relationship is fully symmetric. Our results show that the assigned algebras satisfy strong congruence properties which can be transferred back to the posets. We also mention applications of such posets in certain non-classical logics.
