Calcium phosphate ceramics are one of the most studied biomaterials for bone substitution and regeneration. Bioactivity is one of the most important properties for these materials and it can be evaluated by adsorption of proteins and by hydroxyapatite formation on the surface in simulated body fluid (SBF). The aim of this work was to evaluate the bioactivity of ceramic granules with various hydroxyapatite (HAp) and beta tricalcium phosphate (β-TCP) ratios by adsorption of bovine serum albumin (BSA) in SBF and phosphate saline buffer (PBS) solution. The highest adsorption capacity in both solutions was observed for biphasic calcium phosphate sample with HAp/β-TCP ratio 50/50 but the lowest – for sample 10/90. The adsorption capacity of all samples was more than 2 times higher in SBF media than in PBS. The possibility of hydroxyapatite formation was evaluated by changes of Ca2+ concentration in SBF. After 5 days of incubation at 37oC all samples showed a decrease in Ca2+ concentration.

Studying on the interaction between proteins and calcium phosphate implants is one of the basic subjects in biomaterials science and engineering. In this work, zeta potentials and contact angles of hydroxyapatite (HA) and biphasic calcium phosphate (BCP) ceramics were measured, and the adsorption behaviors of bovine serum albumin (BSA) on the surfaces were investigated. The adsorption isotherms of BSA on both ceramics follow the Langmuir type, however, BCP shows higher BSA adsorption ability. The differences of contact angles and zeta potentials in HA and BCP are the key factors to determine their ability to bind BSA molecules.

This study examined the possibility of synthesis of biphasic calcium phosphate by sintering a mixture of hydroxyapatite and calcium phosphate glass. The effect of the concentration of calcium phosphate glass in a mixture on the proliferation and differentiation of MG-63 preosteoblast-like cells in a hydtoxyapatite scaffold was investigated. The addition of 5 wt% of calcium phosphate glass significantly improved the level of attachment, proliferation and differentiation of MG-63 cells onto the hydroxyapatite scaffolds, particularly when the surface was modified with 2% bovine serum albumin (p<0.05). Under these conditions, type I collagen was expressed and the extracellular matrix was formed by 1 week, and the ALP gene was expressed at 4 weeks.

Sorption experiments of bovine serum albumin (BSA) on hydroxyapatite (HAp) ceramic granules, prepared at three temperatures 900°C, 1000°C and 1150°C were performed at room temperature 18,6 °C and phosphate buffer, pH 5,83; 6.38 and 7,39. Thermal treatment contributed to the decrease of bovine serum albumin immobilization indicating that sorption process depended on HAp ceramics specific surface area and pH values of phosphate buffer solution. However, it was confirmed that granule size was also an important parameter for bovine serum albumin adsorption. As a result of these experiments, the most appropriate adsorption conditions and phosphate buffer pH values influence on to BSA sorption were analyzed.
