The present study aimed to assess the effects of square dance exercise on the cognitive function and quality of life in older women with mild cognitive impairment and to investigate the mediating role of a depressed mood and reduced quality of life in the relationship between square dance exercise and cognition. The study design was a single-blind whole-group controlled trial. A total of 136 eligible participants were divided according to their nursing home into either an experimental or control group. The nursing home grouping was determined by the drawing of lots. The Montreal Cognitive Assessment (MoCA), Quality of Life (SF-12) and Geriatric Depression Scale (GDS-15) were used to assess participants at baseline, week 9, and week 18, respectively. Generalized estimating equations (GEE) were used to compare the results at baseline with mid-test and post-test changes in cognitive function and quality of life. Maximum likelihood estimation (ML) and robust standard errors were used to perform the mediation model. The study results indicated that the experimental group (compared to the control group) had a significant improvement in cognitive function, quality of life, and mood state at baseline in the mid-test and post-test results. The results of this 18-week experiment showed that the exercise–cognition relationship was significantly mediated by a reduction in depressive symptoms (indirect effect: β = −0.375; 95% CI = −0.864 to −0.069) and an improvement in quality of life (indirect effect: β = −0.678; 95% CI = −1.222 to −0.290). This study revealed the effects of moderate-intensity square dance exercise on cognitive function and quality of life in older Chinese women with mild cognitive impairment and explored the potential mediating mechanisms. These findings can be used to inform the development of public health policies to promote brain health in older adults with mild cognitive impairment.

Objectives: We previously reported the therapeutic potential of Bifidobacterium breve A1 (B. breve A1) for preventing cognitive impairment in Alzheimer’s disease model mice, which suggested that supplementation of the probiotics could be an effective therapeutic strategy for managing cognitive function in mild cognitive impairment (MCI). Design and settings: We conducted an open-label, single-arm study to examine the effects of 24-week supplementation of B. breve A1 on elderly with MCI in Aki Orthopedics Rehabilitation Clinic in Japan. Participants: 27 participants were screened by their Mini Mental State Examination (MMSE) scores. Measurements: Cognitive function was assessed using MMSE and Digit Symbol Substitution Test (DSST) at baseline and every 8 weeks. Mental condition and quality of life for gastrointestinal symptoms were measured using the Profile of Mood States 2nd Edition (POMS2), and the Gastrointestinal Symptom Rating Scale (GSRS). Results: Of the 27 participants enrolled, 19 completed the study. MMSE scores were significantly increased during the intervention by mixed model Dunnett’s test and Wilcoxon signed-rank tests (+1.7, P < 0.01). POMS2 and GSRS scores were significantly improved during intervention when analyzed by Wilcoxon signed-rank tests. Conclusion: The present study showed that oral supplementation of B. breve A1 in participants with MCI improved cognitive function, thus suggesting the potential of B. breve A1 for improving cognitive function and maintaining quality of life of the elderly. Further randomized, double-blind placebo-controlled studies are worth conducting to examine the beneficial effect of B. breve A1.

Depressive symptoms may increase the risk of progressing from mild cognitive impairment (MCI) to dementia. Consumption of n-3 PUFA may alleviate both cognitive decline and depression. The aim of the present study was to investigate the benefits of supplementing a diet with n-3 PUFA, DHA and EPA, for depressive symptoms, quality of life (QOL) and cognition in elderly people with MCI. We conducted a 6-month double-blind, randomised controlled trial. A total of fifty people aged >65 years with MCI were allocated to receive a supplement rich in EPA (1·67 g EPA+0·16 g DHA/d; n 17), DHA (1·55 g DHA+0·40 g EPA/d; n 18) or the n-6 PUFA linoleic acid (LA; 2·2 g/d; n 15). Treatment allocation was by minimisation based on age, sex and depressive symptoms (Geriatric Depression Scale, GDS). Physiological and cognitive assessments, questionnaires and fatty acid composition of erythrocytes were obtained at baseline and 6 months (completers: n 40; EPA n 13, DHA n 16, LA n 11). Compared with the LA group, GDS scores improved in the EPA (P = 0·04) and DHA (P = 0·01) groups and verbal fluency (Initial Letter Fluency) in the DHA group (P = 0·04). Improved GDS scores were correlated with increased DHA plus EPA (r 0·39, P = 0·02). Improved self-reported physical health was associated with increased DHA. There were no treatment effects on other cognitive or QOL parameters. Increased intakes of DHA and EPA benefited mental health in older people with MCI. Increasing n-3 PUFA intakes may reduce depressive symptoms and the risk of progressing to dementia. This needs to be investigated in larger, depressed samples with MCI.

Objectives: To examine the mediating effects of depression and anxiety in the relationship between self-efficacy and quality of life among people with mild cognitive impairment (MCI) or mild dementia. Method: A total of 196 patients diagnosed with MCI or dementia due to Alzheimer disease completed structured measures of self-efficacy, quality of life, and depressive and anxiety symptoms. We examined direct and mediated effects by fitting structural equation models to data. Results: Our analyses supported that the effects of self-efficacy on quality of life may be partially mediated by depression and anxiety. Both anxiety and depression had significant mediating effects, with depression showing a stronger effect. Conclusion: These results suggest that increased self-efficacy may have a positive effect on quality of life in people with MCI or dementia—partly by reducing depression and anxiety. These findings may have important practical implications for tailoring therapeutic interventions.
