The effect of feed deprivation on gastrointestinal respiration and Na+, K+-ATPase in chicks

In order to investigate the effect of nutrient availability on energy metabolism of the gastrointestinal tract (GIT) of chicks, two experiments were conducted. In exp. 1, jejunal slices from male White Leghorn chicks at 14 d were incubated in salt-based balanced medium (SBM) or complex medium (CM) for 30 min. The total O2 (TO2) and ouabain-sensitive O2 (OSO2) consumption in jejunal slices were significantly decreased in SBM. The results from exp. 1 imply that when the GIT mucosa is subjected to nutrient deprivation, the utilization of oxygen consumption is altered primarily by shifting active Na+ pump activity. Changes in the TO2, OSO2 and amiloride-sensitive O2 (ASO2) uptake in response to feed deprivation in the small intestine of male White Leghorn chicks were investigated in exp. 2. There was little change in TO2, OSO2 and ASO2 per unit of dry matter of the small intestine from birds subjected to fasting, except for a TO2 decrease in the jejunum of 48 h fasted chicks. However, there were significant reductions in TO2, OSO2 and ASO2 in the jejunum when data were expressed per unit DNA. Na+, K+-ATPase-dependent respiration represented 31–37% of TO2 in the avian jejunoileum. Amiloride-sensitive respiration accounted for 10–18% of TO2 in the jejunoileum. The maximal amount of 3Houabain binding sites of jejunal enterocytes was significantly decreased in chicks fasted for 48 h (P < 0.05). The purpose of this study was to assess the reliability and validity of a portable metabolic system (TEEM 100) during submaximal and maximal [Formula: see text] exercise using a computer-based metabolic system as the reference system (REF). Between repeated trials of submaximal exercise at three constant loads, differences in ventilation [Formula: see text] and oxygen consumption [Formula: see text] were 0.2 ± 4.9 L∙min−1 and 0.03 ± 0.10 L∙min−1 for REF, and 1.9 ± 0.7 L∙min−1 and 0.00 ± 0.17 L∙min−1 for TEEM 100. Pooled intraclass reliability coefficients for [Formula: see text] and [Formula: see text] calculated from the repeated submaximal trials were r =.89 and r =.94 for REF, and r =.86 and r =.94 for the TEEM 100. Respiratory exchange ratio (RER) measured by the TEEM 100 was significantly higher (p =.01) at only the lowest workload. At [Formula: see text], the TEEM 100 recorded significantly higher values for FeO2 (p =.01) and RER (p <.001). These results suggest that the TEEM 100 provides reliable and valid measurements of [Formula: see text] during submaximal and maximal exercise. Four castrated pigs, each fitted with a re-entrant cannula in the terminal ileum, were used to study digestion in the small intestine. A nitrogen-free diet was used for the estimation of metabolic nitrogen and amino acids. Faba beans, as the sole source of dietary protein, were used raw or after autoclaving for 30 or 60 min. The four diets were fed to the pigs in a 4 × 4 latin square experiment. The pigs were fed each diet for 6 days prior to a 24-hr collection of total ileal contents. Autoclaving of faba beans had no significant effect on digestibility of dry matter, gross energy, nitrogen and individual amino acids except arginine, which was significantly increased. The intestinal uptake of arginine was the highest and of cystine the lowest in all faba bean diets. It was concluded that autoclaving faba beans had no beneficial effect on the digestion of nutrients in the small intestine of the pig.

1. In a 4 x 4 Latin square experiment four cows were given, twice daily, diets consisting of (g/kg dry matter (DM)) 500 barley, 400 grass silage and 100 soya-bean meal. The diets were given at either 1.15 (L) or 2.3 (H) times maintenance energy requirements and the soya-bean meal was either untreated (U) or formaldehyde (HCH0)-treated (T).2. The passage of digesta to the duodenum was estimated using chromic oxide as a flow marker;35S was used to estimate the amount of microbial protein entering the small intestine. A microbial fraction was prepared by differential centrifugation from duodenal digesta. Samples of bacteria and of protozoa from rumen digesta were also prepared.3. The total amino acid contents of feedingstuffs, duodenal digesta, duodenal microbial material, rumen bacteria and rumen protozoa were determined by ion-exchange chromatography. The D-alanine and D-glutamic acid contents of the samples were determined by gas–liquid chromatography.4. The quantity of each amino acid entering the small intestine was significantly (P < 0,001) increased by increasing DM intake and tended to be increased by formaldehyde-treatment of the soya-bean meal. There were net losses of all amino acids across the forestomachs except for lysine, methione, o-alanine and D-glutamic acid for which there were net gains.5. There were significant (P < 0.05) differences in amino acid composition between rumen bacteria and duodenal microbial material; differences in amino acid composition between rumen bacteria and rumen protozoa were also observed.6. D-Alanine and D-glutamic acid were present in the silage but not in the barley or either of the soya-bean meals. All samples of microbes and digesta contained D-alanine and D-glutamic acid.7. The use of D-ahine and D-glUtamiC acid as markers for microbial nitrogen entering the small intestine was assessed. Estimates of the quantities of microbial N entering the small intestine based on the D-alanine or D-glutamic acid contents of rumen bacteria or duodenal microbes were significantly higher than those determined using 35S as a marker.

1. The oxygen consumption rates of 3rd- and 4th-instar larvae of Chironomus riparius have been measured at 10 and 20° C. using a constant-volume respirometer. 2. The oxygen consumption is approximately proportional to the 0.7 power of the dry weight: it is not proportional to the estimated surface area. 3. This relationship between oxygen consumption and dry weight is the same at 10 and at 20° C.. 4. The rate of oxygen consumption at 20° C. is greater than at 10° C. by a factor of 2.6. 5. During growth the percentage of dry matter of 4th-instar larvae increases from 10 to 16 and the specific gravity from 1.030 to 1.043. 6. The change in the dry weight/wet weight ratio during the 4 larval instar supports the theory of heterauxesis. 7. At 20° C., ‘summer’ larvae respire faster than ‘winter’ larvae.

Four treatments, involving differences in method of feeding and interval between last feed and slaughter, were employed in a study of the effect of dietary chlortetracycline on protease, amylase, and cellulase activity in the intestinal and cecal contents of 16 pairs of weanling pigs. Most consistent results were obtained with six pairs which were limited pair-fed except for the final feeding during which feed was available ad libitum for a 4-hour period ending 18 hours before slaughter. The mean total activities of all three hydrolases in the contents of the small intestines and of the ceca of the antibiotic-fed animals of these six pairs were significantly greater than in those of the control animals. Expressed as activity per gram dry matter of intestinal contents, significantly higher values for protease and amylase, but not for cellulase, were observed in the pigs that had received chlortetracycline. When the combined results obtained from all 16 pairs of the experimental animals were analyzed, the results showed that on a basis of activity per gram dry matter of intestinal contents, ingested chlortetracycline was associated with significant increases in amylase and cellulase but not in protease activity. Protease, amylase, and cellulase activities per gram dry matter of cecal contents were higher for pigs fed the antibiotic than for their controls. The mean wet weight of the empty small intestine and the mean dry weight of the mucosa scraped from the anterior 3-meter section of the small intestine were lower for the chlortetracycline-fed animals, but the differences were not statistically significant.

In a study with ten pigs of 60–70 kg live weight, provided with a re-entrant cannula at the end of the ileum, and sixteen intact, non-cannulated pigs, the digestion and absorption of a dietary dose of 100 g isomalt/kg, and isomalt given between the meals as a ‘sweet’on the basis of 50 and 100 g/kg feed consumption, were examined. In all three isomalt treatments slightly less than 0.40 of the isomalt consumed was digested in the small intestine when the calculations were based on ileal sugar passage. However, when basing the calculations on energy contents of ileal chyme, only approximately 0.10 was digested in the small intestine. The bacterial fermentation of the isomalt flowing into the large intestine was indicated by a decreased faecal energy digestibility and a slight reduction in faecal dry matter and nitrogen digestibility. The retention of the minerals sodium, potassium, magnesium, calcium and phosphorus was not influenced to any measurable extent when isomalt was fed.

Immune-mediated scouring in sheep is a mucosal hypersensitivity response incited by infective larvae of gastrointestinal nematodes in the southern half of Australia and in New Zealand, regions characterised by a Mediterranean, winter-rainfall climate. To gain insights into the underlying mechanism(s), this study investigated the relationship between nematode infection of sheep in pens, faecal dry matter and release of inflammatory mediators in the intestinal tract. Six worm-free, immune rams received a dose of 3000 Trichostrongylus third stage larvae (L3) and 3000 Ostertagia L3 per week for 6 weeks, after which they were euthanased. Three rams acted as uninfected controls. Faecal dry matter was significantly lower in the infected group from the first week of infection until the end of the trial. Concentrations of cysteinyl leukotrienes and prostaglandin E2 were higher in mucus from both the abomasum and the small intestine of infected rams compared with controls. Within the infected group, concentrations of both mediators were higher in mucus from the small intestine than from the abomasum. There were greater numbers of eosinophilic granulocytes and mucosal mast cells in abomasal and small-intestinal tissue sections from infected rams compared with the controls. The results are consistent with the hypothesis that nematode infection causes an inflammatory response in the gastrointestinal mucosa, characterised by increased fluid secretion.

1. In two experiments, sheep prepared with a rumen cannula and with re-entrant cannulas in the duodenum and ileum were continuously fed on diets of dried grass, dried grass plus formalin-treated casein, or dried grass plus untreated casein. Paper impregnated with chromic oxide was given once daily via the rumen fistula.2. In ten 24 h collections of digesta entering the duodenum and eleven 24 h collections of digesta reaching the ileum of sheep given dried grass, there were highly significant correlations between the 24 h flows of Cr marker and the corresponding flows of dry matter, organic matter, nitrogen, gross energy, hemicellulose and cellulose (P < 0.01) at both sites.3. Daily amounts of non-ammonia N and of individual amino acids entering and leaving the small intestine and of total N excreted in faeces and urine are given.4. Net retention of supplementary N was 36% when the supplement was administered as formalin-treated casein, but only 17% when it was administered as untreated casein.5. Formalin treatment of casein significantly increased the daily amounts of non-ammonia N entering the small intestine (P < 0.01) and the amounts of non-ammonia N apparently absorbed therein (P < 0.05).6. Apparent absorption of amino acids from the small intestine was significantly greater (P < 0.05) with treated casein than with untreated casein. There were relative increases in the small amounts of several free amino acids measured, including taurine, in the ileal digesta of sheep receiving the treated casein supplement.

Distribution of the binding sites for [3H]diprenorphine, a non-selective opiate ligand, was studied in membrane fractions from longitudinal muscle/myenteric plexus and circular muscle containing deep muscular plexus. [3H]saxitoxin was used as a marker for neuronal plasma membranes and 5'-nucleotidase as a marker for smooth muscle plasma membranes. Saxitoxin binding correlated strongly with diprenorphine binding, but 5'-nucleotidase correlated poorly with diprenorphine or saxitoxin binding in these fractions. Opiate binding sites in membranes of myenteric and deep muscular plexus were of high affinity (Kd = 0.12 and 0.18 nM, respectively) with maximum binding capacity of 400 and 500 fmol/mg protein, respectively. Competition experiments using subtype-selective opiate ligands indicated that all three subtypes of opiate receptors were present in the same ratio of 40-45% mu-subtypes, 40-45% delta-subtypes, and 10-15% kappa-subtypes on both plexuses. Opiate receptors of canine small intestine, therefore, are located primarily or exclusively on nerves with similar distributions in nerve membranes containing only axonal varicosities (deep muscular plexus) as in those containing neurons, dendrites, and varicosities (myenteric plexus).
