Complex social structure in eusocial insects can involve worker morphological and behavioural differentiation. Neuroanatomical variation may underscore worker division of labour, but the regulatory mechanisms of size-based task specialization in polymorphic species are unknown. The Australian weaver ant, Oecophylla smaragdina , exhibits worker polyphenism: larger major workers aggressively defend arboreal territories, whereas smaller minors nurse brood. Here, we demonstrate that octopamine (OA) modulates worker size-related aggression in O. smaragdina . We found that the brains of majors had significantly higher titres of OA than those of minors and that OA was positively and specifically correlated with the frequency of aggressive responses to non-nestmates, a key component of territorial defence. Pharmacological manipulations that effectively switched OA action in major and minor worker brains reversed levels of aggression characteristic of each worker size class. Results suggest that altering OA action is sufficient to produce differences in aggression characteristic of size-related social roles. Neuromodulators therefore may generate variation in responsiveness to task-related stimuli associated with worker size differentiation and collateral behavioural specializations, a significant component of division of labour in complex social systems.

Background: The chick chorioallantoic membrane (CAM) model has attracted a great deal of interest in pharmaceutical and biological research as an alternative or complementary in vivo assay to animal models. Traditionally, CAM assay has been widely used to perform some toxicological studies, specifically to evaluate the skin, ocular and embryo toxicity of new drugs and formulations, and perform angiogenesis studies. Due to the possibility to generate the tumors onto the CAM, this model has also become an excellent strategy to evaluate the metastatic potential of different tumours and test the efficacy of novel anticancer therapies in vivo. Moreover, in the recent years, its use has considerably grown in other research areas, including the evaluation of new anti-infective agents, the development of biodistribution studies and tissue engineering research. Objectives: This manuscript provides a critical overview of the use of CAM model in pharmaceutical and biological research, especially to test the toxicity of new drugs and formulations and the biodistribution and the efficacy of novel anticancer and anti-infective therapies, analyzing its advantages and disadvantages compared to animal models. Conclusion: The chick chorioallantoic membrane model shows great utility in several research areas, such as cancer, toxicology, biodistribution studies and anti-infective therapies. In fact, it has become an intermediate stage between in vitro experiments and animal studies, and, in the case of toxicological studies (skin and ocular toxicity), has even replaced the animal models.

Elevated amounts of copper are considered to be contributing factor in the progression of neurodegenerative diseases as they promote oxidative stress conditions. The aim of our study was to examine the effects of ethanolic extract of propolis (EEP) against copper-induced neuronal damage. In cultured P19 neuronal cells, EEP exacerbated copper-provoked neuronal cell death by increasing the generation of reactive oxygen species (ROS) and through the activation of caspase-3/7 activity. EEP augmented copper-induced up-regulation of p53 and Bax mRNA expressions. Neurotoxic effects of EEP were accompanied by a strong induction of glyceraldehyde 3-phosphate dehydrogenase (GAPDH) expression and decrease in the expression of c-fos mRNA. SB203580, an inhibitor of p38 mitogen-activated protein kinase (MAPK) prevented detrimental effects of EEP, whereas SP600125, an inhibitor of c-Jun N-terminal kinase (JNK), exacerbated EEP-induced neuronal cell death. Quercetin, a polyphenolic nutraceutical, which is usually present in propolis, was also able to exacerbate copper-induced neuronal death. Our data indicates a pro-oxidative and apoptotic mode of EEP action in the presence of excess copper, wherein ROS/p53/p38 interactions play an important role in death cascades. Our study also pointed out that detailed pharmacological and toxicological studies must be carried out for propolis and other dietary supplements in order to fully recognize the potential adverse effects in specific conditions.

 Objective: This work assessed the antimicrobial activity and toxicological consequence of the ethanolic extract of Ricinodendron heudelotii (Baill).Methods: The antimicrobial potential of the leaf extract was investigated against 9 different microorganisms using the agar well diffusion method while the minimum inhibitory concentration (MIC) was ascertained. Toxicity evaluations were carried out on five groups of seven albino rats each for 28 days. Rats in Group A received normal saline, while those in Groups B, C, D, and E received 250, 500, 1000, and 2000 mg/kg body weight (b.w.) of the extract. After treatment, they were anesthetized, blood samples collected for biochemical assays; organs isolated, weighed, and processed for histopathological studies.Results: The phytochemical screening showed the presence of glycosides, tannins, polyphenols, terpenoids, and alkaloids while saponin was absent. Antimicrobial effect was observed against 7 of the 9 microorganisms tested with inhibition zone and MIC ranging from 18 to 36 mm and 31.25 to 62.5 mg/mL, respectively. Liver function tests were significantly (p<0.05) increased, whereas no significant change in total protein and creatinine levels was observed. Histology showed a sinusoidal congestion and vacuolar degeneration of hepatocytes in groups administered higher doses, and a dose-dependent reduction was observed in some hematological parameters.Conclusion: The results obtained suggest that the R. heudelotii may not induce significant toxic effect below 3600 mg/kg b.w. and could serve as an effective antibiotic.

Abstract Annona vepretorum is endemic from the Brazilian Caatinga biome and is used in human nutrition. The present study aimed to investigate the toxic effects of the ethanolic extract from the leaves of this species. The leaves of A. vepretorum were collected, dried, pulverized, and macerated with ethanol to yield the crude ethanol extract of A. vepretorum. HPLC-diode array detection was used to determine the fingerprint chromatogram of the extract. In toxicity studies, the acute toxicity experimental group was administered a single dose of the ethanol extract of A. vepretorum (1 g/kg), while in the subacute toxicity experimental group, the ethanol extract of A. vepretorum was administered orally, daily for 30 days, at doses of 100 and 400 mg/kg. Death and signs of toxicity were observed and at the end, the animals were anesthetized, and blood and organs were then collected. The presence of the flavonoid rutin in the extract was confirmed using HPLC-diode array detection. In the evaluation of acute and subacute toxicity, there were no behavioral and physiological changes or signs of toxicity, and no occurrences of mice deaths were registered. The organs had normal color and preserved architecture, and no statistical variations in weight were observed. The results of the hematological and biochemical parameters after the administration of the ethanol extract of A. vepretorum showed no significant change, except in the count of the number of leukocytes and triglycerides. The histopathologic analysis of the liver, kidneys, and stomach indicated architecture with normal aspects. Thus, the toxicity study indicates low toxicity of the ethanol extract of A. vepretorum. Such information will be helpful in future clinical studies.
