A study of average flow in open channel with baffle blocks distributed uniformly has been considered by using channel with varied slopes. In this article, experimental and modelling studies were introduced when the correlation between the water depth and baffle block size is significant. The objective of the work is to give the rudimentary relations between discharge and water level in the channels. When the water depth is large, the effect of bottom channel friction on the flow is relatively small. This paper also gives applications of the software ‘Telemac-2D’ to simulate the flow under different conditions.

Background:: Indole and pyrazoles are one of the prime structural units in the field of medicinal chemistry and have been reported to exhibit a variety of biological activities specifically anti-cancer. In view of their medicinal significance, we synthesized a conjugate of the two moieties to get access to newer and potential anti-cancer agents. Methods: Indolyl pyrazoles [3-(1,3-diphenyl-1H-pyrazol-4-yl)-2-(1-methyl-1H-indole-3-carbon yl)acrylonitriles] (4a-l) were synthesized by adopting simple and greener protocol and all the synthesized derivatives were docked against Bcl-2 protein and the selected chemical moieties were screened for their cytotoxicity by using the MTT assay. Results: : All the synthesized compounds were docked against BCL-2 protein in order to understand their binding pattern. Among the 12 compounds docked, 4d, 4f, 4h, 4j, and 4l compounds exhibited better protein binding interactions and the same were screened for their anti-cancer activity against A549 (lung) cancer cell lines at a concentration of 100 μM using Doxorubicin as standard. Substitutions such as N-benzyl, N-ethyl groups and halogen groups such as Br, Cl on indole ring showed moderate activity against A-549 cell lines. Conclusion:: Among the 5 indolyl pyrazole derivatives screened, compounds 4h and 4j showed significantly better activity with an IC50 of 33.12 and 34.24 μM, respectively. Further, structural tweaking of the synthesized new chemical entities may lead to potential hit/lead-like molecules.

Abstract. Scotty Creek, Northwest Territories (NWT), Canada, has been the focus of hydrological research for nearly three decades. Over this period, field and modelling studies have generated new insights into the thermal and physical mechanisms governing the flux and storage of water in the wetland-dominated regions of discontinuous permafrost that characterises much of the Canadian and circumpolar subarctic. Research at Scotty Creek has coincided with a period of unprecedented climate warming, permafrost thaw, and resulting land cover transformations including the expansion of wetland areas and loss of forests. This paper (1) synthesises field and modelling studies at Scotty Creek, (2) highlights the key insights of these studies on the major water flux and storage processes operating within and between the major land cover types, and (3) provides insights into the rate and pattern of the permafrost-thaw-induced land cover change and how such changes will affect the hydrology and water resources of the study region.

Bayesian methods are advantageous for biological modelling studies due to their ability to quantify and characterize posterior variability in model parameters. When Bayesian methods cannot be applied, due either to non-determinism in the model or limitations on system observability, approximate Bayesian computation (ABC) methods can be used to similar effect, despite producing inflated estimates of the true posterior variance. Owing to generally differing application domains, there are few studies comparing Bayesian and ABC methods, and thus there is little understanding of the properties and magnitude of this uncertainty inflation. To address this problem, we present two popular strategies for ABC sampling that we have adapted to perform exact Bayesian inference, and compare them on several model problems. We find that one sampler was impractical for exact inference due to its sensitivity to a key normalizing constant, and additionally highlight sensitivities of both samplers to various algorithmic parameters and model conditions. We conclude with a study of the O'Hara–Rudy cardiac action potential model to quantify the uncertainty amplification resulting from employing ABC using a set of clinically relevant biomarkers. We hope that this work serves to guide the implementation and comparative assessment of Bayesian and ABC sampling techniques in biological models.
