New first derivative ratio spectrophotometric method was developed for simultaneous estimation of telmisartan hydrochloride (TELM) and hydrochlorothiazide (HCTZ) in combined tablet dosage form without prior separation. The amplitude in the first derivative of the corresponding ratio spectra at 263.2 nm and 278.4 nm of TELM and for HCTZ at 287.2 nm and 304.8 nm were selected for the determination. The calibration graphs were established in the range of 1-10μg/mL and 2-15 μg/mL of TELM and HCTZ, respectively. The percentage recoveries of binary mixture of TELM were found to be 97.81±1.05 (263.2nm) and 96.60±1.91 (278.4nm) and for HCTZ were found to be 97.47±1.41 (287.2), 98.16± 1.45 (304.8nm) by proposed method. This method was successfully applied for the analysis of commercial tablet formulation, with no interference from excipients as indicated by the recovery study results. The proposed method was validated, statistically analyzed, compared with reported HPLC method and can be successfully applied for the routine analysis.

The spectrophotometric method for estimation of CefpodoximeProxetil employed first derivative amplitude UV spectrophotometric method for analysis using methanol as solvent for the drug. CefpodoximeProxetil has absorbance maxima at 235nm and obeys Beer’s law in concentration range 10-50µg/ml with good linearity i.e. r2 about 0.999. The recovery studies established accuracy of the proposed method; result validated according to ICH guideline. Results were found satisfactory and reproducible. The method was successfully for evaluation of CefpodoximeProxetil in tablet dosage form without interference of common excipients.

<p align="center"><strong>Abstrak</strong><strong></strong></p><p align="center"><strong> </strong></p><p>Metode kromatografi cair kinerja tinggi (KCKT) dengan detektor ultra lembayung telah dikembangkan dan divalidasi untuk estimasi kadar secara simultan campuran piridoksin hidroklorida (PH) dan piratiazin teoklat (PT)dalam sediaan tablet antiemetik. Proses pemisahan terjadi dalam kolom Inertsil® ODS-3 pada panjang gelombang 280 nm dengan laju alir 1,0 mL/menit. Fase gerak yang optimal untuk pemisahan adalah campuran methanol-asam asetat 1% (20:80) dengan waktu retensi PH dan PT berturut-turut adalah 1,2 dan 9,8 menit. Perolehan kembali PH dan PT berturut-turut adalah 100,13 dan 99,78 %. Batas deteksi untuk PH dan PT berturut-turut adalah 0,21 dan 0,22 µg/mL, sedangkan batas kuantisasinya berturut-turut adalah 0,70 dan  0,72 µg/mL. The separation process occurs in the Inertsil® ODS-3 column at a wavelength of 280 nm with a flow rate of 1.0 mL /min. The optimal mobile phase for separation is a mixture of methanol-acetic acid 1% (20:80) with the retention times of PH and PT 1.2 and 9.8 minutes respectively. The recoveries of PH and PT were 100.13 and 99.78%, respectively. The detection limits for PH and PT were 0.21 and 0.22 µg / mL respectively, while the quantisation limits were 0.70 and 0.72 µg / mL, respectively. A simple, rapid, sensitive and economical UV spectrophotometric method was developed and validated for the simultaneous estimation of aspirin, caffeine, and orphenadrine citrate by Vierodt’s method in combined tablet dosage form and using mixture of 0.1 N HCl: ethanol (1:1) as solvent. The method employs designing and solving of simultaneous equations using 3 wavelengths, namely 226 nm, 272 nm, and 210 nm. Beer’s law was obeyed in the concentration range of 3-18 μg/mL for aspirin and 0.5-12 μg/mL for both caffeine and orphenadrine citrate with r2 value of 0.993, 0.991, and 0.991 respectively. Accuracy was determined by recovery studies and showed percent recovery ranging from 99.78-100% for aspirin, 96.10-100.92% for caffeine and 100.53-102.5% for orphenadrine citrate. The developed method can be used in QC laboratory for routine analysis to ensure the identity, purity, and performance of the drug product.

A novel, simple, rapid, and highly selective stability indicating RP-HPLC method was developed and validated for simultaneous estimation of azilsartan medoxomil (AZL) and amlodipine besylate HCl (AMLO) in tablet dosage form having strength of 20 mg and 2.5 mg, respectively. The effective chromatographic separation was achieved on a Phenomenex luna ODS C18 (15 cm X 4.6 mm internal diameter, 3.5 μm Particle size) with a mobile phase composed of phosphate buffer (pH-2.5) adjusted with ortho phosphoric acid : acetonitrile in the ratio of 60:40 v/v. The mobile phase was pumped using an isocratic HPLC system at a flow rate of 0.7 mL/min with injection volume 20μl and quantification of the analytes was done at detection wavelength 254 nm. The retention times were found to be 5.918 min and 14.901 min for AMLO and AZL, respectively. The proposed HPLC method was validated with respect to linearity, ranges, precision, accuracy, specificity, robustness, LOD, and LOQ as per ICH Q2 (R1) guideline. Calibration plots were linear over the concentration range of 75-125 µg/mL and 600-1000 µg/mL with correlation coefficients 0.9966 and 0.9948 for AMLO and AZL, respectively. Forced degradation studies were performed using hydrolysis, oxidation, photolytic, and thermal degradation conditions with good resolution between the degradants and analytes. Degradation products resulting from the stress studies did not interfere with the detection of AMLO and AZL, thus the proposed method is sensitive and stability-indicating. The validated HPLC method was successfully applied to the analysis of AMLO and AZL in tablet dosage form.
