Flaxseed oil cake (FOC) was evaluated as a potential substrate for the production of a novel kefir-like fermented beverage. Three variants containing 5%, 10%, and 15% (w/w) of FOC were inoculated with kefir grains and incubated at 25 °C for 24 h. After processing, beverages were stored in refrigerated conditions (6 °C) for 21 days. Changes in microbial population, pH, acidity, levels of proteins, polyphenolics, flavonoids, ascorbic acid, and reducing sugars were estimated. Additionally, viscosity, firmness, color, and antioxidant properties were determined. Results showed that lactic acid bacteria as well as yeast were capable of growing well in the FOC without any supplementation. During refrigerated storage, the viability of the microorganisms were over the recommended minimum level for kefir products. As a result of fermentation, the beverages showed excellent antioxidant activity. Because of the functional characteristics conferred to the FOC beverages, the use of kefir grains showed adequate potential for the industrial application. Therefore, this beverages could be used as a new, non-dairy vehicle for beneficial microflora consumption, especially by vegans and lactose-intolerant consumers.

The phytochemicals of the ethanolic extracts of petals and stamens of lotus flowers from several lotus cultivars belonging to the Nelumbo nucifera Gaertn. (namely 'lotus white flower' and 'lotus pink flower') were studied by color reaction test and HPTLC analysis. The lotus pink flower stamens showed a highest amount of ethanolic extractives (ca. 18.33±2.14% of dry material weight). The several the ethanolic extracts of N. nucifera petals and stamens showed similar qualitative phytochemicals. The crude extracts revealed the presence of flavonoids, terpenoinds, tannins and polyphenols. The antioxidant activity was assessed using the 2,2-diphenyl-1-picrylhydrazyl assay. The analyzed lotus pink flower stamens ethanolic extract showed a highest antioxidant activity at IC50 = 0.61±0.16 μg/ml and lotus pink flower petals ethanolic extract showed a highest tyrosinase inhibitory effect at IC50 = 2.25±0.21 μg/ml. N. nucifera flowers could be provide a potential natural source of phytochemicals and could be beneficial to the cosmetic properties.

Objective: The main objective of the study was to investigate the effect of ethanol seed extracts of Nelumbo nucifera Gaertn (Nymphaeaceae) as antihyperglycemic, anti-hyperlipidemic and antioxidant activity in streptozotocin-induced diabetic rats.Methods: Diabetes was induced in wistar albino rats by administration of streptozotocin (single intraperitoneal dose of 60 mg/kg B. W). The ethanol extract of N. nucifera seed at a dose of 200 and 400 mg/kg body weight was administrated at a single dose per day to STZ induced diabetic rats for a period of 42 d. The different pharmacological parameters were evaluated. The effect of ethanol seed extract of N. nucifera on insulin, blood glucose, urea, creatinine, HbA1C, serum protein, albumin, globulin, serum enzymes, serum lipid profiles, lipid peroxides (LPO) and other antioxidant enzymes like catalase (CAT), glutathione peroxidase (GPx), superoxide dismutase (SOD) and reduced glutathione (GSH) were measured in the diabetic rats.Results: In the acute toxicity study ethanol seed extract of Nelumbo nucifera were non-toxic at 2000 mg/kg in rats. The ethanol extract of Nelumbo nucifera seed showed significant reduction in blood glucose (p<0.05), serum enzymes (SGPT, SGOT, ALP) (p<0.05), lipid parameters (TC, TG, VLDL, LDL) (p<0.05) except HDL and significantly increased insulin (p<0.05), HDL (p<0.05), GPx, GSH, SOD and CAT (p<0.05) at the dose of 400 mg/kg when compared with the diabetic-induced control.Conclusion: The present study suggested that the NNSE has significant (p<0.05) antihyperglycemic, antihyperlipidemic and antioxidant activity in STZ induced diabetic rats. These results clearly indicate that Nelumbo nucifera is effective against free radical-mediated diseases, thus replacing the synthetic ones.

Background: Microorganisms from understudied habitats have been shown to be an important source of novel bioactive compounds. Endophytes constitute an underexplored group of microorganisms, of which those from aquatic plants have been even less studied. Nelumbo nucifera (lotus) is an aquatic plant with medicinal properties. A screening program for endophytes from N. nucifera by our research group resulted in many microbial isolates, of which isolate L-003 was a promising candidate, exhibiting antimicrobial and antioxidant activities. Objectives: The major objectives were to characterize the endophyte L-003 for its antimicrobial and antioxidant properties, identify the constituent bioactive compounds by GC-MS and characterize their activities further using in silico software. Methods: L-003 was identified by PIBWin software. Antimicrobial activity of the aqueous and organic extracts of culture supernatant of L-003 was checked against a panel of bacteria and fungi. Since the ethyl acetate extract showed the best antimicrobial activity, it was further characterized by thin layer chromatography, an activity confirmed by bioautography and purified by column chromatography. Total antioxidant capacity was assayed by standard techniques. Partially purified metabolite fingerprints were identified by GC-MS analysis. Results: Based on morphological and biochemical analyses, isolate L-003 was identified as belonging to Streptococcus sp. All the organic solvent extracts showed antimicrobial activity. Ethyl acetate extract showed maximum antimicrobial activity against all selected targets and exhibited antioxidant activity too, though butanol and aqueous extracts showed higher antioxidant activity. Two compounds, Acetic acid,-hydroxy, methyl ester and Disulfide, dipropyl, were identified by GC-MS in the metabolite fingerprint. These compounds showed differences in observed and calculated retention indices and could, therefore, be novel. In silico activity, characterization confirmed the antimicrobial and antioxidant properties attributed to these compounds. Conclusion: This is the first study reporting metabolite fingerprinting, identification and characterization of bioactive compounds from an endophytic isolate of Nelumbo nucifera. We conclude that endophytes from aquatic plants could be prospective sources of bioactive compounds, in this case with antimicrobial and antioxidant activities.
