G protein-coupled receptors (GPCRs) are associated with a great variety of biological activities. Yeasts are often utilized as a host for heterologous GPCR assay. We engineered the intense reporter plasmids for fission yeast to produce green fluorescent protein (GFP) through its endogenous GPCR pathway. As a control region of GFP expression on the reporter plasmid, we focused on seven endogenous genes specifically activated through the pathway. When upstream regions of these genes were used as an inducible promoter in combination with LPI terminator, themam2upstream region produced GFP most rapidly and intensely despite the high background. Subsequently, LPI terminator was replaced with the corresponding downstream regions. The SPBC4.01 downstream region enhanced the response with the low background. Furthermore, combining SPBC4.01 downstream region with the sxa2 upstream region, the signal to noise ratio was obviously better than those of other regions. We also evaluated the time- and dose-dependent GFP productions of the strains transformed with the reporter plasmids. Finally, we exhibited a model of simplified GPCR assay with the reporter plasmid by expressing endogenous GPCR under the control of the foreign promoter.

Take a typical cross-section of rockfill embankment slope in Yaan-Luku highway as the research object, reliability analysis is studied under the condition of water table of 840.85m, 851.50m, and loading condition of natural state and horizontal seismic acceleration of 0.2g, respectively. Raw data use Kolmogorov-Smirnov test (K-S test) to determine the distribution type of parametric variation. And the parameters were sampling with Latin hypercube sampling (LHS) method and Monte Carlo (MC) method, respectively, to obtain state function and determine safety factors and reliability indexes. A conclusion is drawn that the times of simulation based on LHS method were less than Monte Carlo method. Also the convergence of failure probability is better than the Monte Carlo method. The safety factor is greater than one and the failure probability has reached to 35.45% in condition of earthquake, which indicating that the instability of rockfill embankment slope is still possible.

Pollen productivity еstimate (PPE) and relevant source area of pollen (RSAP) are critical parameters for quantitative interpretations of pollen data in palaeolandscape and palaeoecological reconstructions, and for analyses of the landscapes evolution and anthropogenisation as well. In light of this, the present paper endeavours to calculate PPE of key plant taxa and to define the RSAP in the Kamchia River Downstream Region (Eastern Bulgaria) in order to use them in landscape simulations and estimations. For the purposes of this research, a dataset of pollen counts from 10 modern pollen samples together with corresponding vegetation data, measured around each sample point in concentric rings, were collected in 2020. Three submodels of the Extended R-Value (ERV) model were used to relate pollen percentages to vegetation composition. Therewith, in order to create a calibrated model, the plant abundance of each pollen type was weighed by distance in GIS environment. The findings led to the conclusion that most of the tree taxa have PPE higher than 1 (ERV3 submodel). Cichoriceae, Fabaceae and Asteraceae have lower PPE.
