Plant tissue culture is becoming an important technique for the massive propagation of date palm and the available source lead to the success of this process. The mature female inflorescence is a novel and available source for in vitro propagation of date palm, nowadays become needed at the large scale to usage of organic nature materials in agriculture practice which are rich in plant growth regulators as well as micro elements. This study describes in vitro plant regeneration from mature female inflorescence explants of evaluating date palm (Phoenix dactylifera L.) by using the methanol extract of Moringa oleifera leaves which has great potential to be used as a natural source of nutraceutical in micropropagation. Mature inflorescences explants were cultured on Murashige and Skoog (MS) medium supplemented with different concentrations of an extract of  Moringa leaves with or without add TDZ at 0.2 and 0.6 BA mg/l. Indoles, phenols,  proteins,  amino acids, and carbohydrates were determined. The addition of different concentrations of an extract of Moringa oleifera leaves with TDZ at 0.2 and 0.6 BA mg/l gave the highest significant average percentage of callus formation, friable callus, germination of embryos and numbers of embryos comparing with other treatments. Also, given the highest significant values of free amino acids and carbohydrates were noted in callus and embryo germination stage.

Moringa oleifera is a foliated tree widely cultivated in tropical latitudes, which is highly adaptable to climatic conditions and dry soils. Every part of the plant has nutritional, therapeutic or industrial benefits. This is due to its phytochemicals such as glucosinolates, phenolic compounds, alkaloids, terpenoids and tannins, high values of crude protein, carbohydrates, starch and lipids. In addition, the use of the leaves has increased considerably by the agro-food and biochemical industries since they are a valuable source of dietary proteins and essential amino acids. This work aimed to characterize three types of leaf from Moringa oleifera seeds with different origins (Thai (C1), Ghana (C2) and India (C3)), grown in the same plot, but with different cultural practices (intended for leaf production (C1 and C2) or sheath production (C3). For this, water content and optical properties were determined in the fresh leaves. Later the leaves were dried (50 °C for 8 h) and pulverized, analyzing their water content, antioxidant capacity, color and amino acid content. No significant differences were observed in fresh leaves in terms of humidity and color. In dry powder, a higher antioxidant capacity was registered in moringa type C2, with a% DPPH inhibition of 83.7%, although in all cases, it exceeded 60%, showing the high persistence of the antioxidants after drying. Serine, glutamic acid and alanine were the major amino acids with values of 373 ± 78, 301 ± 51 and 248 ± 9 mg/100 g of powder, respectively, without influencing the applied field treatment or origin.

After inoculation of young leaves of date palm offshoot required about six months to complete the morphogenesis process. Fourteen weeks were required for embryogenic callus formation under continuous dark condition and nine weeks for shoot initiation (under 16/8 h light/dark). The highest number of explants (80%) produced callus in modified MS containing 5 mg/l 2,4-D + 2 mg/l 2ip. Sixty per cent of explants produced callus in the modified medium containing 5 mg/l 2,4-D + 5 mg/l NAA. while only 50 per cent of the explants formed callus in the same medium when supplemented with only 5 mg/l 2,4-D. The induced calli were transferred to modified MS for shoot proliferation. A combination of two cytokines showed better performance than single ones in shoot induction. The highest percentage (70) of shoot developed in modified MS containing 2 mg/l BAP + 1 mg/l Kn. Forty per cent shoot induction was found in the same medium supplemented with 2 mg/l of BAP. Thirty per cent shoot formed in MS containing 1 mg/l of Kn. The shoots were subcultured at three- four week intervals throughout culture duration. D. O. Background: Regular brushing teeth with scaling and root planning (SRP) cannot effectively decrease the periopatogen bacterial colonies. Even with the addition of antibiotics to support SRP, such as tetracycline given with low doses and for a long time may cause bacteria to become resistant and the effectiveness to eliminate colonies of bacteria being reduced. Photodynamic is a treatment modality that does not cause resistance and potentially to eliminate the growth of bacterial colonies. Moringa oleifera is a plant that can be easily found in Indonesia, by extracting chlorophyll of Moringa oleifera leaves, it can be used as a photosensitizer agent to increase the absorption of light on photodynamic method. Purpose: This study aimed to determine the potential photodynamic inactivation therapy to inactivate (eliminate) periopatogen bacterium. Method: This study used Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans). Laser diode 660nm as a light source with 8mm optical fiber to guide the beam, also used 20% extract chlorophyll of Moringa oleifera leaf as photosensitizer. Four diode lasers energy density exposures (2,5J.cm-2, 5J.cm-2, 7,5J.cm-2, and 10J.cm-2) are used from both at the in vitro photodynamic inactivation test. Result: The highest percentage of deaths occurred in the group treated with addition of photosensitizer and exposed by 660 nm diode laser with 10J.cm-2 energy density, which is 83.01%, compared to the results obtained in the group without addition of the photosensitizer. Conclusion: Chlorophyll of Moringa oleifera leaf after activated by diode laser effectively eliminates A. actinomycetemcomitans.

The effect of different explants and concentrations of BAP and NAA on induction of callus and plant regeneration of brinjal cv. Jhumki were investigated. The treatment combinations were BAP (0. 2.0. 3.0, and 4.0 mg/l) and NAA (0. 0.1, 0.5, and 1.0 mg/l). The rate of callus formation varied in different treatments. The highest amount of callus (48.66%) was produced on MS medium containing 2.0 mg/l BAP and 0.5 mg/l NAA from stem, and 8.2 days required for callus induction. The highest fresh weight of callus was 1.12g from stem and 0.48g from root. The number of shoot regenerated through callus from stem containing 2.0 mg/l BAP and 0.5 mg/l NAA was 3.4 (23.287%) and days required for 38.8 days. Rich in phenolic compounds, Moringa oleifera leaf extract (ME) exhibits significant antioxidant activity both in vitro and in vivo. ME has already been widely used in fields of medicine, functional food, and cosmetics. Ultrasonic extraction (UE) method has been improved to be one of the most effective ways to extract phenols from M. oleifera leaves. The purpose of this study was to optimize ultrasonic extraction of phenols by response surface methodology (RSM). Four parameters were discussed, such as ethanol concentration, solvent-sample ratio, extraction temperature, and extraction time. Also, purification methods of the crude ME by organic solvent extraction and column chromatography were examined. Antioxidant activities of ME and each fraction were evaluated by DPPH, ABTS, and hydroxy radical-scavenging activities and reducing power. The phenol content of the purified ME reached up to 962.6 mg RE/g, extremely higher than the crude extract 107.22 ± 1.93 mg RE/g. The antioxidant activity of the purified ME was also significantly improved. Furthermore, phenols were identified by using the HPLC-MS method, and the results showed that there were 6 phenolic acids and derivatives and 7 flavonoids in ME. Quercetin-3-O-β-D-glucoside isolated from ME showed excellent DPPH and ABTS radical-scavenging abilities, which were comparable to VC.
