Рибництво – важлива та невід’ємна складова частина бізнесу в Україні, у якій розведення окре-мих видів риб відіграє вирішальну роль. Варто відмітити, що антропогенний вплив людини на водні об’єкти, сучасний стан екології, зростання темпів поселення чужорідних видів організмів до водних екосистем України сукупно сприяють формуванню нових симбіотичних відносин між представника-ми іхтіофауни та паразитичними організмами, зокрема й паразитичними найпростішими. Так звані «нові відносини» зазвичай призводять до погіршення епізоотичної ситуації у водоймах, спричинюючи появу масових захворювань риби. Внаслідок цього знижується кількість та якість рибної продукції, що призводить до економічних збитків. Зважаючи на це, метою цього огляду є визначення сучасного стану фауни паразитичних найпростіших організмів окремих видів риб прісноводних водойм України. Як об’єкт огляду було обрано різні види риб – білий амур, білий товстолобик, бичок-пісочник, окунь, карась (звичайний – золотий та карась сріблястий), щука, краснопірка, короп та плітка. Саме наведені види риб входять у ядро іхтіоценозу переважної кількості прісноводних водойм України та мають різний характер харчування. У праці висвітлено дані щодо фауни парази-тичних найпростіших зазначених видів риб. Встановлено, що наразі фауна протозоозів риб налічує 59 видів, які відносяться до 5-и класів: Kinetoplastomonada (Honigberg, 1963), Coccidiomorphea (Doflein, 1901), Myxozoa (Grassé, 1970), Oligohymenophorea (de Puytorac et al., 1974) та Rinostomata (Jankowski, 1978), 14 родин та 18 родів. Домінуючим класом виявився Oligohymenophorea. Найбільш багатою, за даними науковців, виявилася фауна білого амура (34 види), білого товстолобика (29 видів) та карася (20 видів). Найменшого ураження паразитичними найпростішими, за даними дослідників, зазнають окунь, щука, бичок-пісочник та краснопірка, у яких зафіксовано по 2‒3 види паразитичних найпростіших. Варто відмітити, що з-поміж виявлених паразитичних найпростіших 10 видів (Costia necatrix Henneguy, 1884, Sphaerospora carassii Kudo, 1919, Myxobolus dispar Thélohan, 1895, M. ellipsoides Thélohan, 1892, Ichthyophthirius multifiliis Fouquet, 1876, Trichodina reticulata Hirschmann et Partsch, 1955, Trichodinella epizootica Raabe, 1950, Chloromyxum fluviatile Thélohan, 1892, Tripartiella bulbosa Davis, 1947 та Balantidium ctenopharyngodoni Chen, 1955) є епізоотично важливими та можуть бути надзвичайно небезпечними для аквакультури.

Ichthyophthirius multifiliis is a major pathogen that causes a high mortality rate in trout farms. However, systemic responses to the pathogen and its interactions with multiple organs during the course of infection have not been well described. In this study, dual-organ transcriptomic responses in the liver and head kidney and hemato-serological indexes were profiled under I. multifiliis infection and recovery to investigate systemic immuno-physiological characteristics. Several strategies for massive transcriptomic interpretation, such as differentially expressed genes (DEGs), Poisson linear discriminant (PLDA), and weighted gene co-expression network analysis (WGCNA) models were used to investigate the featured genes/pathways while minimizing the disadvantages of individual methods. During the course of infection, 6,097 and 2,931 DEGs were identified in the head kidney and liver, respectively. Markers of protein processing in the endoplasmic reticulum, oxidative phosphorylation, and the proteasome were highly expressed. Likewise, simultaneous ferroptosis and cellular reconstruction was observed, which is strongly linked to multiple organ dysfunction. In contrast, pathways relevant to cellular replication were up-regulated in only the head kidney, while endocytosis- and phagosome-related pathways were notably expressed in the liver. Moreover, interestingly, most immune-relevant pathways (e.g., leukocyte trans-endothelial migration, Fc gamma R-mediated phagocytosis) were highly activated in the liver, but the same pathways in the head kidney were down-regulated. These conflicting results from different organs suggest that interpretation of co-expression among organs is crucial for profiling of systemic responses during infection. The dual-organ transcriptomics approaches presented in this study will greatly contribute to our understanding of multi-organ interactions under I. multifiliis infection from a broader perspective.

The skin mucus is the fish primary defense barrier protecting from infections via the skin epidermis. In a previous study, we have investigated the proteome of common carp (Cyprinus carpio) skin mucus at two different time points (1 and 9 days) post-exposure to Ichthyophthirius multifiliis. Applying a nano-LC ESI MS/MS technique, we have earlier revealed that the abundance of 44 skin mucus proteins has been differentially regulated including proteins associated with host immune responses and wound healing. Herein, in skin mucus samples, we identified six proteins of I. multifiliis associated with the skin mucus in common carp. Alpha and beta tubulins were detected in addition to the elongation factor alpha, 26S proteasome regulatory subunit, 26S protease regulatory subunit 6B, and heat shock protein 90. The identified proteins are likely involved in motility, virulence, and general stress during parasite growth and development after parasite attachment and invasion. Two KEGG pathways, phagosome and proteasome, were identified among these parasite proteins, mirroring the proteolytic and phagocytic activities of this parasite during host invasion, growth, and development, which represent a plausible host invasion strategy of this parasite. The results obtained from this study can support revealing molecular aspects of the interplay between carp and I. multifiliis and may help us understand the I. multifiliis invasion strategy at the skin mucus barrier. The data may advance the development of novel drugs, vaccines, and diagnostics suitable for the management and prevention of ichthyophthiriosis in fish.

The objective of this research was to establish loop-mediated isothermal amplifications (LAMP) that could be used to detect parasitic ciliate Ichthyophthirius multifiliis (I. multifiliis) in freshwater cyprinid fish. Primers were developed from the distinguishing fragments of 18S ribosomal RNA of I. multifiliis and the LAMP test was then used to evaluate and optimize various concentrations of chemicals, time and temperature. The results indicated that LAMP required 1.6 μM of FIP primers and BIP primers, 0.2 μM of F3 and B3, 2 mM of Mg2+, 1 M of Betaine, and 0.6 mM of dNTP. This assay was able to detect parasite DNA within a 40 min period of incubation and at a constant optimal temperature of 64oC. The positive sample appeared as a clear ladder like pattern on gel electrophoresis, while a yellowish green color appeared with SYBR Green I under ultraviolet light with the use of a heating block. The LAMP test was determined to be more sensitive than conventional PCR in the detection of I. multifiliis. In conclusion, we have presented a sensitive and specific rapid detection system for I. multifiliis based on isothermal DNA amplification. Importantly, this system could then be employed as an alternative and effective diagnostic method in place of other molecular techniques.
