Both the detrimental effect of prenatal exposure to di-(2-ethylhexyl)-phthalate (DEHP) and the beneficial effects of physical exercise on brain functions have been reported. The oxytocin pathway has been implicated in the onset of maternal behaviors. Epigenetic modification of the oxytocin receptor gene (OXTR) through DNA methylation has been associated with the pathogenesis of neuropsychiatric disorders. The purpose of this study was to investigate the effects of prenatal DEHP exposure on oxytocin-regulated maternal behaviors and to examine the protective effect of exercise. Pregnant rats (F0) were fed with vehicle or DEHP during gestation and the offspring females (F1) were assessed for their maternal behaviors by pup retrieval test at postpartum. The results showed that reduced pup retrieval activities without significant alteration of stress responses were observed in the prenatally DEHP-exposed females. Prenatal DEHP exposure decreased the expressions of oxytocin, Oxtr mRNA, and oxytocin receptor, and increased Oxtr methylation in the hypothalamus of postpartum female rats. There were no significant effects of exercise on behavioral, biochemical, and epigenetic measurements. These results suggest that prenatal DEHP exposure has a long-term adverse effect on maternal behaviors; Oxtr hyper-methylation may be a potential epigenetic mechanism for this alteration, which cannot be prevented by physical exercise during childhood.

Defining the activity patterns and social relationships of animals can provide valuable information related to animal welfare. Bottlenose dolphins under professional care engage in a variety of social and solitary activities, which may be influenced by conspecifics, time of day, and maternal behaviors. This study examined the social, solitary, and maternal behaviors of eight dolphins at one zoological facility. Data about social and solitary behaviors, nursing occurrences, and mother–calf pair swimming were collected five days per week over a ten-month period. The simple ratio index was used to calculate association rates for all dyads. Affiliative behaviors were the dominant behavior class and all behavior categories and swim states showed temporal oscillations throughout the day. The high association values within the mother–calf dyads suggested these ties remain strong through the second and third years of life. Preferred associates remained the same for some individuals in both social group compositions, while they differed for others. However, no large reductions in associations were recorded, suggesting strong relationships in the subgroups continued when other individuals were present. Examining sociality and activity levels may be a useful tool in continuing to improve welfare as they may be related to social groupings and environmental factors.

AbstractThe preoptic area (POA) is a brain structure classically involved in a wide variety of animal behavior including sleep and maternal care. In the current study, we evaluate the specific effect of disinhibition of two specific regions of the POA, the medial POA nucleus (mPOA) and the ventrolateral POA area (VLPO) on sleep and maternal behavior in lactating rats. For this purpose, mother rats on postpartum day 1 (PPD1) were implanted for polysomnographic recordings and with bilateral cannulae either in the mPOA or in the VLPO. The rats were tested for sleep and maternal behavior on PPD4-8 after the infusion of the GABA-A antagonist, bicuculline (0, 10 or 30 ng/0.2 µl/side). Infusion of bicuculline into the mPOA augmented retrieving and nest building behaviors and reduced both nursing and milk ejections but had almost no effect on sleep. When bicuculine was microinjected into the VLPO, the rats significantly increase the number of retrievings and mouthings and reduced the nursing time without changes in milk ejections, which was associated with an increase in wakefulness and a reduction in light sleep.Our results show that disinhibition of the mPOA, a key area in the control of maternal behavior, increased active maternal behaviors and reduced nursing without affecting wakefulness or sleep time. In contrast, the enhancement of some active maternal behaviors when the drug was infused into the VLPO, a sleep-promoting area, with a concomitant increase in wakefulness suggests that mother rats devote this additional waking time in the active maternal care of the pups. We hypothesize that maternal behavior changes after bicuculine microinjection into the VLPO is caused by a reduction in the sleep drive, rather than a direct effect on maternal behavior.

Childhood aggression is important to acknowledge due to its social impact and importance in predicting future problems. The temperament of a child and parental socialization have been essential in explaining behavioral problems, particularly in the case of childhood aggression. The aim of this study is to examine—from the parents’ perspective—the role of childhood temperament in the dynamic by which mothers’ reactions socialize their children’s aggression. We also explore how children’s gender and age differences affect these relationships. The sample was composed of 904 participants between 1 and 6 years old. The Early Childhood Behavior Questionnaire and the Children’s Behavior Questionnaire were used to evaluate children’s negative affect and effortful control. The Parent–Child Relationship Inventory Maternal was used to assess maternal communication and discipline, and child aggression was assessed using the Children’s Behavior Checklist. The results supported the mediating role of temperament in the processes by which perceived mothers’ reactions socialize their children’s aggression and suggested that maternal behaviors may not have the same consequences for girls and boys. Specifically, the aggressiveness of girls is dependent on a negative affect throughout toddlerhood and early childhood, while for boys, the duration of the negative affect’s contribution is shorter, and aggressiveness is more sensitive to the maternal behaviors of discipline and communication.
