O-GlcNAcylation is a reversible and dynamic post-translational protein modification catalyzed by O-GlcNAc transferase (OGT). Despite the reported association of O-GlcNAcylation with cancer metastasis, the O-GlcNAc proteome profile for cancer aggressiveness remains largely uncharacterized. Here, we report our comparative O-GlcNAc proteome profiling of two differentially invasive lung adenocarcinoma cell lines, which identified 158 down-regulated and 106 up-regulated candidates in highly invasive cells. Among these differential proteins, a nuclear RNA-binding protein, SAM68 (SRC associated in mitosis of 68 kDa), was further investigated. Results showed that SAM68 is O-GlcNAcylated and may interact with OGT in the nucleus. Eleven O-GlcNAcylation sites were identified, and data from mutant analysis suggested that multiple serine residues in the N-terminal region are important for O-GlcNAcylation and the function of SAM68 in modulating cancer cell migration and invasion. Analysis of clinical specimens found that high SAM68 expression was associated with late cancer stages, and patients with high-OGT/high-SAM68 expression in their tumors had poorer overall survival compared to those with low-OGT/low-SAM68 expression. Our study revealed an invasiveness-associated O-GlcNAc proteome profile and connected O-GlcNAcylated SAM68 to lung cancer aggressiveness.

AbstractMoral rules allow humans to cooperate by indirect reciprocity. Yet, it is not clear which moral rules best implement indirect reciprocity and are favoured by natural selection. Previous studies either considered only public assessment, where individuals are deemed good or bad by all others, or compared a subset of possible strategies. Here we fill this gap by identifying which rules are evolutionary stable strategies (ESS) among all possible moral rules while considering private assessment. We develop an analytical model describing the frequency of long-term cooperation, determining when a strategy can be invaded by another. We show that there are numerous ESSs in absence of errors, which however cease to exist when errors are present. We identify the underlying properties of cooperative ESSs. Overall, this paper provides a first exhaustive evolutionary invasion analysis of moral rules considering private assessment. Moreover, this model is extendable to incorporate higher-order rules and other processes.

In ecological communities, the behaviour of individuals and the interaction between species may change between seasons, yet this seasonal variation is often not represented explicitly in mathematical models. As global change is predicted to alter season length and other climatic aspects, such seasonal variation needs to be included in models in order to make reasonable predictions for community dynamics. The resulting mathematical descriptions are nonautonomous models with a large number of parameters, and are therefore challenging to analyze. We present a model for two predators and one prey, whereby one predator switches hunting behaviour to seasonally include alternative prey when available. We use a combination of temporal averaging and invasion analysis to derive simplified models and determine the behaviour of the system, in particular to gain insight into conditions under which the two predators can coexist in a changing climate. We compare our results with numerical simulations of the temporally varying model.

The article provides information on monitoring studies of polar bear helminthiasis in the wild population of Franz Josef Land and in zoos in Russia. Wild bears were found to be free from invasion. Analysis of fecal samples from polar bears from 17 zoos showed that the total extensiveness of the invasion was 21%, 2 types of helminths were identified: Baylisascaris transfuga and Diphyllobothrium sp. Samples with helminth eggs were found in zoos in Nizhny Novgorod, Rostov-on-Don, Penza, Udmurtia, Khabarovsk, Novosibirsk, Kazan, Krasnoyarsk, Moscow, Volokolamsk, Seversk, Perm.
