Entomopathogenic fungi (EPF) in Croatian forests are known only from observations of insect cadavers that show obvious signs of disease. To date, their presence in soils has not been investigated. The aim of this study was to investigate their occurrence, diversity, and distribution, and to assess their density in tested soils. Soil samples were collected during 2018, 2019, and 2020 at different localities throughout the country, and analyzed by using a method of isolation of fungi on selective culture media. To assess the density of EPF in tested soils, colonies of individual fungal species were counted and recorded; the results were expressed as the number of colony-forming units (CFU) per gram of dry soil. After morphological and molecular analysis, five entomopathogenic fungal genera were identified: Beauveria spp., Metarhizium spp., Purpureocillium spp., Lecanicillium spp., and Paecilomyces spp. Results also showed that the range of a total EPF colony density in the soil varies from 4 × 103 to 27.4 × 103 CFU g−1. The most common were EPF of the genus Beauveria, which were recorded at four of five locations, and at 16 of 25 sampling points, but the highest average number (density) of colonies belonged to the genus Metarhizium. Since this type of research was never conducted in Croatia previously, this is the first evidence that insect pathogenic fungi are present in soils of different natural forest habitats. Such research can be useful in selecting and utilizing entomopathogens that are suitable for biological pest control in certain target areas.

Abstract The Mormon cricket (MC), Anabrus simplex Haldeman, 1852 (Orthoptera: Tettigoniidae), has a long and negative history with agriculture in Utah and other western states of the USA. Most A. simplex populations migrate in large groups, and their feeding can cause significant damage to forage plants and cultivated crops. Chemical pesticides are often applied, but some settings (e.g. habitats of threatened and endangered species) call for non-chemical control measures. Studies in Africa, South America, and Australia have assessed certain isolates of Metarhizium acridum as very promising pathogens for Orthoptera: Acrididae (locust) biocontrol. In the current study, two isolates of Metarhizium robertsii, one isolate of Metarhizium brunneum, one isolate of Metarhizium guizhouense, and three isolates of M. acridum were tested for infectivity to MC nymphs and adults of either sex. Based on the speed of mortality, M. robertsii (ARSEF 23 and ARSEF 2575) and M. brunneum (ARSEF 7711) were the most virulent to instars 2 to 5 MC nymphs. M. guizhouense (ARSEF 7847) from Arizona was intermediate and the M. acridum isolates (ARSEF 324, 3341, and 3609) were the slowest killers. ARSEF 2575 was also the most virulent to instar 6 and 7 nymphs and adults of MC. All of the isolates at the conidial concentration of 1 × 107 conidia ml−1 induced approximately 100% mortality by 6 days post application of fungal conidia. In conclusion, isolates ARSEF 23, ARSEF 2575, and ARSEF 7711 acted most rapidly to kill MC under laboratory conditions. The M. acridum isolates, however, have much higher tolerance to heat and UV-B radiation, which may be critical to their successful use in field application.

The red flour beetle is a very important primary insect pest of wheat worldwide. The experiments were performed to check the virulence of Beauveria bassiana and Isaria fumosorosea by insect dipping method on the adults and 2nd instar of Tribolium castaneum. Conidia were taken from 15 days old fungi and subsequently four concentrations i.e., 2×108, 3×108, 4×108 and 5×108 spores/ml of both fungi were prepared in 0.05% Tween 80 solution. Minimum 12.5% and maximum 32.5% mortality of adult insects was recorded on 7th day after the treatment at 2×108 and 5×108 spores/ml concentrations of B. bassiana, respectively while on larval stages, minimum 2.5% on 5th day and maximum 80.0% mortality was observed on 7th day post treatment of B. bassiana, respectively. On the other side, minimum 7.5 and maximum 22.5 mortality percentage was noted on 7th day post application of I. fumosorosea, respectively while on immatures minimum 5% on 6th day and highest 70% mortality was noted on 7th day post infection with 2×108 and 5×108 spores/ml of I. fumosorosea, correspondingly. This study showed the effectiveness of insect pathogenic fungi against the important stored grain insect pest and proved to be a positive management strategy.

Background: In our previous study, we have isolated a new compound, named Fumosorinone (FU) from insect pathogenic fungi, and was found to inhibit proliferation, migration, and invasion of breast cancer MDA-MB-231 cells. Objective: The aim of this study was to identify the underlying molecular mechanisms for FU effects on MDAMB- 231 cells. Methods: After MDA-MB-231 cells were treated with FU for 48h, RNA sequencing was used to identify the effect of FU on the transcriptome of MDA-MB-231 cells. The validation of the relative expression of the selective genes was done using quantitative real-time PCR (qRT-PCR). Results: The transcriptome results showed that 2733 genes were differentially expressed between the untreated and the FU-treated cells, including 1614 up-regulated and 1119 down-regulated genes. The multiple genes are associated with cancer cell growth, migration, and invasion. Functional analysis identified multitude of pathways related to cancer, such as cell cycle, ECM–receptor interaction, p53 signaling pathway. We selected 4 upregulated and 9 downregulated genes, which are associated with breast cancer to verify their expression using qRT-PCR. The validation showed that HSD3B1, ALOX5, AQP5, COL1A2, CCNB1, CCND1, VCAM-1, PTPN1 and PTPN11 were significantly downregulated while DUSP1, DUSP5, GADD45A, EGR1 were upregulated in FU-treated MDA-MB-231cells. Conclusion: These aberrantly expressed genes and pathways may play pivotal roles in the anti-cancer activity of FU, and maybe potential targets of FU treatments for TNBC. Further investigations are required to evaluate the FU mechanisms of anti-cancer action in vivo.
