Abstract Background Cells show directed migration response to electric signals, namely electrotaxis or galvanotaxis. PI3K and PTEN jointly play counterbalancing roles in this event via a bilateral regulation of PIP3 signaling. PI3K has been proved essential in anterior signaling of electrotaxing cells, whilst the role of PTEN remains elusive. Methods Dictyostelium cells with different genetic backgrounds were treated with direct current electric signals to investigate the genetic regulation of electrotaxis. Results We demonstrated that electric signals promoted PTEN phosphatase activity and asymmetrical translocation to the posterior plasma membrane of the electrotaxing cells. Electric stimulation produced a similar but delayed rear redistribution of myosin II, immediately before electrotaxis started. Actin polymerization is required for the asymmetric membrane translocation of PTEN and myosin. PTEN signaling is also responsible for the asymmetric anterior redistribution of PIP3/F-actin, and a biased redistribution of pseudopod protrusion in the forwarding direction of electrotaxing cells. Conclusions PTEN controls electrotaxis by coordinately regulating asymmetric redistribution of myosin to the posterior, and PIP3/F-actin to the anterior region of the directed migration cells.

Abstract This chapter examines a cluster of wildfire conflagrations that hit northern California during October 2017, which resulted in significant loss of housing units (6874 residential structures destroyed or damaged). To assess the magnitude of the migration response and network of destinations, a method to estimate migration drawing from a proxy universe of households with students enrolled in public schools was proposed, using data on school exits and re-enrollments from a longitudinal student database. The analysis finds that a small minority of households affected by the fires moved out of the area. Out of nearly 7800 persons displaced by the central fire complex in one city, this study estimated fewer than 1000 changed neighborhoods; of those, fewer than 500 moved out of Sonoma County. These findings are applicable to other wildfires and localized disasters where a substantial portion of housing is lost but public infrastructure in the region remains intact.

Accumulating evidence suggests that NKX6.1 (NK homeobox 1) plays a role in various types of cancer. In our previous studies, we identified NKX6.1 hypermethylation as a promising marker and demonstrated that the NKX6.1 gene functions as a metastasis suppressor through the epigenetic regulation of the epithelial-to-mesenchymal transition (EMT) in cervical cancer. More recently, we have demonstrated that NKX6.1 methylation is related to the chemotherapy response in colorectal cancer (CRC). Nevertheless, the biological function of NKX6.1 in the tumorigenesis of CRC remains unclear. In this study, we showed that NKX6.1 suppresses tumorigenic and metastatic ability both in vitro and in vivo. NKX6.1 represses cell invasion partly through the modulation of EMT. The overexpression of NKX6.1 enhances chemosensitivity in CRC cells. To further explore how NKX6.1 exerts its tumor-suppressive function, we used RNA sequencing technology for comprehensive analysis. The results showed that differentially expressed genes (DEGs) were mainly related to cell migration, response to drug, transcription factor activity, and growth factor activity, suggesting that these DEGs are involved in the function of NKX6.1 suppressing cancer invasion and metastasis. Our results demonstrated that NKX6.1 functions as a tumor suppressor partly by repressing EMT and enhancing chemosensitivity in CRC, making it a potential therapeutic target.

This article argues for the interplay of fear of God’s judgement and responsibility compulsion as a nexus for encouraging responsible migration response. This arises from the need of theology of migration that challenges the church to respond to migration challenges in a comprehensive manner. Given this, the aim of this article is to contribute migration theology that respond to the aforementioned proposed need. It utilises the available literature to establish the role of Israel in God’s redemptive history that includes her positive treatment of people from other nations, thereby also reminding the nations of what God expects of all people. In bringing the issue of the interplay of fear of God’s judgement and responsibility compulsion as a nexus for encouraging responsible migration response, it advances that God promises to judge and punish his people when they oppose his care and love for the vulnerable people such as aliens, widows and orphans (Ex 22:21–27). However, in remaining faithful to his aforementioned promises to the Israelites, God confronts them (the Israelites) in anger, judgement and punishment when they disobey him in the way they manage their relationship with people of other nations (Jr 7:1–15, 52; 2 Ki 24:10–20). With this in mind, it concludes by advancing the interplay of fear of God’s judgement and responsibility compulsion as nexus for encouraging responsible migration response within contemporary churches in South Africa and beyond.Intradisciplinary and/or interdisciplinary implications: This article outlines the issue of fear of judgement and responsibility compulsion as a nexus for responsible migration response. It underscores the aforementioned by delving into a thorough exegesis of the relevant Old Testament passages and looking at Israel’s role in redemptive history as God’s people. It then presents the role of Israel as accomplished by the God-man, Jesus Christ, who established the church as a new covenant community of God. As such, it is a theological interdisciplinary article integrating exegesis, biblical theology, systematic theology and practical theology.
