AbstractIn light of Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) variants potentially undermining humoral immunity, it is important to understand the fine specificity of the antiviral antibodies. We screened 20 COVID-19 patients for antibodies against 9 different SARS-CoV-2 proteins observing responses against the spike (S) proteins, the receptor-binding domain (RBD), and the nucleocapsid (N) protein which were of the IgG1 and IgG3 subtypes. Importantly, mutations which typically occur in the B.1.351 “South African” variant, significantly reduced the binding of anti-RBD antibodies. Nine of 20 patients were critically ill and were considered high-risk (HR). These patients showed significantly higher levels of transforming growth factor beta (TGF-β) and myeloid-derived suppressor cells (MDSC), and lower levels of CD4+ T cells expressing LAG-3 compared to standard-risk (SR) patients. HR patients evidenced significantly higher anti-S1/RBD IgG antibody levels and an increased neutralizing activity. Importantly, a large proportion of S protein-specific antibodies were glycosylation-dependent and we identified a number of immunodominant linear epitopes within the S1 and N proteins. Findings derived from this study will not only help us to identify the most relevant component of the anti-SARS-CoV-2 humoral immune response but will also enable us to design more meaningful immunomonitoring methods for anti-COVID-19 vaccines.

<p>Slot machines are a remarkably popular mode of gambling even though they are programmed to make a profit by paying out less money than is put in. One common feature of slot machines, which may increase the likelihood of persistent gambling in the face of this monetary loss, is the near win. This study’s aim was to investigate the conditioned reinforcing properties of near wins using an observing response procedure in the context of a simulated slot machine. In an observing response procedure, participants can use an observing button to produce a stimulus correlated with the availability of reinforcement (S+) or a stimulus correlated with no reinforcement or less reinforcement (S-). The percentage of observing responses made for each stimulus is thought to reflect the reinforcing efficacy of the reinforcer correlated with each stimulus. Experiment 1 successfully tested the procedure with an obvious reinforcer - wins - and found consistently more observing for the S+. In Experiment 2 and 3 the S+ was correlated with near wins, and in Experiment 2 only those with slot-machine experience had consistently more observing for the S+. Experiment 3 increased the probability of wins to enhance the reinforcing efficacy of near wins, but failed to find consistently more observing for the S+, regardless of slot machine or scratchie card experience. These results indicated that near wins are not conditioned reinforcers. However, participants tended to bet more following near wins than losses, which suggested that near wins may instead function as discriminative stimuli.</p>

Various food products distributed throughout the cold chain can present a health risk for consumers due to the presence of psychrotolerant B. cereus group species that possess enterotoxin genes and antibiotic resistance. As these bacteria can grow at the low temperatures used in the food industry, this study evaluated the antimicrobial efficacy of acetic acid, sodium hypochlorite, and thermal treatments for inhibition of psychrotolerant strains and the effect that differences in activation temperature (30 °C and 10 °C) have on their efficacy. The minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and bacterial growth assay of acetic acid and thermal treatment showed an equal or higher antimicrobial efficacy in isolates activated at 10 °C than in those activated at 30 °C. In particular, psychrotolerant strains from the B. cereus group were completely eliminated with 0.25% acetic acid, regardless of the activation temperature. The possibility of tolerance was determined by observing responses in cells activated at 10 and 30 °C when exposed to different concentrations of sodium hypochlorite. Five isolates activated at 10 °C exhibited enhanced survivability in sodium hypochlorite compared to isolates activated at 30 °C, and these isolates were able to grow in sodium hypochlorite at concentrations of 250 ppm or higher. Although a significant difference in antimicrobial efficacy was observed for psychrotolerant B. cereus group strains depending on the activation temperature, acetic acid may be the most effective antimicrobial agent against psychrotolerant B. cereus species isolated from food products distributed in a cold chain.

The feasibility of a neural network method was discussed in terms of a self-tuning proportional–integral–derivative (PID) controller. The proposed method was configured with two neural networks to dramatically reduce the number of tuning attempts with a practically achievable small amount of data acquisition. The first network identified the target system from response data, previous PID parameters, and response characteristics. The second network recommended PID parameters based on the results of the first network. The results showed that it could recommend PID parameters within 2 s of observing responses. When the number of trained data was as low as 1000, the performance efficiency of these methods was 92.9%, and the tuning was completed in an average of 2.94 attempts. Additionally, the robustness of these methods was determined by considering a system with noise or a situation when the target position was modified. These methods are also applicable for traditional PID controllers, thus enabling conservative industries to continue using PID controllers.
