This study examined the thermal and catalytic pyrolysis of waste Achyranthes Root (AR) using pyrolyzer-gas chromatography/mass spectrometry (Py-GC/MS). The non-catalytic pyrolysis of waste AR produced various kinds of oxygenates, such as acetic acid, hydroxy propanone, furfural, phenol, cresol, guaiacols, syringols, and so on. By applying nanoporous Al-MCM-41 with acidic properties and mesopores to the pyrolysis of waste AR, the levels of furan and aromatic hydrocarbons production increased with a concomitant decrease in the other oxygenates. The formation efficiency of furans was improved further by increasing the amount of Al-MCM-41 applied to the catalytic pyrolysis of waste AR.

Achyranthes bidentata Blume, a traditional Chinese medicine, is widely acknowledged for its function of invigorating the liver and kidneys and as a stranguria-relieving diuretic and used in the treatment of edema, gonorrhea, and other diseases. Polysaccharide (ABPS), isolated from Achyranthes bidentata Blume, has been demonstrated to have multiple biological activities including immunomodulatory effects. However, the mechanisms underlying the effects of ABPS have not been fully investigated. The present study is conducted to explore the underlying mechanism of immunomodulatory activities of ABPS. Results showed that ABPS significantly increased the secretion of IL-1β and TNF-α in J744 A.1 cells. Nitric oxide (NO) also significantly increased after ABPS treatment. The special antibodies (Toll-like receptor 4 (TLR4) antibody and CD14/TLR4 antibody) significantly decreased the activation, while the Toll-like receptor 2 (TLR2) antibody could not abolish this activation. Meanwhile, pyrrolidine dithiocarbamate (PDTC), a specific inhibitor of NF-κB, remarkably inhibited the secretion of IL-1β and TNF-α induced by ABPS in J744 A.1 cells. Western blotting (WB) and confocal laser scanning microscopy (CLSM) showed that ABPS promoted NF-κB translocation into the nucleus. Furthermore, the mRNA and protein expression of TLR4 and MyD88 were significantly increased after ABPS treatment. Taken together, these findings suggested that the immunomodulatory mechanism of ABPS was associated with the secretion of cytokines by stimulating the NF-κB pathway through TLR4/MyD88 signaling.

Achyranthes bidentata polysaccharides (ABPS), a major active ingredient of Achyranthes bidentata, has many kinds of well-known pharmacological activity, such as anti-coagulation, indirect anti-tumor, anti-aging and hypoglycemic effect, and also have certain effects on the immune system. However, the direct anti-tumor activity of ABPS remain unclear. In this paper, ABPS was efficiently isolated from Achyranthes bidentata adopted orthogonal experimental method, and then its anti-tumor effect on LTEP-a-2 cells was investigated. LTEP-a-2 cells were treated with various concentrations of ABPS (0, 0.2, 0.4, 0.6, 0.8 and 1 mg/mL) for 48h. Subsequently, cell viability was analyzed by MTT method and the effects of ABPS on the apoptosis of LTEP-a-2 cells was explored by DNA ladder, PI and Annexin V FITC/PI double staining. The results showed that the optimum extraction conditions of ABPS was that the extraction temperature at 90oC, 1:40(W/V) of solid to liquid, and extraction time 4 h. In addition, the yield and purity of ABP was measured up to 15.52% and 97.21% respectively. Furthermore, ABPS significantly inhibited the proliferation and induced the apoptosis of LTEP-a-2 cells. So ABPS might be utilized as a new potential therapeutic agent against Lung Adenocarcinoma.
