The present study aims to assess the antimicrobial and antioxidant activity of selected medicinal plants (Achyranthes bidentata, Linum usitatissimum, Pedalium murex, Sphaeranthus indicus and, Terminalia bellirica) extracts against seven different microorganisms Candida albicans, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Staphylococcus epidermidis, Staphylococcus hominis, and Streptococcus mutants. Leaf, root, and flower extracts of plants were prepared in different solvents like methanol, distilled water, dichloromethane, ethanol, ethyl acetate, chloroform, petroleum ether, propanol, benzene, and hexane. All the prepared extracts showed very good antimicrobial activity except distilled water extract. Most of the extracts were found to have antimicrobial potential against pathogens but Linum usitatissimum and T. bellirica leaf and seed extract prepared in methanol and chloroform solvents show a higher zone of inhibition against E. coli. Only Linum usitatissimum shows activity aganist Candida albicans. Minimum Inhibitory Concentration for Achyranthes bidentata extracts varied from 150µl/ml to 200µl/ml in different solvents. Antioxidant studies were carried out in methanolic extracts of all the plants. The maximum scavenging activity of methanolic leaf extracts was observed between 80 -100 μg/ml concentrations.

Achyranthes bidentata Blume, a traditional Chinese medicine, is widely acknowledged for its function of invigorating the liver and kidneys and as a stranguria-relieving diuretic and used in the treatment of edema, gonorrhea, and other diseases. Polysaccharide (ABPS), isolated from Achyranthes bidentata Blume, has been demonstrated to have multiple biological activities including immunomodulatory effects. However, the mechanisms underlying the effects of ABPS have not been fully investigated. The present study is conducted to explore the underlying mechanism of immunomodulatory activities of ABPS. Results showed that ABPS significantly increased the secretion of IL-1β and TNF-α in J744 A.1 cells. Nitric oxide (NO) also significantly increased after ABPS treatment. The special antibodies (Toll-like receptor 4 (TLR4) antibody and CD14/TLR4 antibody) significantly decreased the activation, while the Toll-like receptor 2 (TLR2) antibody could not abolish this activation. Meanwhile, pyrrolidine dithiocarbamate (PDTC), a specific inhibitor of NF-κB, remarkably inhibited the secretion of IL-1β and TNF-α induced by ABPS in J744 A.1 cells. Western blotting (WB) and confocal laser scanning microscopy (CLSM) showed that ABPS promoted NF-κB translocation into the nucleus. Furthermore, the mRNA and protein expression of TLR4 and MyD88 were significantly increased after ABPS treatment. Taken together, these findings suggested that the immunomodulatory mechanism of ABPS was associated with the secretion of cytokines by stimulating the NF-κB pathway through TLR4/MyD88 signaling.

The experiment aimed to investigate the effects of probiotics and Achyranthes bidentata polysaccharides on the growth performance, nutrients digestibility, and immune function of weaned pigs. One hundred and twenty weaned pigs (about 7 kg BW, 23 ± 2 d) were allotted to five dietary treatments (CON: antibiotics-free basal diet; ANT: CON + antibiotics; PRO: CON + probiotics; ABPS: CON + Achyranthes bidentata polysaccharides; P-ABPS: PRO + ABPS) for a 28-day trial. Compared with CON, pigs in ANT, PRO, ABPS, and P-ABPS had greater (p < 0.05) ADG, ATTD of CP and GE, serum ALB, IgA and IL-2, duodenal intraepithelial lymphocyte, ileal VH and jejunal mucosa sIgA, but lower (p < 0.05) fecal scores, serum BUN, and IL-1β. Meanwhile, ANT, PRO, ABPS, and P-ABPS exhibited similar beneficial roles on growth performance, nutrients digestibility, serum parameters, and immune function. Interestingly, P-ABPS effects were similar to those obtained with ANT rather than with PRO or ABPS. In conclusion, Dietary PRO or ABPS used alone or in combination (P-ABPS), the combination augmenting the positive effect more than the independent supplement, could improve piglets’ growth performance via promoting intestinal nutrient digestion and absorption and enhancing immune function, indicating it had the potential to act as an alternative to in-feed antibiotics used in piglet diets.

Increasing attention is being focused on the use of polypeptide-based N-methyl-d-aspartate (NMDA) receptor antagonists for the treatment of nervous system disorders. In our study on Achyranthes bidentata Blume, we identified an NMDA receptor subtype 2B (NR2B) antagonist that exerts distinct neuroprotective actions. This antagonist is a 33 amino acid peptide, named bidentatide, which contains three disulfide bridges that form a cysteine knot motif. We determined the neuroactive potential of bidentatide by evaluating its in vitro effects against NMDA-mediated excitotoxicity. The results showed that pretreating primary cultured hippocampal neurons with bidentatide prevented NMDA-induced cell death and apoptosis via multiple mechanisms that involved intracellular Ca2+ inhibition, NMDA current inhibition, and apoptosis-related protein expression regulation. These mechanisms were all dependent on bidentatide-induced inhibitory regulation of NR2B-containing NMDA receptors; thus, bidentatide may contribute to the development of neuroprotective agents that would likely possess the high selectivity and safety profiles inherent in peptide drugs.

The complex composition and interaction of root-associated microbes are critical to plant health and performance. In this study, we presented a detailed characterization of three rhizocompartment (rhizosphere, rhizoplane, and root) microbiomes of Achyranthes bidentata under different years of consecutive monoculture by deep sequencing in order to determine keystone microorganisms via co-occurrence network analysis. The network analysis showed that multiple consecutive monoculture (MCM, represented 5Y and 10Y) soils generated some distinct beneficial bacterial taxa such as Bacillus, Fictibacillus, Bradyrhizobium, Shinella, and Herbaspirillum. For fungi, Mortierella substituted for Fusarium in occupying an important position in different rhizocompartments under A. bidentate monoculture. Quantitative PCR analysis confirmed a significant increase in Bacillus, Pseudomonas, and Burkholderia spp. The results of the inoculation assay showed that addition of beneficial bacteria Bacillus subtilis 74 and Bacillus halodurans 75 significantly increased the root length and fresh weight of A. bidentata. Furthermore, three types of phytosterones, as the main allochemicals, were identified both in the rhizosphere soil and in culture medium under sterile conditions by LC-MS/MS. When looking at in vitro interactions, it was found that phytosterones displayed a positive interaction with dominant beneficial species (Bacillus amyloliquefaciens 4 and B. halodurans 75) and had a negative effect on the presence of the pathogenic fungi Fusarium solani and Fusarium oxysporum. Overall, this study demonstrated that consecutive monoculture of A. bidentata can alter the bacterial and fungal community by secreting root exudates, leading to recruitment of beneficial microbes and replacement of plant-specific pathogenic fungi with plant beneficial fungi.
