High Level Transient Expression in Mammalian Cells: Identification and Optimization of Physico-Chemical Parameters of the Calcium Phosphate Transfection Method

ABSTRACTAchieving reproducible, stable, and high-level transgene expression in mammalian cells remains problematic. Previously, we attained copy-number-dependent, chromosome-position-independent expression of reporter minigenes by embedding them within a BAC containing the mouseMsh3-Dhfrlocus (DHFR BAC). Here we extend this “BAC TG-EMBED” approach. First, we report a toolkit of endogenous promoters capable of driving transgene expression over a 0.01-5 fold expression range relative to the CMV promoter, allowing fine-tuning of relative expression levels of multiple reporter genes expressed on a single BAC. Second, we show small variability in both the expression level and long-term expression stability of a reporter gene embedded in BACs containing either transcriptionally active or inactive genomic regions, making choice of BACs more flexible. Third, we describe an intriguing phenomenon in which BAC transgenes are maintained as episomes in a large fraction of stably selected clones. Finally, we demonstrate the utility of BAC TG-EMBED by simultaneously labeling three nuclear compartments in 94% of stable clones using a multi-reporter DHFR BAC, constructed with a combination of synthetic biology and BAC recombineering tools. Our extended BAC TG-EMBED method provides a versatile platform for achieving reproducible, stable simultaneous expression of multiple transgenes maintained either as episomes or stably integrated copies.

Water plays a key role in the survival, growth and reproduction of aquatic organisms. Therefore maintaining good quality of water physico-chemical parameters would ensure optimum productivity of aquatic fauna. The study evaluated water physical and chemical parameters (temperature, hydrogen ion concentration (pH), conductivity, dissolved oxygen (DO) and biochemical oxygen demand (BOD) of Bodo Creek. Sample was collected for 5 months (July 2020 – November 2020) at locations previously studied (before-spill baseline studies, 3 years, 5 years, and 7 years after-spill); by using the sampling methods used in previous studies. Spatially, there was no statistically significant difference in temperature (p > 0.05 = 0.997), pH (p > 0.05 = 0.496), DO (p > 0.05= 0.34) and BOD (p > 0.05 = 0.644). However, the difference in conductivity between the stations was statistically significant (p < 0.05 = 0.006). On monthly basis, there was a significant difference in temperature (p < 0.05 = 0.000) and pH (p < 0.05 = 0.005). In contrast, there was no statistically significant difference in conductivity (p > 0.05 = 0.633), DO (p > 0.05 = 0.559) and BOD (p > 0.05 = 0.75). Most of the physico-chemical parameters were not within the WHO recommended levels for the survival of aquatic lives, indicating a high level of environmental pollution in Bodo Creek. This study provides dataset for future evaluation of the water physico-chemistry of Bodo Creek.

AbstractThe Actinidia genus includes a number of commercial fruit species with a wide range of countries of origin. Some species such as A. arguta or A. kolomikta are characterized by a high level of resistance to frost. The aim of this study was to compare the fruit of selected cultivars of kiwiberry. In the kiwiberry fruit, sucrose was the predominant type of sugar. Citric acid was dominant in all samples; it was most represented in the pulp of kiwiberry seedlings and in the skin of kiwiberry ‘Issai’. Kiwiberry fruits feature a higher ascorbic acid content in the skin in comparison with the commercial A. chinensis cultivars. Antioxidant activity of kiwiberry fruit skin was up to 18 times higher than that of the pulp. In view of the kiwiberry fruit having no hair, it is possible to eat the whole fruit, including the skin containing substances beneficial to health.
