Biotechnological interventions of in vitro propagation and production of valuable secondary metabolites in Stevia rebaudiana

Stevia rebaudiana, belongs to the family of Asteraceae, is a perennial and medicinal shrub. It’s leaves dry powder widely used as a natural sweetener which has no calories but reported to be 50– 300 times sweetener as sweet as sugar, that are said to be having insulin balancing properties without any side effects. This research was first time conducted in Bangladesh where a quantitative analysis were performed for determining the major constituents of in vitro propagated Stevia rebaudiana plant leaves dry powder that was prepared through oven dry after sun dry. The analysis revealed the powder as mild acidic (PH 5.345). Total chlorophyll contents were found to be 0.845mg% where chlorophyll–a and -b were 0.088 and 0.761 mg% respectively.  The major macro nutrients such as ash, crude fiber, total carbohydrate, total protein and fat were determined at the amount of 7.05, 10.5, 53.52, 13.13 and 3.55 gm% respectively where the micro nutrients like Iron (Fe), Sodium (Na), Potassium (K), Calcium (Ca), Magnesium (Mg), Phosphorus (P) and Chloride (Cl) were found at the amount of 34.2, 184.3, 2500, 534.43, 465.34, 304.7 and 49.6 mg% accordingly.  Screening for secondary metabolites showed the presence of alkaloids, flavonoids, saponins and tannins as the amount of 2.83, 17.20, 6.69 and 5.84 mg% respectively in dry powder. These findings also have been compared with the other reported values on Stevia from different countries of the world.   Int J Appl Sci Biotechnol, Vol 3(4): 721-726

Diplocyclos palmatus (L.) C. Jeffrey, commonly referred to as “Shivalingi” or “Lollipop climber” is a valuable medicinal plant with a climbing growth habit used in traditional medicine. It is reputed to have antiarthritic, anti-diabetic properties and to be useful in various skin and reproductive problems. Overexploitation of wild plants and low seed germination have resulted in the decline of the species in the wild. Thus, the present investigation was aimed to establish an effective in vitro propagation procedure for its large-scale production and conservation. Nodal explants, obtained from an established mother plant were grown on MS basal medium augmented with various cytokinins, alone or in combination with auxins, to study the morphogenic response. A maximum of 8.3 shoots/explants with an average shoot length of 7.2 cm were produced after six weeks on MS containing benzylaminopurine 5.0 µM + 1-naphthaleneacetic acid 2.0 µM. After 4 weeks of transfer, microshoots rooted well on a low nutrient medium of ½ MS + 1.0 µM indole-3-butyric acid, with a maximum of 11.0 roots/microshoot and an average root length of 7.4 cm. With an 80% survival rate, the regenerated plantlets were effectively acclimatized to natural conditions. DNA-based molecular markers were used to investigate the genetic uniformity. Scanning Electron Microscopic examination of leaves indicated the adaptation of the plantlets to natural, as evidenced by the formation of normal stomata. Gas chromatography-mass spectrometry analyses of mother and micropropagated plants were performed to identify essential secondary metabolites. The results obtained show that the in vitro propagation system can be adopted for preservation, large-scale production and secondary metabolites’ production in D. palmatus.

Stevia rebaudiana is of great importance due to its steviol glycosides (SGs) which are natural sweeteners used by the food industry as well as having medicinal purposes. In the present study, the effect of plant growth regulators (PGRs) and explant types on in vitro propagation and shoot growth of S. rebaudiana were studied, the effect of PGRs on SGs production was determined. For this purpose, nodal explants and shoot tip explants were cultured on woody plant medium (WPM) supplemented with cytokinins [6-benzyladenine (BA), kinetin (Kn), or thidiazuron (TDZ)] or cytokinins + auxins combinations [BA + indoleacetic acid (IAA); BA + naphthaleneacetic acid (NAA); Kn + IAA; Kn + NAA]. Although, the best shoot proliferation was obtained on WPM supplemented with BA + NAA combinations, shoots grown on PGR-containing media produced callus at the base of the shoots and showed chlorosis and necrosis. Additionally, shoots showed at all concentrations of TDZ, and at higher concentrations of BA, morphological changes such as malformed leaves and poor shoot growth. In contrast to PGR-containing media, on the PGR-free control medium, the development of shoots and roots occurred simultaneously and healthy and well-developed plantlets were obtained. Thus, we developed an economical viable means of in vitro propagation by minimizing the micropropagation steps and removing the requirement of PGRs. According to the high-performance liquid chromatography (HPLC) results, PGR-free control medium (WPM) led to considerably higher stevioside content in the leaves compared with the PGR(s)-containing media and the highest stevioside [34 mg·g−1 dry weight (DW)] and rebaudioside A content was only detected on the control medium without PGRs. Steviolbioside, rubusoside, and dulcoside A were detected qualitatively in the leaves of shoots grown on WPM supplemented with 2.27 μm TDZ, 4.54 μm TDZ, 2.22 μm BA + 2.69 μm NAA, 2.22 μm BA + 5.37 μm NAA, 2.32 μm Kn + 5.71 μm IAA, or 2.32 μm Kn + 2.69 μm NAA.
